High Mobility Group Box 1 and Traumatic Brain Injury  [PDF]

Abstract: Traumatic brain injury (TBI) is a leading cause of death and disability worldwide. We identify High Mobility Group Box 1 (HMGB1) as a novel causative factor in the development of cerebral oedema, mediating coagulation, preventing secondary brain damage as well as serving as a novel therapeutic target to limit secondary neurological injury after TBI. As a prototypical danger associated molecular patterns (DAMP), HMGB1 is released from necrotic neurons via a NR2B-mediated mechanism during TBI. The secretion of HMGB1 requires severe injury and tissue hypoperfusion, and is associated with posttraumatic coagulation abnormalities, activation of complement and severe systemic inflammatory response. HMGB1 is clinically associated with elevated intracranial pressure (ICP) in patients and functionally promoted cerebral edema after TBI. The detrimental effects of HMGB1 is mediated at least in part between activation of microglial toll-like receptor 4 (TLR4) and the subsequent expression of the astrocytic water channel aquaporin-4 (AQP4). Anti-HMGB1mAb remarkably inhibited fluid percussion-induced brain edema by inhibiting HMGB1 translocation, protection of blood-brain barrier (BBB) integrity, suppression of inflammatory molecule expression and improvement of motor function. Our review demonstrates the pathological role of HMGB1 as well as the possible therapeutic valve of HMGB1 in patients with TBI.
Notch Signaling Mediates TNF-α-Induced IL-6 Production in Cultured Fibroblast-Like Synoviocytes from Rheumatoid Arthritis

Abstract: It has been reported that Notch family proteins are expressed in synovium tissue and involved in the proliferation of synoviocyte from rheumatoid arthritis (RA). The aim of this paper was to investigate whether Notch signaling mediated TNF-α-induced cytokine production of cultured fibroblast-like synoviocytes (FLSs) from RA. Exposure of RA FLSs to TNF-α (10 ng/ml) led to increase of Hes-1, a target gene of Notch signaling, and a marked upregulation of Notch 2, Delta-like 1, and Delta-like 3 mRNA levels. Blockage of Notch signaling by a γ-secretase inhibitor (DAPT) inhibited IL-6 secretion of RA FLSs in response to TNF-α while treatment with recombinant fusion protein of Notch ligand Delta-like 1 promoted such response. TNF-α stimulation also induced IL-6 secretion in OA FLSs; however, the Hes-1 level remained unaffected. Our data confirm the functional involvement of Notch pathway in the pathophysiology of RA FLSs which may provide a new target for RA therapy.

Abstract: In recent years, some problems existing in the interior auditing of Chinese enterprise groups have directly or indirectly influenced the enhancement of the management and economic benefit of enterprise group. In this article, based on detailed analysis of these problems, following countermeasures are put forward, including perfecting the management mode of interior auditing, perfecting the program of interior auditing, performing the interior auditing by computer, and enhancing the auditors’ quality of interior auditing.

Abstract: Plot and field experiments showed that the NH3 volatilization loss from rice field reached its maximum in 1-3 days after N-fertilization, which was affected by the local climate conditions (e.g., sun illumination, temperature, humidity, wind speed, and rainfall), fertilization time, and ammonium concentration in surface water of the rice field. The wet deposition of atmospheric nitrogen was correlated with the application rate of N fertilizer and the rainfall. The amount of nitrogen brought into soil or surface water by the wet deposition in rice growing season reached 7.5 kg.hm-2. The percent of NH4(+)-N in the wet deposition was about 39.8%-73.2%, with an average of 55.5%. There was a significant correlation of total ammonia volatilization loss with the average concentration of NH4(+)-N in wet deposition and total amount of wet deposition in rice growing season.
Enhancing Specific-Antibody Production to the ragB Vaccine with GITRL That Expand Tfh, IFN-γ+ T Cells and Attenuates Porphyromonas gingivalis Infection in Mice

Abstract: The outer membrane protein RagB is one of the major virulence factors of the periodontal pathogen Porphyromonas gingivalis (P. gingivalis). In order to induce protective immune response against P. gingivalis infection, an mGITRL gene-linked ragB DNA vaccine (pIRES-ragB-mGITRL ) was constructed. Six-week-old female BALB/c mice were immunized with pIRES-ragB-mGITRL through intramuscular injection and then challenged by subcutaneous injection in the abdomen with P. gingivalis. RagB-specific antibody-forming cells were evaluated by an Enzyme-linked immunosorbent spot, and specific antibody was determined by enzyme-linked immunosorbent assay. In addition, the frequencies of Tfh and IFN-γ+ T cells in spleen were measured using flow cytometer, and the levels of IL-21 and IFN-γ mRNA or proteins were detected by real time RT-PCR or ELISA. The data showed that the mGITRL-linked ragB DNA vaccine induced higher levels of RagB-specific IgG in serum and RagB-specific antibody-forming cells in spleen. The frequencies of Tfh and IFN-γ+ T cells were obviously expanded in mice immunized by pIRES-ragB-mGITRL compared with other groups (pIRES or pIRES-ragB ). The levels of Tfh and IFN-γ+ T cells associated cytokines were also significantly increased in pIRES-ragB-mGITRL group. Therefore, the mice immunized with ragB plus mGITRL showed the stronger resistant to P. gingivalis infection and a significant reduction of the lesion size caused by P. gingivalis infection comparing with other groups. Taken together, our findings demonstrated that intramuscular injection of DNA vaccine ragB together with mGITRL induced protective immune response dramatically by increasing Tfh and IFN-γ+ T cells and antibody production to P. gingivalis.
Evaluating Relational Ranking Queries Involving both Text Attributes and Numeric Attributes  [PDF]

Abstract: In many database applications, ranking queries may reference both text and numeric attributes, where the ranking functions are based on both semantic distances/similarities for text attributes and numeric distances for numeric attributes. In this paper, we propose a new method for evaluating such type of ranking queries over a relational database. By statistics and training, this method builds a mechanism that combines the semantic and numeric distances, and the mechanism can be used to balance the effects of text attributes and numeric attributes on matching a given query and tuples in database search. The basic idea of the method is to create an index based on WordNet to expand the tuple words semantically for text attributes and on the information of numeric attributes. The candidate results for a query are retrieved by the index and a simple SQL selection statement, and then top-N answers are obtained. The results of extensive experiments indicate that the performance of this new strategy is efficient and effective.

Abstract: Arrhythmogenic right ventricular cardiomyopathy/dysplasia (ARVC/D) is an autosomal dominant genetic form of cardiomyopathy (CM), which primarily affects the right ventricle (RV) and results in life threatening ventricular arrhythmias and sudden cardiac death (SCD). Diagnosis is difficult due to the broad spectrum of phenoltypic variations, especially in the early stage. Clinical suspicion should be raised in the setting of refractory ventricular tachycardia originating from the RV, and the final diagnosis could be made based on the combination of electrocardiography, echocardiography, cardiac magnetic resonance imaging (CMRI) and myocardial biopsy. Implantable cardioverter-defibrillator (ICD) implantation is an effective option for the treatment of ARVC.

Abstract: Numerical integration formulas in $n$-dimensional Euclidean space of degree three are discussed. For the integrals with permutation symmetry we present a method to construct its third-degree integration formulas with $2n$ real points. We present a decomposition method and only need to deal with $n$ one-dimensional moment problems independently.
On the Singularity of Multivariate Hermite Interpolation

Abstract: In this paper we study the singularity of multivariate Hermite interpolation of type total degree. We present a method to judge the singularity of the interpolation scheme considered and by the method to be developed, we show that all Hermite interpolation of type total degree on $m=d+k$ points in $\R^d$ is singular if $d\geq 2k$. And then we solve the Hermite interpolation problem on $m\leq d+3$ nodes completely. Precisely, all Hermite interpolations of type total degree on $m\leq d+1$ points with $d\geq 2$ are singular; for $m=d+2$ and $m=d+3$, only three cases and one case can produce regular Hermite interpolation schemes, respectively. Besides, we also present a method to compute the interpolation space for Hermite interpolation of type total degree.
