%0 Journal Article %T SelectiverecruitmentofhostfactorsbyHSV-1replicationcenters %A Feng-Chao LANG %A Xin LI %A Olga VLADMIROVA %A Zhuo-Ran LI %A Gui-Jun CHEN %A Yu XIAO %A Li-Hong LI %A Dan-Feng LU %A Hong-Bo HAN %A Ju-Min ZHOU %J 动物学研究 %P 142-151 %D 2015 %X Herpessimplexvirustype1(HSV-1)entersproductiveinfectionafterinfectingepithelialcells,whereitcontrolsthehostnucleustomakeviralproteins,startsviralDNAsynthesisandassemblesinfectiousvirions.Inthisprocess,replicatingviralgenomesareorganizedintoreplicationcenterstofacilitateviralgrowth.HSV-1isknowntousehostfactors,includinghostchromatinandhosttranscriptionregulators,totranscribeitsgenes;however,theinvadingvirusalsoencountershostdefenseandstressresponsestoinhibitviralgrowth.Recently,wefoundthatHSV-1replicationcentersrecruithostfactorCTCFbutexcludeγH2A.X.Thus,HSV-1replicationcentersmayselectivelyrecruitcellularfactorsneededforviralgrowth,whileexcludinghostfactorsthataredeleteriousforviraltranscriptionorreplication.HerewereportthattheviralreplicationcentersselectivelyexcludedmodifiedhistoneH3,includingheterochromatinmarkH3K9me3,H3S10PandactivechromatinmarkH3K4me3,butnotunmodifiedH3.WefoundadynamicassociationbetweentheviralreplicationcentersandhostRNApolymeraseII.ThecentersalsorecruitedcomponentsoftheDNAdamageresponsepathway,including53BP1,BRCA1andhostantiviralproteinSP100.Importantly,wefoundthatATMkinasewasneededfortherecruitmentofCTCFtotheviralcenters.TheseresultssuggestthattheHSV-1replicationcenterstookadvantageofhostsignalingpathwaystoactivelyrecruitorexcludehostfactorstobenefitviralgrowth. %K HSV-1 %K CTCF %K &gamma %K H2AX %K Viralreplicationcente %K RNAPolII %U http://www.zoores.ac.cn/CN/abstract/abstract3640.shtml
