%X Micropipette aspiration technique was adopted to investigate the adhesion of hepatocellular carcinoma (HCC) cells onto type IV collagen/laminin (LN)/fibronectin (FN) coated surfaces. Then, four synthetic peptides, namely Arg-Gly-Asp-Ser (RGDS), Gly-Arg-Gly-Asp-Thr-Pro (GRGDTP), Tyr-Ile-Gly-Ser-Arg (YIGSR) and Cys-Asp-Pro-Gly-Tyr-Ile-Gly-Ser-Arg (CDPGYIGSR) were used to study the dependence of HCC cell adhesion on the two adhesive recognition sequences, i.e., RGD and YIGSR. To summarize the experimental results, we adopted a simple function F=b/(I a) c to fit the relation between the mean cell adhesion force (F) and the concentration of competitively inhibitory peptides (I) based on the principles of competitive inhibition. The results show that co-coating of FN along with type IV collagen did not result in an increase of HCC cell adhesion forces onto type IV collagen coated surfaces. Adhesion of HCC cells onto type IV collagen coated surfaces was RGD dependent but not YIGSR dependent while that onto LN coated surfaces YIGSR dependent and FN coated surfaces RGD dependent. A brief discussion about these results was presented.
