%0 Journal Article %T PurificationandcharacterizationofcholecystokininfromtheskinofsalamanderTylototritonverrucosus %A Wen-Bin JIANG %A Ma HAKIM %A Lei LUO %A Bo-Wen LI %A Shi-Long YANG %A Yu-Zhu SONG %A Ren LAI %A Qiu-Min LU %J 动物学研究 %P 174-177 %D 2015 %X Asagroupofintestinalhormonesandneurotransmitters,cholecystokinins(CCKs)regulateandaffectpancreaticenzymesecretion,gastrointestinalmotility,painhypersensitivity,digestionandsatiety,andgenerallycontainaDYMGWMDFGsequenceattheC-terminus.ManyCCKshavebeenreportedinmammals.However,onlyafewhavebeenreportedinamphibians,suchasHylanigrovittata,Xenopuslaevis,andRanacatesbeiana,withnonereportedinurodeleamphibianslikenewtsandsalamanders.Here,aCCKcalledCCK-TVwasidentifiedandcharacterizedfromtheskinofthesalamanderTylototritonverrucosus.ThisCCKcontainedanaminoacidsequenceofDYMGWMDF-NH2asseeninotherCCKs.AcDNAencodingtheCCKprecursorcontaining129aminoacidresidueswasclonedfromthecDNAlibraryofT.verrucosusskin.TheCCK-TVhadthepotentialtoinducethecontractionofsmoothmusclestripsisolatedfromporcinegallbladder,elicitingcontractionataconcentrationof5.0x10-11mol/Landinducingmaximalcontractionataconcentrationof2.0x10-6mol/L.TheEC50was13.6nmol/L.Tothebestofourknowledge,thisisthefirstreporttoidentifythepresenceofaCCKinanurodeleamphibian. %K Cholecystokinin %K Salamander %K Skin %K Amphibian %U http://www.zoores.ac.cn/CN/abstract/abstract3644.shtml
