%X Andrographolide is the active principle component of the Andrographis paniculata (Acanthaceae) which is attributed for its diverse pharmacological properties. Traditionally Andrographis paniculata was extracted the leaves , steams, roots and other parts of plant. However, leaf found to be a maximum andrographolide content 2 -3% in compare to other parts of plants. In the present study callus was established and explored for andrographolide content on them. Callus was induced by culturing leaf discs on MS medium with varying concentration of cytokinins (BAP, KN, TDZ) and auxins (NAA, 2,4D,IAA, IBA).Maximum callus induction were obtained in lower concentration of 2,4-D (0.5 & 1.0 mg/l) BAP+2,4-D (0.5+1mg /l), 2,4 D+NAA (1 mg l/1mg/l),2,4-D+KN (1+0.5 mg/l).The highest andrographolide content found to be in a callus culture 2,4-D +BAP(1.0 mg/l +0.5 mg/l) followed by 2,4-D,BAP(1+1 mg/l) and 2,4-D +NAA(1 +1 mg/l).These studies used for the production of andrographolidein large scale level from callus culture.
