%0 Journal Article %T EstablishmentofHIV-1modelcelllineGHOST(3)withstableDRiP78andNHERF1knockdown %A Lin ZHANG %A Xu-He HUANG %A Ping-Ping ZHOU %A Guo-Long YU %A Jin YAN %A Bing QIN %A Xin-Ge YAN %A Li-Mei DIAO %A Peng LIN %A Yi-Qun KUANG %J 动物学研究 %P 161-166 %D 2015 %X ChemokinereceptorsCXCR4andCCR5areindispensableco-receptorsforHIV-1entryintohostcells.Inourpreviousstudy,weidentifiedthatdopaminereceptor-interactingprotein78(DRiP78)andNa+-H+exchangerregulatoryfactor1(NHERF1)aretheCXCR4andCCR5homo-orhetero-dimer-interactingproteins.DRiP78andNHERF1areabletoinfluencetheco-receptorinternalizationandintracellulartrafficking.Over-expressionofNHERF1affectstheligandsorHIV-1gp120-inducedCCR5internalizationandHIV-1production.ItisreasonabletospeculatethatDRiP78andNHERF1,aswellasthesignalingpathwaysinvolvedinviralreplication,wouldprobablyaffectHIV-1replicationthroughregulatingtheco-receptors.Inthispresentstudy,wedesignedtwoshorthairpinRNAs(shRNAs)targetingtheDRiP78andNHERF1,respectively,andconstructedthepLenti6/BLOCK-iT-DESTlentiviralplasmidsexpressingDRiP78orNHERF1shRNA.Thepackagedlentiviruseswereusedtotransducethewidely-appliedHIV-1modelcelllineGHOST(3).Then,cellswithstableknockdownwereestablishedthroughselectingtransducedcellswithBlasticidin.Thisstudy,forthefirsttime,reportedtheestablishmentoftheGHOST(3)withDRiP78andNHERF1knockdown,whichisthefirststablecelllinewithHIV-1co-receptor-interactingmoleculardefects. %K HIV-1 %K DRiP78 %K NHERF1 %K shRNA %K GHOST(3)cells %U http://www.zoores.ac.cn/CN/abstract/abstract3642.shtml
