Abstract: 为筛选具抗疟疾活性的的内生真菌,采用平板分离法对海南金鸡那树内生真菌进行分离纯化。利用“4日抑制疟疾”测试内生真菌发酵液的抗疟活性,采用TLC、HPLC等分析技术对内生菌发酵液等成分进行分析。从金鸡那树叶片分离到31种内生真菌,通过筛选找到了1株具有抗疟疾活性的产奎宁或奎宁类似物的内生真菌。从金鸡那树叶片中分离得到1株产奎宁的结实串孢霉属内生真菌,为奎宁的资源开发开辟了新途径。
Toxicarioside A Inhibits Tumor Growth and Angiogenesis: Involvement of TGF-β/Endoglin Signaling

Abstract: Toxicarioside A is a cardenolide isolated mainly from plants and animals. Emerging evidence demonstrate that cardenolides not only have cardiac effects but also anticancer effects. In this study, we used in vivo models to investigate the antitumor activities of toxicarioside A and the potential mechanisms behind them. Murine colorectal carcinoma (CT26) and Lewis lung carcinoma (LL/2) models were established in syngeneic BALB/c and C57BL/6 mice, respectively. We found that the optimum effective dose of toxicarioside A treatment significantly suppressed tumor growth and angiogenesis in CT and LL/2 tumor models in vivo. Northern and Western blot analysis showed significant inhibition of endoglin expression in toxicarioside A-treated human umbilical vein endothelial cells (HUVECs) in vitro and tumor tissues in vivo. Toxicarioside A treatment significantly inhibited cell proliferation, migration and invasion, but did not cause significant cell apoptosis and affected other membrane protein (such as CD31 and MHC I) expression. In addition, TGF-β expression was also significantly inhibited in CT26 and LL/2 tumor cells treated with toxicarioside A. Western blot analysis indicated that Smad1 and phosphorylated Smad1 but not Smad2/3 and phosphorylated Smad2/3 were attenuated in HUVECs treated with toxicarioside A. Smad1 and Smad2/3 signaling remained unchanged in CT26 and LL/2 tumor cells treated with toxicarioside A. Endoglin knockout by small interfering RNA against endoglin induced alternations in Smad1 and Smad2/3 signaling in HUVECs. Our results indicate that toxicarioside A suppresses tumor growth through inhibition of endoglin-related tumor angiogenesis, which involves in the endoglin/TGF-β signal pathway.

Modeling Chinese Microblogs with Five Ws for Topic Hashtags Extraction

Abstract: Hashtags are important metadata in microblogs and are used to mark topics or index messages. However, statistics show that hashtags are absent from most microblogs. This poses great challenges for the retrieval and analysis of these tagless microblogs. In this paper, we summarize the similarity between microblogs and short-message-style news, and then propose an algorithm, named 5WTAG, for detecting microblog topics based on a model of five Ws (When, Where, Who, What, hoW). As five-W attributes are the core components in event description, it is guaranteed theoretically that 5WTAG can properly extract semantic topics from microblogs. We introduce the detailed procedure of the algorithm in this paper including spam microblog identification, microblog segmentation, and candidate hashtag construction. In addition, we propose a novel recommendation computing method for ranking candidate hashtags, which combines syntax and semantic analysis and observes the distribution of artificial topic hashtags. Finally, we conduct comprehensive experiments to verify the semantic correctness and completeness of the candidate hashtags, as well as the accuracy of the recommendation method using real data from Sina Weibo.

Abstract: This paper breaks through the relationship between the administrative appointment manager and the State-Owned controlling shareholders, through taking the professional managers with fairness preferences into the State-owned enterprises corporate governance framework. It finds that the equity incentive for the manager will not affect the tunneling behavior of state-owned controlling shareholders in the State-owned enterprise which is administrative, but the equity incentive for the Professional Managers with fairness preferences can restrain the tunneling of State-Owned controlling shareholders to a certain degree. And that a more perfect legal supervision can also restrain the tunneling of State-Owned controlling shareholders. Finally, main strategies for government were suggested, including developing professional manager market, establishing an institution of professional managers, establishing a fair and open competition environment in the market, and making a more perfect legal supervision.
Evaluation of Intersection Performance Under Atc System in Wuhan  [PDF]

Abstract: In order to accurately evaluate the implementation of Area Traffic Control (ATC) system, two situations were compared in this paper, i.e., comparison of intersection performance under two different operation conditions before and after the application of ATC system. Based on floating car theorem, this paper investigated the average intersection delay and the average vehicle stop times. During the traffic investigation, ArcGIS, GPS, VC++ and other technologies have been applied. In addition, the evaluation indices of intersection performance average intersection delay and average vehicle stop times (Guiyan Jiang et al., 2006) were selected and compared. The results showed that both average intersection delay and average vehicle stop times have been reduced after the implementation of ATC system. Improvement of traffic condition is more obvious under cooperative control status. This paper provided a reference for testing the performance of intersections under ATC system.
Inhibition of Angiogenesis, Fibrosis and Thrombosis by Tetramethylpyrazine: Mechanisms Contributing to the SDF-1/CXCR4 Axis

Abstract: Background Tetramethylpyrazine (TMP) is one of the active ingredients extracted from the Chinese herb Chuanxiong, which has been used to treat cerebrovascular and cardiovascular diseases, pulmonary diseases and cancer. However, the molecular mechanisms underlying the actions of TMP have not been fully elucidated. In a previous study we showed that TMP-mediated glioma suppression and neural protection involves the inhibition of CXCR4 expression. The SDF-1/CXCR4 axis plays a fundamental role in many physiological and pathological processes. In this study, we further investigated whether the regulation of the SDF-1/CXCR4 pathway is also involved in the TMP-mediated inhibition of neovascularization or fibrosis and improvement of microcirculation. Methodology/Principal Findings Using a scratch-wound assay, we demonstrated that TMP significantly suppressed the migration and tubule formation of the human umbilical vein endothelial cell line ECV304 in vitro. The expression of CXCR4 in ECV304 cells is notably down-regulated after TMP treatment. In addition, TMP significantly suppresses corneal neovascularization in a rat model of corneal alkali burn injury. The expression of CXCR4 on days 1, 3 and 7 post-injury was determined through RT-PCR analysis. Consistent with our hypotheses, the expression of CXCR4 in the rat cornea is significantly increased with alkali burn and dramatically down-regulated with TMP treatment. Moreover, TMP treatment significantly attenuates bleomycin-induced rat pulmonary fibrosis, while immunofluorescence shows a notably decreased amount of CXCR4-positive cells in the TMP-treated group. Furthermore, TMP significantly down-regulates the expression of CXCR4 in platelets, lymphocytes and red blood cells. Whole-blood viscosity and platelet aggregation in rats are significantly decreased by TMP treatment. Conclusions These results show that TMP exerts potent effects in inhibiting neovascularization, fibrosis and thrombosis under pathological conditions; thus, the underlying mechanism of TMP might partially contribute to the down-regulation of CXCR4.
BRCA1 Silencing Is Associated with Failure of DNA Repairing in Retinal Neurocytes

Abstract: Retinal post-mitotic neurocytes display genomic instability after damage induced by physiological or pathological factors. The involvement of BRCA1, an important factor in development and DNA repair in mature retinal neurocytes remains unclear. Thus, we investigated the developmental expression profile of BRCA1 in the retina and defined the role of BRCA1 in DNA repair in retinal neurocytes. Our data show the expression of BRCA1 is developmentally down-regulated in the retinas of mice after birth. Similarly, BRCA1 is down-regulated after differentiation induced by TSA in retinal precursor cells. An end-joining activity assay and DNA fragmentation analysis indicated that the DNA repair capacity is significantly reduced. Moreover, DNA damage in differentiated cells or cells in which BRCA1 is silenced by siRNA interference is more extensive than that in precursor cells subjected to ionizing radiation. To further investigate non-homologous end joining (NHEJ), the major repair pathway in non-divided neurons, we utilized an NHEJ substrate (pEPI-NHEJ) in which double strand breaks are generated by I-SceI. Our data showed that differentiation and the down-regulation of BRCA1 respectively result in a 2.39-fold and 1.68-fold reduction in the total NHEJ frequency compared with that in cells with normal BRCA1. Furthermore, the analysis of NHEJ repair junctions of the plasmid substrate indicated that BRCA1 is involved in the fidelity of NHEJ. In addition, as expected, the down-regulation of BRCA1 significantly inhibits the viability of retina precursor cells. Therefore, our data suggest that BRCA1 plays a critical role in retinal development and repairs DNA damage of mature retina neurocytes.

Abstract: This paper abandons the “rational man” hypothesis in traditional economics and utilizes the existing “reciprocal” principal-agent model (Pu Y. J, 2007) to analyze the benefits of environmental protection inputs. The results of the study show that if humans increase their environmental investment on the basis of rational input is set to , naturally, the increase in revenue under the assumption of “reciprocity” is r, when η > 2r, the environmental investment made by humans under irrational assumptions can bring Higher income level than “rational people”. Thus, the enlightenment of government behavior is: first, the government should fully recognize the “reciprocal” characteristics of the natural environment when formulating environmental protection policies; second, the government should further increase investment on the basis of rational input, when the increased investment meets the aforementioned conditions, the purpose of increasing human income can be achieved.
