%0 Journal Article %T ThecomingofRNA-basedpestcontrols %A Xu Linghua %A Zeng Baosheng %A Noland Jeffery %A Huang Yongping %A Zhou Xuguo %J 植物保护学报 %P 673-690 %D 2015 %X RNAinterference(RNAi)isagenesilencingtoolthattargetsmessengerRNA(mRNA)transcriptsinasequencespecificmanneranddownregulatesgeneexpressionbyinteractingwithsmallinterferingRNAs(siRNAs).mRNAscanbesilencedeitherthroughendogenousdegradationvianucleaseactivity(e.g.,RNAipathway),orbyinhibitingtranslation(e.g.,miRNA).Overthepastdecade,RNAihasbeenusedbroadlyinentomologicalresearchtodecipherthegenefunctionsininsects.WiththesuccessofRNAiinfunctionalgenomicsresearch,muchattentionhasshiftedtothepotentialapplicationsofRNAiinagriculture,especiallyforthecontrolofinsectpests.InadditiontoRNAi-basedgenesilencing,genomeediting,anexcitingnewbiotechnology,offersyetanotheroptionforpestcontrols.Themodificationofplantgenomestoknock-ingenesthatareheritabletoincreasethetoleranceofplantstoinsectinfestation,orknock-outgenesinpestinsectsthroughgeneticallymodified(GM)-insectreleasesareontheforefrontofthegenetic-basedpestmanagements.Inthisreview,wesummarizethecurrentknowledgeregardingRNAi-basedgenesilencingandCRISPR/Cas9-basedgenomeediting.TechnicalchallengesandregulatoryconcernsforthisnewwaveofRNA-basedpestcontrolsarediscussedingreatdetail.WealsosharetheperspectiveofmodifyingcurrentenvironmentalriskassessmentframeworkstobetterfittheRNA-basedpestmanagementstrategies.Giventhecurrentdiscussions/attentionsoverthesafetyofGeneticallyModifiedOrganisms(GMO),we,respectively,comparedprosandconsofRNAi-basedgenesilencingandCRISPR/Gas9-basedgenomeediting,andlaterweidentifiedtheregulatoryissuesthatshouldbeaddressedbeforetheseemergingbiotechnologiescanmovefromthebenchtoptothetabletop. %K RNAi %K genomeediting %K CRISPR/Cas9 %K genesilencing %K pestcontrol %K technicalchallenges %K regulatoryhurdles %K emergingbiotechnology %K ecologicalriskassessment %U http://www.zwbhxb.com.cn/ch/reader/view_abstract.aspx?file_no=20150501&flag=1
