%X H-leu incorporation and cell area of cardiocytes cultured in silicone membrane were measured when collagen type I, fibronectin or laminin was coated on as adhesive substrates with or without fibronectin,RGD peptide or laminin as integrin receptor competing inhibitors to investigate the role of extracellular matrix-integrin in the transmembrane signal transduction of heart hypertrophy induced by pressure overload. The Results showed that either collagen, fibronectin or laminin as adhesion substances were very helpful for cultured cardiocytes adhesion and spreading. 3H-leu incorporation and cell area of cultured cardiocytes were larger in stretch groups (collagen and fibronectin as adhesive substratum) than that in non-stretch group, however laminin had no effect. Moreover,when collagen was coated as adhensive substrate,3H-leu incorporation and cell area of cultured cardiocytes were inhibited significantly by soluble fibronectin and RGD peptide stimulated by stretch, laminin also had no effect. It is demonsdrated that specific extracellular matrix-integrin play a transmembrane signal transduction role in the mechanism of heart hypertrophy induced by pressure overload.
