A Gene Score Test for Disease Association with Multiple Genes  [PDF]

Abstract: The traditional method for creating a gene score to predict a given outcome is to use the most statistically significant single nucleotide polymorphisms (SNPs) from all SNPs which were tested. There are several disadvantages of this approach such as excluding SNPs that do not have strong single effects when tested on their own but do have strong joint effects when tested together with other SNPs. The interpretation of results from the traditional gene score may lack biological insight since the functional unit of interest is often the gene, not the single SNP. In this paper we present a new gene scoring method, which overcomes these problems as it generates a gene score for each gene, and the total gene score for all the genes available. First, we calculate a gene score for each gene and second, we test the association between this gene score and the outcome of interest (i.e. trait). Only the gene scores which are significantly associated with the outcome after multiple testing correction for the number of gene tests (not SNPs) are considered in the total gene score calculation. This method controls false positive results caused by multiple tests within genes and between genes separately, and has the advantage of identifying multi-locus genetic effects, compared with the Bonferroni correction, false discovery rate (FDR), and permutation tests for all SNPs. Another main feature of this method is that we select the SNPs, which have different effects within a gene by using adjustment in multiple regressions and then combine the information from the selected SNPs within a gene to create a gene score. A simulation study has been conducted to evaluate finite sample performance of the proposed method.
Catalyst Composition and Content Effects on the Synthesis of Single-Walled Carbon Nanotubes by Arc Discharge

Abstract: Single-walled carbon nanotubes (SWCNTs) were prepared by a modified arc discharging furnace using Fe-Ni-Mg powders as catalyst at 600∘C. The effects of catalyst composition and content on the production rate and purity of SWCNTs are investigated in this paper. When the Fe-Ni-Mg catalyst composition is 2 : 1 : 2 wt% and the catalyst content is 5 wt%, the experimental results indicate that the production of SWCNTs is 12 grams per hour, and the purity and diameter of SWCNTs are 70% and 1.22 ∼1.38 nm, respectively. The results indicate that the cooperative function of catalyst composition and content plays an important role in the production of SWCNTs. The aim of this work is to control the production process of SWCNTs efficiently.
Wavelet transform element of SAW type

Abstract: This paper proposes to use substrate materials of small electromechanical coupling coefficient k2 (such as X-112YLiTaO3) to manufacture wavelet transform element of SAW type so as to reduce finger reflections, i.e. to reduce the error of wavelet transform element of SAW type. And it is concluded that the smaller the center frequency of the transmitting IDT of wavelet type, the smaller the error. We suggest to choose substrate material with electromechanical coupling coefficient smaller than that of X-112Y LiTaO3 in the manufacture of the transmitting IDTs of wavelet type and the receiving IDTs at center frequencies above 100MHz, so as to reduce the errors of the transmitting IDTs of wavelet type and the receiving IDTs at center frequencies above 100MHz.
Field Emission Properties of the Dendritic Carbon Nanotubes Film Embedded with ZnO Quantum Dots

Abstract: Response on the effects of individual differences of common carbon nanotubes on the field emission current stability and the luminescence uniformity of cathode film, a new type of cathode film made of dendritic carbon nanotubes embedded with Zinc oxide quantum dots is proposed. The film of dendritic carbon nanotubes was synthesized through high-temperature pyrolysis of iron phthalocyanine on a silicon substrate coated with zinc oxide nanoparticles. The dendritic structure looks like many small branches protrude from the main branches in SEM and TEM images, and both the branch and the trunk are embedded with Zinc oxide quantum dots. The turn-on field of the dendritic structure film is ~1.3 V/m at a current of 2 A, which is much lower than that of the common carbon nanotube film, and the emission current and the luminescence uniformity are better than that of the common one. The whole film emission uniformity has been improved because the multi-emission sites out from the dendritic structure carbon nanotubes cover up the failure and defects of the single emission site.
Understanding Haemophilus parasuis infection in porcine spleen through a transcriptomics approach

Abstract: A total of 931 differentially expressed (DE) transcripts were identified in the porcine spleen 7 days after HPS infection; of these, 92 unique genes showed differential expression patterns based on analysis using BLASTX and Gene Ontology. The DE genes involved in the immune response included genes for inflammasomes (RETN, S100A8, S100A9, S100A12), adhesion molecules (CLDN3, CSPG2, CD44, LGALS8), transcription factors (ZBTB16, SLC39A14, CEBPD, CEBPB), acute-phase proteins and complement (SAA1, LTF, HP, C3), differentiation genes for epithelial cells and keratinocytes (TGM1, MS4A8B, CSTA), and genes related to antigen processing and presentation (HLA-B, HLA-DRB1). Further immunostimulation analyses indicated that mRNA levels of S100A8, S100A9, and S100A12 in porcine PK-15 cells increased within 48 h and were sustained after administration of lipopolysaccharide (LPS) and Poly(I:C) respectively. In addition, mapping of DE genes to porcine health traits QTL regions showed that 70 genes were distributed in 7 different known porcine QTL regions. Finally, 10 DE genes were validated by quantitative PCR.Our findings demonstrate previously unrecognized changes in gene transcription that are associated with HPS infection in vivo, and many potential cascades identified in the study clearly merit further investigation. Our data provide new clues to the nature of the immune response in mammals, and we have identified candidate genes that are related to resistance to HPS.The pig is an important agricultural animal and is an excellent mammalian model for biomedical research [1]. Abstract: An IGBT subcircuit model is proposed and optimized,which is fully SPICE compatible.Based on analytical equations describing the semiconductor device physics,the model parameters are extracted accurately from the measured data without device destruction.The IGBT n - layer conductivity modulated resistor is effectively modeled as a voltage controlled resistor.The proposed model can be used to accurately predict the IGBT output I-V characteristics and low current gain etc.The simulation results are verified by the comparison with measurements and found to be in good agreement with them.The error in average is within 8%,which is better than the results of semi-mathematical models reported previously.

Adaptive changes of gastro-fntero-pancreatic system endocrine cells in the black-spotted pond frog Rana nigromaculata after fasting

Abstract: Changes in distribution density, morphology and secretory content of endocrine cells in the gastro-entero-pancreatic system of black-spotted frogs Rana nigromaculata before and after fasting were investigated using immunohistochemistry and antisera to six gut hormones. Six types of endocrine cells were detected in the digestive tract of Rana nigromaculata, including 5-hydroxytryptamine (5-HT), gastrin (GAS), somatostatin (SOM), glucagon (GLU), pancreatic polypeptide (PP) and vasoactive intestinal polypeptide (VIP) cells. After fasting, the density of 5-HT cells in the esophagus, cardia and fundus, GAS cells in fundus and pylorus, PP cells in the fundus decreased significantly(P <0.01), while SOM cells in the cardia, GLU cells in the rectum increased significantly (P <0.01). The cytoplasmic processes of 5-HT cells became shorter or not detectable. The secretory content of GAS cells reduced in the cardia. The positive immunostaining reaction in the perinuclear region of SOM cells in the cardia, fundus and pylorus became weaker, while the staining intensity in the periphery of these cells became stronger. VIP cells were not detectable in the whole digestive tract after fasting. Five types of endocrine cells were found in the pancreas of Rana nigromaculata, including 5-HT, GAS, SOM, GLU and PP cells. After fasting, the density of 5-HT cells decreased slightly (P >0.05), while SOM, GAS, GLU and PP cells increased significantly (P <0.01). Furthermore, the secretory content of GLU cells increased significantly. Considering their functionalities, our results indicate that the changes of GEP endocrine cells in Rana nigromaculata responded adaptively to starvation-induced stress [Current Zoology 55 (4):301–308, 2009].
