It is assumed that the base liquid composition will definitely changes as the cells growth, since the cells will consume the nutrition in it, and also excrete toxic and waste to it. This can be recognized from the changing in color of the base liquid, and also the degradation of the transparency of the liquid. This paper attempts to introduce the design protocols of oil palm tissue culture weight measurement system using optical sensor, rule-based Expert System (ES), and MySQL acts as database. This system measures indirectly the weight of callus on explants raised on a nutrient medium in flask from the voltage of the base liquid composition density measured by an optical sensor. We found that the weight of callus and the base liquid composition density were inversely proportional to the voltages outputs of the optical sensor. Finally, our proposed system reduces the callus weight measurement process time compared to convention method %K Optical sensor %K culture growth %K callus weight %K rule-based expert system %U http://ijecct.coe.edu.my/journal/index.php/ijecct/article/view/49
