游翠红 %A 童金苟 %A 俞小牧 %J 水生态学杂志 %D 2012 %I %X By using the microsatellites of pond loach Misgurnus anguillcaudatus, cross-species PCR amplification and family segregation test were carried out in Paramisgurnus dabryanus, and six loci were proved to be suitable to analyze genetic diversity and population structure of the seven populations of P. dabryanus. The average number of alleles, observed heterozygosity (Ho) and expected heterozygosity (He) ranged from 3.167 to 4.833, 0.248 to 0.417 and 0.379 to 0.505, respectively. These results showed that the level of genetic diversity in P. dabryanus was relatively low. Null alleles and apparent deficiency of heterozygotes were detected at some loci. The cluster analysis by UPGMA method was carried out based on the DA genetic distance of seven populations. The results showed that all populations were pooled into four groups: Luzhou (upstream of the Yangtze River), Guangzhou (the Zhujiang water system), Shishou and other four populations (including Shashi, Lixian, Wuhan and Poyanghu, the middle and lower reaches of the Yangtze River). The value of the F-statistics was 0.2987 and this suggested that P. dabryanus exhibited a significant genetic differentiation, mostly resulting from genetic divergence between Luzhou and the other six populations. %K Paramisgurnus dabryanus %K microsatellite %K genetic diversity %K population genetic differentiation
