%X The ¦Â isoform of glycogen synthase kinase 3 enzyme(GSK3¦Â), which is found at particularly highconcentrations in the central nervous system, is crucialfor its proper development and functioning. The role ofGSK3¦Â in the pathogenesis of affective disorders,schizophrenia and Alzheimer¡¯s disease, as well as itsinvolvement in other neurological disorders andneurogenesis, is discussed. A hypothesis is proposed thatinhibitory phosphorylation of GSK3¦Â induced bynon-selective cyclooxygenase (COX) inhibitors mediatesthe non-selective COX inhibitors¡¯ Alzheimer¡¯s diseaseand Parkinson¡¯s disease risk-lowering effects.Throughout the review special attention is paid tothe therapeutic potential of GSK3¦Â inhibitors.Supplementary information on GSK3¦Â cellular activityregulation, the place of GSK3¦Â in the Wnt signallingpathway, the link between GSK3¦Â and memory,neuroplasticity and circadian rhythms is provided.
