%X Two strategies introduced for whole genome sequencing,one is clone by clone method,the other is whole genome shotgun sequencing,for microbes which are very important to us,whole genome shotgun sequencing method is very convenient.In this article we discussed the library construction?long-to-short-ratio of insert,?total number of reads should be sequenced?assembly and gap filling technologies of the whole microbial genome shotgun sequencing method while some examples presented.
