Diseases of the thyroid are accompanied by changes in antioxidant levels which could result in significant organ dysfunction. The aim of current research was to determine plasma oxidative protein damage in patients with thyroid dysfunction. 40 hypothyroid patients (mean age 44.6¡À2.1 years); 40 hyperthyroid patients (mean age 44.75¡À2.6 years) and 40 patients with euthyroid state (mean age 37.2¡À1.6 years) were included in the study. Plasma protein thiols, protein carbonyls, erythrocyte glutathione and glutathione reductase activity were measured spectrophotometrically to determine the oxidative protein damage. A significant decrease (p<0.001) in protein thiols levels in hyperthyroid by 53.22% (114.15¡À2.0 nmoles/L) and hypothyroid patients by 40.24 % (145.8¡À2.7nmoles/L) compared to euthyroid controls (244.06¡À3.1nmoles/L). Protein carbonyl levels were significantly elevated (p<0.001) in hyperthyroid by 91% (1.93¡À0.03 ng/mg protein) and in hypothyroid patients by 68.5% (1.71¡À0.02ng/mg protein) compared to euthyroid controls (1.01¡À0.01 ng/mg protein). Erythrocyte glutathione levels was significantly elevated (15.6%) in hyperthyroid patients (11.74¡À0.09 mg/g Hb) compared to euthyroid (10.147¡À0.11 mg/g Hb) (p<0.001) and significantly lower (14.06%) in hypothyroid patients (8.72¡À0.07 mg/g Hb) (p<0.001). Glutathione reductase levels expressed as activity co-efficient was significantly increased (1.47¡À0.01 in hyperthyroid and 1.23¡À0.01 in hypothyroid) in both groups when compared to euthyroid controls (1.009¡À0.03) (p<0.001) indicating a significant reduction of antioxidant capacity.
