%0 Journal Article %T MolecularCloningofaCytosolicG6PDHGenefromPopulussuaveolensandItsExpressiontoImprovetheColdResistanceofTobaccoPlants %A LINYuan-zhen %A GUOHai %A ZHANGZhi-yi %A LINShan-zhi %J 冰川冻土 %P 1178-1185 %D 2009 %I 科学出版社 %X Glucose-6-phosphatedehydrogenase(G6PDH,EC1.1.1.49)isthefirstandmainregulatedenzymeofoxidativepentosephosphatepathway(OPPP),catalyzingtheconversionofglucose-6-phosphateto6-phospho-gluconolactoneandplayingimportantrolesinthegrowthanddevelopmentofplants.WepreciouslyreportedthattheenhancementoffreezingresistanceofPopulussuaveolenscuttingswasclearrelatedtothedistinctincreaseincytosolicG6PDHactivity.Here,a1697bpcDNAfragment(PsG6PDH)wasamplifiedbyRT-PCRfromcold-inducedtotalRNAofthefreezing-tolerantP.suaveolens.ThesequenceanalysisshowedthatPsG6PDHcodingregionhad1530bpandencoded510predictedaminoacidresidues.GenomicSouthernanalysisrevealedthattheisoformisencodedbyafewcopiesofthegeneinthepoplargenome.TheclonedgenePsG6PDHwasclonedintobinaryvectorpBI121andusedtotransformtobacco.PCRandSouthernblottingresultsverifiedintegrationofthisgeneintothegenomeoftobacco.Moreover,coldtreatmentexperimentsandmembranedefenseenzymeactivityanalysisconfirmedthatoverexpressionofthePsG6PDHgenecouldenhancethetolerancetocoldorfrigidstressesintransgenicplants. %K Populus %K suaveolens %K glucose-6-phosphate %K dehydrogenase %K cold %K resistance %K gene %K engineering %U http://bcdt.westgis.ac.cn/CN/abstract/abstract221.shtml
