%X The formulation of the relative mode content of base elements was revised. Using it, the preferential modes of tri-nucleotides in the E.coli and yeast genes were analyzed. The results suggest that the relative mode content of start codon modes in the leading region is lower than that in the other regions. Furthermore, in the E.coli genes, the relative mode content of the mode "SSS"in the trailing region and code-terminal region is higher on the average. The relation between the number of the pre-ferred or forbidden modes and gene expression level was also investigated. It is found that in the leading region, the number of the preferred modes is positively correlated to the gene expression level. However, the number of the forbidden modes is in the inverse ratio of the gene expression level. In the trailing region, the relation differs in the two species.
