Nuclear reconstitution of demembranatedOrychophragmus violaceus sperm inXenopus laevis egg extracts

Abstract: The cell-free extracts from animalXenopus laevis egg could induce chromatin decondensation and pronuclear formation from demembranated plant (Orychophragmus violaceu) sperm. The demembranatedOrychophragmus violaceus sperm began to swell in 30 min incubation, and then were gradually decondensed. The reassembly of nuclear envelope in the reconstituted nuclei had been visualized by means of electron microscopy and fluorescent microscopy. Membrane vesicles fused to form the double envelope around the periphery of the decondensed chromatin. The morphology of the newly formed nucleus, with a double membrane, was similar to those nuclei after fertilization. Transmission electron microscope micrograph of the whole mount prepared nuclear matrix-lamina showed the reconstituted nucleus to be filled with a dense network.

Demembranated Xenopus sperm began to swell after 15 min of incubation with Nicotiana ovule extracts. Accompanying the process of incubation, Xenopus sperm decondensed and their shapes changed gradually from long and ellipse to round. The completely decondensed chromatin was surrounded with membrane structure, which was a mixture envelope of a double membrane and a single membrane. Nucleosome assembly was verified by means of micrococcal nuclease digestion to reconstituted nuclei and DNA electrophoresis. Nicotiana ovule extracts supplied one more experimental model and system. The new system could promote powerfully the research on mechanisms of cell division and cell cycle regulation.

Abstract:

During pregnancy, a cellular immunosuppressant status is produced. It is characterized by anti-inflammatory cellular responses that allow embryonic implantation. The reversal of these changes during the postpartum period may result in overt clinical manifestations of otherwise quiescent or latent infections. We present an Acute Respiratory Distress Syndrome (ARDS) case after a caesarean section related to the Immune Reconstitution Inflammatory Syndrome (IRIS) in a Guinean black woman. The patient was treated with large doses of corticosteroids showing a great clinical and radiological improvement.

Effect of Cooking and Reconstitution Methods on the Loss of Bioactive Compounds in Pigmented and Unpigmented Potatoes  [PDF]

Abstract: Total phenolics, anthocyanins and antioxidant capacity of five coloured/pigmented (AR2009-10, Adirondack Red, Adirondack Blue, Congo, and POROIPG22-1) and two unpigmented potato genotypes (“Anuschka” and “Russet Burbank”) were assessed in fresh (with and without skin) and commercially processed/cooked/reconstituted products. Ascorbate profiles of the seven genotypes also were investigated using fresh tuber/tissue only. The results showed that genotypes greatly varied in their contents of bioactive compounds. Ascorbate profiles of the genotypes were not associated to any particular flesh colour/pigment. However, the pigmented potatoes had 1.5 to 2.5 times more the phenolics, 2 to 3 times more antioxidant capacity and higher levels of anthocyanins (13.98 to 38.57 mg C3GE-100g FW) compared to unpigmented genotypes. No anthocyanins were detected in the unpigmented potatoes. Significant losses of total phenolics, anthocyanins and total antioxidant capacity were found during peeling (18% - 23%), blanching process (40% - 60%) and further cooking/ reconstitution (7% - 12%) with no prominent genotype differences. Together, 65 to 90% of these bioactive compounds were lost during processing. The results suggested that pigmented potatoes contained higher amounts of total phenolics and anthocyanins and blanching step took away the most of the original bioactive compounds.

Abstract: Purpose. Twenty-six HIV patients with biopsy diagnosed spongiotic dermatitis (SD) were studied to determine their demographic and clinical characteristics. The condition was also investigated to determine if it was linked to the immune reconstitution inflammatory syndrome (IRIS). Methods. All patients whose skin biopsies were diagnosed by a pathologist as SD were identified. Medical records were reviewed to extract demographic descriptors, viral loads and CD4+ counts at the time that SD occurred. In an attempt to determine if the condition could be linked to Immune Reconstitution Syndrome (IRIS), the time from nadir CD4+ count to the occurrence of SD was determined. Results. SD patients were found to be older than expected but were similar to other clinic patients with regard to race/ethnicity and gender. CD4+ counts, viral loads were not related to SD. SD did not seem to be related to IRIS because few cases occurred within the time frame associated with IRIS. SD frequently appeared on the limbs although the literature suggests a more frequent appearance on the trunk and buttocks. Discussion. HIV clinicians may expect to see SD on occasion and it may appear in unusual body sites and at any time during the course of the infection.
Studies on the reconstitution properties of stored dried spotted spanish mackerel (Scomberomorus guttatus)

Abstract: A study was carried out to compare and evaluate the percentage of reconstitution media (tap water, hot water, 5% NaCl and 5% Ca(OH)2) uptake of stored dried control, irradiated (1 kGy) and heat treated (50 °C) spotted spanish mackerel Scomberomorus guttatus at room temperature for 1, 4 and 24 dipping hours upto 180 days of storage period after each 30 days of intervals. Maximum media uptake found in 5% Ca(OH)2followed by 5% NaCl, hot water and tap water in all samples. The highest (121.81%) and lowest (4.07%) percentage of media uptake observed in Ca(OH)2 and tap water respectively from the irradiated fish sample. This study also reveals that percentage of media uptake depends on reconstitution medias and dipping hours significantly but not with the various samples.
