Effects of Bisphenol A on apoptosis of spermatogenic cells and expression of Bax and Bcl-2 proteins in the Siberian frog Rana chensinensis

Abstract: In order to investigate the effects of bisphenol A (BPA) on apoptosis and the expression levels of Bax and Bcl-2 protein in amphibian spermatogenic cells, we treated Rana chensinensis with concentrations of 10-7, 10-6, 10-5mol/L BPA respectively. Testes were removed after R.chensinensis had been treated for 3 , 5 or 7 days. Apoptotic cells were examined by TUNEL and Methyl Green-Pyronine. The expression levels of Bax and Bcl-2 protein in spermatogenic cells were detected by immunohistochemical staining. The results show that apoptotic index (AI)of spermatogenic cells of R.chensinensis treated with BPA is significantly higher than that of the control groups. AI of spermatogenic cells in 10-6 and 10-7 mol/L BPA groups are similar (P>0.05), but AI in 10-5 mol/L is significantly higher than that of in 10-6 and 10-7 mol/L BPA groups (P<0.05). AI of spermatogenic cells increases with prolonged treatment. The relative intensity of Bax immunostaining in BPA treated groups is higher than that of control groups (P<0.05) while the relative intensity of Bcl-2 in BPA treated groups is lower than that of control groups (P<0.05). Positive correlations exist between the AI of spermatogenic cells and the expression ratio of Bax/Bcl-2. These results indicate that BPA can enhance the apoptosis of spermatogenic cells in R.chensinensis, and that the change of the expression ratio of Bax /Bcl-2 might have caused the increased apoptosis of spermatogenic cells in BPA treated R. chensinensis [Acta Zoologica Sinica 54(5): 867–876, 2008].

Abstract: Inthecurrentstudy,5-nydroxytryptamine(5-HT)andgastrin(GAS)cellsinthedigestivecanalsofRanachensinensistadpolesatdifferentdevelopmentalstageswereinvestigatedbyimmunohistochemistry.Resultsshowedthatthe5-HTcellswereonlydetectedintheduodenumbeforemetamorphosisbegan,andwereextensivelydistributedinthestomach,duodenum,smallintestine,andrectumthereafter,withthehighestcountsfoundintheduodenumandrectumwhenmetamorphosiswascompleted.TheGAScellswereonlydistributedinthestomachandduodenum,andonlyrarelydetectedintheduodenumbeforemetamorphosisbegan,butincreasedinthestomachduringmetamorphosisandshowedzonaldistributioninthegastricmucosawhenmetamorphosiswascompleted.Metamorphosisisacriticalperiodforamphibians,duringwhichstructuralandfunctionalphysiologicaladaptationsarerequiredtotransitionfromaquatictoterrestrialenvironments.Duringmetamorphosis,thedifferentiationsof5-HTcellsinthegastrointestinalcanalsoftadpolescouldfacilitatemucussecretionregulation,improvedigestivecanallubrication,andhelpwater-shortagefooddigestioninterrestrialenvironments.Conversely,GAScelldifferentiationsduringmetamorphosismightcontributetothedigestiveandabsorptivefunctiontransitionfromherbivoretoomnivore.

Abstract: 水环境是环境雌激素最大的储存库,环境雌激素可通过水体传递,对水生动物产生严重危害. 雌激素的生理功能通过雌激素受体(ER)介导. ER是一种配体依赖性转录因子,通过雌激素应答元件调节靶基因的转录. 大量研究证实,在环境雌激素类化合物污染的评价中,ER可作为检测环境雌激素效应的生物标记物之一. 但不同亚型的ER具有组织特异性且对配体的结合存在差异,因此在环境雌激素效应的检测中,应注意化学污染物的类型以及受检动物的物种及组织的特异性. 检测ER的方法包括ER蛋白直接定量检测和ER mRNA定量分析,检测技术的选择要依据实验设计而定. 论文对环境雌激素效应检测中ER的研究进展进行了综述,为环境雌激素污染检测工作提供了理论依据和基本方法.

Abstract: 卵黄蛋白原(Vitellogenin,Vtg)是卵黄蛋白(Vitellins,Vn)的前体. 无脊椎动物的Vtg主要在肝胰腺、卵巢或脂肪体合成,脊椎动物主要在肝脏合成. 在总结国内外相关研究基础上,对基于环境雌激素评估的Vtg研究进展进行了综述. 目前的研究已经证明,Vtg的N末端氨基酸序列在动物进化中既具有高度的保守性,又具有一定的特异性. Vtg作为检测环境雌激素效应的生物标志物之一,在环境雌激素类化合物污染的评价中被广泛采用. Vtg的检测方法包括蛋白直接定量检测和mRNA定量分析,选择检测技术要依据实验设计,对于长时间的暴露实验,可通过检测蛋白来完成,短时间的暴露实验则适合半定量RT-PCR检测Vtg mRNA的表达. 在Vtg的检测中应考虑动物种属的特异性.
Expression of c-fos, Oestradiol and Oestrogen Receptor in the Testis of Rana chensinensis at Different Stages

Abstract: 用免疫细胞化学方法检测了原癌基因FOS蛋白、17β-雌二醇(E2)和雌激素受体(ER)在中国林蛙生精周期中不同时期精巢内的表达定位。结果显示:在中国林蛙生精周期的Ⅰ—Ⅴ期,E2和ER在精原细胞、精母细胞、精子细胞、精子、支持细胞和间质细胞内均有表达。在不同时期的精巢中,E2和ER在生精细胞的定位具有一致性:在Ⅱ—Ⅲ期,精子细胞的E2和ER阳性表达最强;在Ⅲ期,精子中E2和ER阳性反应强度显著高于Ⅳ—Ⅴ期(P<0.01)。在生精周期的Ⅰ—Ⅴ期,支持细胞中,E2和ER表达强度经历由强减弱再增强的变化过程。在生精周期的Ⅲ期,间质细胞中E2和ER阳性反应强度高于其他各期。除精子外的生精细胞,支持细胞和间质细胞内均有FOS阳性反应,其表达强度呈现阶段性变化。

Abstract: 对两栖类卵巢滤泡细胞的研究已有一些报道。Thornton等 ( 1973)用电镜观察比较了蟾蜍 (Bufobufo)的成熟滤泡细胞和排卵后滤泡细胞 ,认为两栖类没有膜细胞和颗粒细胞的分化。Dumont等 ( 1978)较为系统地观察并描述了光滑爪蟾 (Xenopuslaevis)卵泡壁的超微结构。Kwon等( 1994)对黑斑蛙 (Rananigromaculata)卵巢组织细胞的离体培养研究表明 ,其类固醇激素的生成是在卵泡壁上完成的 ,并提出了两栖类卵泡类固醇生成的两类细胞模型。但目前有关有尾两栖类滤泡细胞结构与功能的研究仍少见…

Abstract: Testicular microstructure was investigated in 38 stream salamanders (Batrachuperus tibetanus) collected at the north slope of Qinling Mountain from Jan.to Dec.of 2001 and 2003.The results showed that spermatogenesis in this species followed an annual cycle,was the discontinuous type.Proliferation of spermatogonia was divided into two stages,one was from Sep.to Nov.,and the other was from Apr.to May next year.Meiosis occurs in late Jun.-Jul.,and the spermiogenesis began late Jul.and spermatozoa were present in Aug.Each lobule was distinguished from the immature region and mature region.The immature region served as a reservoir of spermatogonia,which can mitose into a proliferative region.The proliferative region transformed into the mature region,which served as the spermiogenesis site and store sperm.After spermiation,the mature region changed into the evacuated region,which was substituted by the proliferative region progressively.Spermatogenic progress was synchronized in all testicular lobules,and no maturational wave was discernible in the testis.

Abstract: A concurrent negotiation model with automated learning capability was developed in trading environments. By using Q-learning algorithm to propose own proposal and similarity criteria to evaluate the opposing party's proposal, agents can participate in concurrent multi-issue negotiation in semi-competitive situations in which there exists information uncertainty and deadlines. This model enables the negotiation more effective.

Abstract: In multi-agent system, the social commitment mechanism between agents needs to be built for solving prob- lem. This paper develops an agent social commitment mechanism based on time-utility for one-to-many negotiation in semi-competitive situations in which there exists information uncertainty and deadlines. The mechanism guarantees ra- tional actions and enables the negotiation more effective and increases utilities of the system.
