Clinical implications of increased lymph vessel density in the lymphatic metastasis of early-stage invasive cervical carcinoma: a clinical immunohistochemical method study

Abstract: We used immunohistochemical (IHC) staining with the antibody against LYVE-1 to measure lymph vessel density in 41 cases of early-stage invasive cervical carcinoma and 12 cases of normal cervical samples. We then analyzed the correlation between lymph vessel density and clinicopathological features of the tumors.(1) The majority of peritumoral lymphatics were enlarged, dilated, and irregular. In contrast, intratumoral lymph vessels were small and collapsed. The peritumoral lymphatic vessel density (PLVD) was significantly higher than the intratumoral lymphatic vessel density (ILVD) (P < 0.01). (2) Both ILVD and PLVD were significantly higher than the LVD of the control cervixes (P < 0.01). (3) Both ILVD and PLVD were significantly associated with lymph node metastasis (ILVD, P < 0.05; PLVD, P < 0.01) and lymphatic vessel invasion (ILVD, P < 0.05; PLVD, P < 0.01). Both the ILVD and PLVD in patients with histological grade HG2 and HG3 were significantly higher than those with HG1 (P < 0.05).Tumor lymphangiogenesis in early-stage invasive cervical carcinoma may play an important role in the process of lymphatic metastasis.The bloodstream and lymph system are two common pathways by which tumor cells spread. A significant amount of laboratory and clinical research indicates that tumor angiogenesis plays important roles in tumor growth and metastasis. On the other hand, research on tumor lymphangiogenesis lags behind, mostly because of the lack of specific markers of lymphatic endothelium. Recently, several markers for lymphatic endothelium have been discovered, including transmembrane protein lymphatic vessel endothelial hyaluronan receptor-1 (LYVE-1), and these provide powerful tools for studying tumor lymphangiogenesis.Cervical cancer is the most common malignant gynecological cancer, and lymphatic metastasis can occur in the early stage of tumor growth. Abstract: Bone marrow mesenchymal stem cells (MSCs) can differentiate into smooth muscle cells (SMCs) and have tremendous potential for cell therapy and tissue engineering. In this study, to understand the effects of TGF-β3 on rat bone marrow-derived MSCs and the underlying molecular mechanism of this differentiation process, we investigated that the changes of myocardin-related transcription factors (MRTFs) at the transcriptional level after rat MSCs were treated with TGF-β3. The results showed that TGF-β3 increased the expression of contractile genes, such as SM22, smooth muscle-myosin heavy chain (SM- MHC), SM-α-actin in MSCs. When TGF-β3 induced MSCs differentiation into SMCs, myocardin and MRTF-A were activated. The data indicated that TGF-β3 induced rat bone marrow-derived MSCs differentiation into SMCs by activating mypcardin and MRTF-A.

Abstract: A new analytical design method of decoupling controller matrix is proposed in terms of the standard internal model control structure for two-input two-output processes with time delays that are often encountered in chemical and industrial practice.Its advantage is that absolute decoupling for the nominal responses of system outputs can be implemented,and moreover,the controller parameters can be tuned on-line in a monotonous manner to cope with the plant unmodelled dynamics,so that significant decoupling regulation can be achieved.At the same time,for the process additive and multiplicative uncertainties mostly encountered in practice,the sufficient and necessary conditions for holding the control system robust stability are analyzed,and thus,an intuitive determination method is provided based on the spectral radius criterion.Finally,simulation examples are included to demonstrate the superiority of the proposed method over other recent approaches.

Abstract: Turbostratic carbon nanotubes (CNTs) were treated in nitrogen atmosphere at 2700 Higha2resolution transmission electron microscope (HRTEM) was used to investigate the structural changes of CNTs. Meanwhile, the conducting property was measured via a selfa2built setup installed in TEM. The structures of CNTs are found to change into bambooa2like or tubular cones after graphitization. The apex angles of cones rang from 10 o 30 and the tubular diameter ranges from 10nm to 40nm. Electron diffraction analysis reveals that an additional overlap angle is involved in the disclination operation of cones, suggesting the helicala2conical structural characteristics of CNTs. The highly curved filamental structures are stabilized via interlayer coincidence site lattices of 3a2, a2, and 9a2type. The conducting measurement indicates that the helical conical CNTs are semiconductors.

Abstract: Regardless of material regulations at the supply source or post-construction quality inspection systems, certain levels of model management systems have been developed. However, in the hospital waiting room, the measured formaldehyde concentration in the air reached 0.99 ppm, which is 12 times higher than that of the standard value. The formaldehyde emission rates measured in the building materials of the private housings or hospital waiting room ranged from 158 to 238 μg/m2?h, which exceed both the ABSL and BSMI standards. Moreover, the decoration locations, construction staff, and construction behaviors varied despite their claims of using low formaldehydeemitting materials. This study compiled the current objectives and regulations established by various supervising authorities and implemented a supporting management system in material grading, amount of material usage, ventilation path control, and ventilation effectiveness for facilitating the promotion of healthy indoor air quality, thereby enhancing public health.
Efficacies of -L-D4A against hepatitis B virus in 2.2.15 cells

Abstract: AIM: To investigate the antiviral effect of beta-L-enantiomer of 2’,3’-didehydro-2’,3’-dideoxyadenosine (β-L-D4A) on 2.2.15 cells transfected with the hepatitis B virus (HBV) genome.METHODS: Lamivudine (3TC) as a positive control. Then, HBV DNA in treated 2.2.15 cells and the Hepatitis B surface antigen (HBsAg) in the culture supernatants were detected to determine the inhibitory effect of β-L-D4A. At the same time, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) was used to detect the survival ratio of 2.2.15 cells.RESULTS: β-L-D4A has a dose-dependent inhibitory effect on HBV DNA replication; this effect was apparent when the concentration was above 1 mol/L. When β-L-D4A was at the highest concentration, 100 mol/L, the HBsAg inhibition ratio was above 50%. The Therapeutic index (TI) of β-L-D4A was above 2.1.CONCLUSION: β-L-D4A has a dose-dependent inhibitory effect on the replication of HBV DNA and the secretion of HBsAg at low toxicity.

Abstract: 长牡蛎(Crassostrea gigas)串联重复序列分析研究较少, 为了研究其在基因组转录本的基本结构特征并为长牡蛎中遗传多样性研究中提供有益的信息, 利用NCBI 公共数据库中的57 139 条长牡蛎ESTs 序列对串联重复序列结构类型、分布、丰度等进行系统比较分析。分析结果表明: 1) 长牡蛎EST中共有小卫星串联重复序列8 392 个, 在大于100 bp 重复类型中, 162~167bp 含量最高; 2)长牡蛎ESTSSR 含量丰富, 1 954 个位点是EST-SSR标记开发的候选资源; 3)EST-SSR 重复数目和重复类型在5′UTR,CDS 和3′UTR 具有显著差异, CDS 区承受更大的选择压力。

Comparative Analysis of the Shadoo Gene between Cattle and Buffalo Reveals Significant Differences

Abstract: Background While prions play a central role in the pathogenesis of transmissible spongiform encephalopathies, the biology of these proteins and the pathophysiology of these diseases remain largely unknown. Since no case of bovine spongiform encephalopathy (BSE) has ever been reported in buffalo despite their phylogenetic proximity to cattle, genetic differences may be driving the different susceptibilities of these two species to BSE. We thus hypothesized that differences in expression of the most recently identified member of the prion family or Shadoo (SPRN) gene may relate to these species-specific differences. Principal Findings We first analyzed and compared the polymorphisms of the SPRN gene (~4.4 kb), including the putative promoter, coding and 3′ regions, and further verified the entire ORF and putative promoter. This yielded a total of 117 fixed differences, remarkably: 1) a 12-bp insertion/deletion polymorphism in the hydrophobic domain of the cattle but not buffalo gene, introducing a four amino acid expansion/contraction in a series of 5 tandem Ala/Gly-containing repeats; 2) two fixed missense mutations (102Ser→Gly and 119Thr→Ala), and three missense mutations (92Pro>Thr/Met, 122Thr>Ile and 139Arg>Trp) in the coding region presenting different (P<0.05) genotypic and allelic frequency distributions between cattle and buffalo; and, 3) functional luciferase-reporter experiments for the predicted promoter region, consistent with a significantly higher activity in buffalo than cattle. Supporting these findings, immunoblotting revealed higher relative expression levels of Sho protein in cerebrum from buffalo than from cattle. In addition, for cattle, highest Sho expression was detected in obex, as compared to cerebrum or cerebellum. Significance Our findings support Sho as a non-PrP specific marker for prion infections, with obex as the best tissue source for the detection of Sho in TSE rapid tests. Moreover, these discoveries may prove advantageous for further understanding the biology of prion diseases.
