%X Se purific車 y se hizo la caracterizaci車n proteica de una de las cepas aisladas en Cuba del virus de la hepatitis A. Para ello se procedi車 a separar al virus de la c谷lula infectada mediante pasos de extracci車n con detergentes, concentraci車n por ultrafiltraci車n y finalmente ultracentrifugaci車n en gradiente discontinuo sacarosa-glicerol. Se determin車 la concentraci車n de prote赤nas, as赤 como la actividad antig谷nica en las diferentes fracciones del gradiente. Para la caracterizaci車n proteica del microorganismo se analizaron, por electroforesis en gel de poliacrilamida y por Western Blotting, aquellas fraccciones con la mayor actividad espec赤fica. Se demostr車 que el material viral se purific車 y concentr車 en las 迆ltimas fracciones del gradiente. Mediante la electrolisis y el Western Blotting se observaron las bandas correspondientes a las prote赤nas estructurales del virus de la hepatitis A. The purification and protein characterization of one of the Cuban isolated strains of hepatitis A virus was carried out. For this, it was necessary to separate the virus from the infected cell by extraction steps with detergents, concentration by ultrafiltration and finally, ultracentrifugation in saccharoseglycerol discontinuous gradient. Protein concentration, as well as the antigenic activity in the different fractions of the gradient were determined. For the protein characterization of the microorganism, those fractions with the greatest specific activity were analyzed by polyacrylamide gel electrophoresis and by Western blotting. It was shown that the viral material was purified and concentrated in the last fractions of the gradient. Bands corresponding to the structural proteins of hepatitis A were observed through electrophoresis and Western blotting.
