%X OntheMSmediumsupplementedwith2ppm2,4-D,calliwereinducedafter4-6weeksfromthepetiolesofanAmericanceleryplant£¨Apiumgraveolensvar.Dulcepers.cv.Florida£©.Suspensionculturewasstartedfromthecalliinahormone-freeliquidMSmediumonagyratoryshakerat110rpm,andkeptat26¡æ.Tostimulatecelldivisionanddedifferentiation,thesubcultureswereconductedfor7dayseachonthesamemedium.Theliquidsuspensioncontainingsinglecells,cellaggregates,andsomaticembryosindifferentstageswerescreened2-3weekslaterand1.0-1.5mmsomaticembryoswereobtained.Theseembryoswereencapsulatedwithsodiumalginatebydropping-beadmethodandsolidifiedwith0.1molCaCl2,.Thesesyntheticseedsgerminatedanddevelopedwellintoseedlingsinthesterilizedvermiculitesubstrate.
