%X The optimum medium and optimal cultivation conditions of Hirsutella sinensis were studied,and the accumulation of nucleosides in the mycelia of shake culture were investigated by ultraviolet spectrophotometry. The results showed that the formula of appropriated culture medium was as follows: glucose 2%,yeast powder 3%,milk powder 1%,MgSO_4.7H_2O 0.1% and KH_2PO_4 0.2%.The suitable culture conditions were as follows: pH 6.5,medium volume 100mL/500mL,rotation rate 180r/rain,inoculum amount 10%.The total nucleoside content reached 6.5748mg/g in the mycelia fermented under optimized culture condition,improved by 33.6% as compared with that under initial culture condition.
