%X Apply AFM to study the best situation observed the fixed cells in the air and the living cells in buffer solution in real time. Useing AFM to observe the cells treated with different fixative and the same fixative but different concentration as well as the cells without treated with any fixative. Imagine with good quality can be acquired when cells were treated with 0.5%-1% glutaraldehyde for 15 min then washed with Hanks solution. Thus, the imagine obtained from the living cells in the Hanks solution is poorer than that of cells fixed in the air, but they can be kept in the living state. Utilize the ATM to acquire imagines with high resolution of living cells, the improvement on the preparation of the cells and the observation system is necessary.
