%X TherearesixmicronucleardivisionsduringconjugationofParameciumcaudatum:threeprezygoticandthreepostzygoticdivisions.Fourhaploidnucleiareformedduringthefirsttwomeioticprezygoticdivisions.Usuallyonlyonemeioticproductislocatedintheparoralcone(PC)regionatthecompletionofmeiosis,whichsurvivesanddividesmitoticallytocompletethethirdprezygoticdivisiontoyieldastationaryandamigratorypronucleus.TheremainingthreelocatedoutsideofthePCdegenerate.Themigratorypronucleiarethenexchangedbetweentwoconjugantsandfusewiththestationarypronucleitoformsynkarya,whichundergothreesuccessivedivisions(postzygoticdivisions).However,littleisknownaboutthesurvivingmechanismofthePCnuclei.Inthecurrentstudy,stage-specificappearanceofcytoplasmicmicrotubules(cMTs)wasindicatedduringthethirdprezygoticdivisionbyimmunofluorescencelabelingwithanti-alphatubulinantibodiessurroundingthesurvivingnuclei,includingthePCnucleiandthetwotypesofprospectivepronuclei.ThissuggestedthatcMTswereinvolvedintheformationofaphysicalbarrier,whosefunctionmayrelatetosequesteringandprotectingthesurvivingnucleifromthemajorcytoplasm,wheredegenerationofextra-meioticproductsoccurs,anotherimportantnucleareventduringthethirdprezygoticdivision.
