Abstract: objective: to study the variations of the atrial natriuretic peptide (anp) introduced by extracorporeal circulation (ecc) during cardiac surgery, and to test the hypothesis that there is a correlation between the anp plasma levels, right atrial pressure (rap), left atrial pressure (lap), diuresis and natriuresis. method: study of a cohort of 15 patients submitted to myocardial revascularization using ecc. the intervals of the observation times were: t0= 10 minutes before ecc (control value); t1= 10 minutes after total flow in ecc; t2=30 minutes into total flow in ecc; t3= end stage of ecc at a nasopharingeal temperature of 36o c; and t4= 30 minutes after ecc has ended. results: the anp, lap and rap values varied significantly (p<0.001). the anp dropped from t0 to t1 (ns), and then rose progressively until t4(p<0.001). the lap and rap were reduced (p <0.001) between t0 and t1, rising progressively until t4 (p<0.001). the urinary na+ rose between t0 and t3 (p<0.001) with a drop at t4. the diuresis increased progressively at all times considered (p<0.001). a significant correlation was found between anp and the diuresis volume at t0, correlation coefficient of 0.535 (p=0.040) and at the time equal to t2 between anp and rap, a correlation coefficient of 0.590 (p=0.021). conclusion: the anp concentrations present variations during revascularization surgery with ecc, favoring the idea that they are related to atrial pressures and, at the end of ecc, they have an important function in sodium excretion and the volume of diuresis.
Participación de los péptidos natriuréticos en la hipertensión renovascular y su correlación con la evolución de la hipertrofia de miocardio

Abstract: the interactions between pressure and volume overload that occur in hypertension lead to different patterns of cardiac hypertrophy and to increase natriuretic peptides (nps) levels. the profiles of atrial natriuretic peptide (anp) and brain natriuretic peptide (bnp) synthesis and secretion have been investigated in models of hypertension. however, the different evolution of these profiles during the acute and chronic periods of pressure overload-induced cardiac hypertrophy is still unknown. for this reason, we studied one-kidney, one clip model using sprague-dawley rats at weeks 2, 4, 6 and 12 and correlated the evolution of these profiles with cardiac hypertrophy and hypertension. we observed a positive correlation between blood pressure elevation and the degree of cardiac hypertrophy, with a time-dependent increase in both parameters from week 2. levels of bnp expression showed an early increase after 2 weeks of treatment while anp increased significantly after 6 weeks. neverless, the increase in anp expression was gradual, allowing its correlation with hypertrophy and hypertension. the np expression has a differential response in the early stages of the development of hypertrophy induced by the renovascular model, with an early increase in bnp expression. once hypertrophy develops, bnp expression is no longer specific and the increase of both nps depends on and correlates with the degree of cardiac hypertrophy.
Atrial Natriuretic Factor Downregulation During Breast Cancer Progression

Abstract: We researched in a series of human breast cancers of different stages the presence and the expression of Atrial Natriuretic Factor (ANF) and Oxytocin (OX), by immunohistochemistry and RT-PCR. We found a reduction of expression of ANF during cancer progression. By contrast, OX resulted lightly expressed by immunohistochemistry and negative at RT-PCR. We hypothesize that ANF could be considered a novel marker to predict breast cancer progression; by contrast, OX expression should be better investigated, to clarify also the putative role of oxytocinases in human breast carcinogenesis.
Atrial natriuretic factor receptor guanylate cyclase, ANF-RGC, transduces two independent signals, ANF and Ca2+

Abstract: Atrial natriuretic factor receptor guanylate cyclase (ANF-RGC), was the first discovered member of the mammalian membrane guanylate cyclase family. The hallmark feature of the family is that a single protein contains both the site for recognition of the regulatory signal and the ability to transduce it into the production of the second messenger, cyclic GMP. For over two decades, the family has been classified into two subfamilies, the hormone receptor subfamily with ANF-RGC being its paramount member, and the Ca2+ modulated subfamily, which includes the rod outer segment guanylate cyclases, ROS-GC1 and 2, and the olfactory neuroepithelial guanylate cyclase. ANF-RGC is the receptor and the signal transducer of the most hypotensive hormones, ANF– and B-type natriuretic peptide (BNP). After binding these hormones at the extracellular domain it, at its intracellular domain, signals activation of the C-terminal catalytic module and accelerates the production of cyclic GMP. Cyclic GMP then serves the second messenger role in biological responses of ANF and BNP such as natriuresis, diuresis, vasorelaxation, and anti-proliferation. Very recently another modus operandus for ANF-RGC was revealed. Its crux is that ANF-RGC activity is also regulated by Ca2+. The Ca2+ sensor neurocalcin d mediates this signaling mechanism. Strikingly, the Ca2+ and ANF signaling mechanisms employ separate structural motifs of ANF-RGC in modulating its core catalytic domain in accelerating the production of cyclic GMP. In this review the biochemistry and physiology of these mechanisms with emphasis on cardiovascular regulation will be discussed.

Abstract: objective: the aims of this article are to verify if there is any alteration in the secretion of natriuretic atrial factor (naf) in children submitted to mechanical pulmonary ventilation and if these possible alterations would lead to modification in the urinary volume and in the urinary sodium excretion. methodology: a prospective study in a tertiary pediatric intensive care unit, nine children submitted to mechanical pulmonary ventilation for recuperation from anesthetics, except one who suffered a not surgical cranioencephalic trauma. the period of the study was from september 1995 to march 1996. patients with renal pathology, pulmonary pathology, in use of diuretics until 24 hours before the collection of laboratory dosages, hemodynamic instability in need of vasoactive and inothropic agents, and patients with alterations of sodium blood levels were excluded from this work. in order to have an adequate alveolar recruitment at least 3 hours were taken. after this period a blood sample was collected for dosage of natriuretic atrial factor, arterial gas analysis, glucose analysis, creatinine and electrolytes dosage. urine was collected in a 6 hours interval during the patient's period under mechanical pulmonary ventilation in order to verify the urinary volume and urinary sodium excretion. fifteen to thirty minutes after extubation, blood an urine were collected for the same dosages done before. urine was collected in a 6 hour period. a statistical comparison was done by the non parametric test of wilcoxon signed rank, with a significance level of 0,05. results: the natriuretic atrial factor showed tendency to increase when mechanical pulmonary ventilation was taken away (p=0,0547). the urinary volume and natriuresis were not altered by mechanical pulmonary ventilation.

Analysis of secondary damage to myocardial tissues after severe crush injury in rats

Abstract: Objective To examine atrial natriuretic peptide(ANP) and endothelin-1(ET-1) and ultrastructural pathologic changes in myocardial tissues in rabbits after severe crush injury and to investigate the secondary damage to myocardial tissues after severe injury and its mechanism.Methods A crush injury model was established in rabbits.Forty-two rabbits were randomly assigned into one control group(n=6) and one crush injury group(n=36).The crush injury group was further divided to six subgroups based on the time after crush release,i.e.,immediately after release and at 6,12,24,48,and 72h after release;each subgroup has six rabbits.The level of blood urea nitrogen,Cr,K+,and CK in serum is detected with an automatic biochemical analyzer.Myoglobin(MYO),cardiac troponin I(c-TnI),and CK-MB in the serum were measured using the chemiluminescence method.The myocardial tissues were collected from the same part of rabbit in each group and the content of ANP and ET-1 was determined by radio-immunoassay and the morphologic changes(at different times after the injury) in hematoxylin and eosin-stained myocardial tissue samples were observed under light microscopy and electron microscopy.Results The levels of CK,CK-MB and c-TnI in serum 12-24h after crush are apparently higher than that before crush(P < 0.05),whereas the content of ANP and ET-1 in the myocardial tissue drops(P < 0.05).Observation under light microscopy and electron microscopy revealed progressive pathologic changes in the myocytes,with the most severe myocardial damage observed within 12-24h after crush injury.The damage subsequently decreased and eventually disappears.Conclusions Severe crush injury may induce secondary damage to myocardial tissues and cardiac function.The changing trend in the cardiac markers ANP and ET-1 is consistent with the pathologic changes in myocytes during crush injury.

Abstract: background atrial natriuretic peptide (anp) and nitric oxide (no) increase diuresis and natriuresis and reduce vascular tone. we have previously demonstrated that no is involved in anp hypotensive effect in normotensive rats. objective to assess the effect of anp on mean blood pressure (mbp) and on no system in spontaneously hypertensive rats (shr) and wistar kyoto (wky), and the role of the inducible isoform of nitric oxide synthase (inos). material and methods protocol 1: animals were instilled with saline solution (0.05 ml/min) or with anp (0.2 μg/kg/min) for an hour. mbp and urinary nitrites and nitrates (nox) were assessed. the heart was extracted and inos and endothelial inos (enos) activity (with l-[u14c]-arginine) and expression (western blot) were determined. protocol 2: after adding anp (1 μm), canp(4-23) (npr-c agonist, 1μm) or aminoguanidine (inos inhibitor, 1 μm) nos activity in the right atrium and left ventricle of shr and wky was determined. results instillation with anp reduced mbp and increased nox in both groups. nos activity was greater in shr, and increased with the instillation of anp. in shr, greater enos and inos protein levels were observed, which were not modified by anp. inos basal activity was greater in shr. in the atrium, anp interacts only with npr-c in order to activate nos, and npr-a/b receptors would also take part in the ventricle. in this experimental model, the development and maintenance of hypertension could involve alterations in the interaction between both systems, anp and no.
óxido nítrico e peptídeo atrial natriurético na predi??o de complica??es da gesta??o

Abstract: purpose: to verify the effectiveness of the maternal blood serum assays of the atrial natriuretic peptide (anp) and nitric oxide (no) to predict pregnancy complications. methods: the sample was made of 49 primigravidae women. they were included in the study at the 18th week of gestation, when blood sample was collected in order to analyze the serum assays. anp was assayed by radioimmunoassay, using euro-dianostica kits (2000), considering abnormal values over 237.4 pg/ml (95 percentil). no level was evaluated by the chemiluminescence method, considering abnormal values over 17.8 mmol/l (percentil 95). for the statistical analysis of continuous quantitative variables with normal distribution, the unpaired t test was used; mann-whitney?s test was used for non parametrical quantitative samples; fisher?s exact test, for the qualitative parameter assessment; and pearson?s test for the assessment of correlations. results: there was no significant difference in the blood serum concentration of anp between the group that presented complications during pregnancy and/or peridelivery (139.3±77.1 pg/ml) and the control group (119.6±47.0 pg/ml), nor in the serum concentration of no, either, among the ones with complications in the pregnancy and/or in the peridelivery (11.1±4,6 mmol/l) and the control group (10.0±3.4 mmol/l). conclusions: the results show that anp and no serum levels are not good predictors of pregnancy complications.
