%0 Journal Article %T Research: In-vitro INTERACTION STUDIES OF LUMEFANTRINE WITH ANTACIDS %A Chinedum Babalola* %A Oluwole Ajayi %A Olayinka Kotila %A Sunday Ameh %J Pharmacie Globale : International Journal of Comprehensive Pharmacy %D 2012 %I %X In-vitro adsorption of lumefantrine (LMF) by three antacids - magnesium trisilicate (MT) light magnesium carbonate (LMC) and aluminium hydroxide (AH) was investigated with a view to deducing the likely effect of antacid co-administration on the bioavailability of LMF. A standard curve of LMF (absorbance at 266nm versus concentration in ¦Ìg/ml) was generated from LMF, extracted and re-crystallized from Coartem tablets by taking advantage of the differences in the solubilities of LMF in ethylacetate and acetonitrile as compared with those artemether (ART) and the excipients. Adsorption and desorption curves of LMF were generated from absorbance and concentration readings of LMF under different experimental conditions. The adsorptive capacities of the antacids for LMF were: MT, 28 ¨C 71 %; AH, 81.06-99.95% and LMC well above 100%. At 37¡ãC, the desorption of LMF from LMC-LMF complex with 0.1M HCl showed a rapid but limited elution of only 0.945 - 0.005% while water did not show any desorption.
