%0 Journal Article %T Cytotoxicityandgenome-widemicroarrayanalysisofintestinalsmoothmusclecellsinresponsetohexavalentchromiuminduction %A Li-Fang JIN %A Yuan-Yuan WANG %A Zi-Dong ZHANG %A Yi-Meng YUAN %A Yi-Rui HU %A Yang-Feng WEI %A Jian NI %J 动物学研究 %D 2013 %R 10.11813/j.issn.0254-5853.2013.E3.E93 %X Chronicingestionofhighconcentrationsofhexavalentchromium[Cr(VI)]indrinkingwaterinducesintestinaltumorsinmice;however,informationonitstoxicityonintestinalsmoothmusclecellsislimited.ThepresentstudyaimedtoassesstheinvitroandinvivotoxicologicaleffectsofCr(VI)onintestinalsmoothmusclecells.Humanintestinalsmoothmusclecells(HISMcells)wereculturedwithdifferentconcentrationsofCr(VI)toevaluateeffectsoncellproliferationability,oxidativestresslevels,andantioxidantsystem.Furthermore,tissuesectionsinCr(VI)exposedrabbitswereanalyzedtoevaluatetoxicityonintestinalmusclecellsinvivo.Genechipswereutilizedtoassessdifferentialgeneexpressionprofilesatthegenome-widelevelin1μmol/LCr(VI)treatedcells.IntestinaltissuebiopsyresultsshowedthatCr(VI)increasedtheincidencesofdiffuseepithelialhyperplasiainintestinaljejunumbutcausednoobviousdamagetothestructureofthemuscularis.Cellproliferationanalysisrevealedthathighconcentrations(≥64μmol/L)butnotlowconcentrationsofCr(VI)(≤16μmol/L)significantlyinhibitedthegrowthofHISMcells.Foroxidativestresslevels,theexpressionofreactiveoxygenspecies(ROS)andnitricoxide(NO)waselevatedathighconcentrations(≥64μmol/L)butnotatlowconcentrationsofCr(VI)(≤16μmol/L).Inaddition,dose-dependentincreasesintheactivityofoxidizedglutathione(GSSH)/total-glutathione(T-GSH)werealsoobserved.Genechipscreened491differentiallyexpressedgenesincludinggenesassociatedwithcellapoptosis,oxidations,andcytoskeletons.SomeofthesedifferentiallyexpressedgenesmaybeuniquetosmoothmusclecellsinresponsetoCr(VI)induction. %K Intestinalsmoothmusclecells %K Hexavalentchromium %K Cytotoxicity %K Genechip %U http://www.zoores.ac.cn/CN/abstract/abstract3350.shtml
