%X 12% sucrose, 0.5% glycin and 1 hour enzymolysis of lysozyme were the optimal conditions for preparation of Streptomyces aureofaciens protoplast. The effect of protoplast regeneration and screen was enhanced when R2YE medium was replaced by bran medium. P-buffer was better than T-buffer in plasmid transformation process. 33% PEG1000 was the fit concentrations for plasmid transformation.
