The Development of Treatment for Parkinson’s Disease  [PDF]

Abstract: Parkinson’s disease (PD) is a slowly progressive, age-related, second most common neurodegenerative disorder after Alzheimer’s disease of unknown etiology. Dopamine replacement therapies were introduced five decades ago and still remain the mainstay of treatment for Parkinson’s disease. However, with long-term treatment with L-dopa, more than 50% of patients were found to develop motor response complications approximately after 4 - 5 years of initiation of continuous treatment, in 80% of patients treated for 10 years, and in nearly 100% patients with young-onset disease. The complications of long–term treatment with levodopa include-motor fluctuations, dyskinesias, and nonmotor fluctuations are such as mood disturbance, cognitive dysfunction, dysautonomia and pain. Till date, there are various therapeutic approaches having been developed for the treatment of advanced PD comprising Pharmacotherapy, neurotrophic factors, surgical procedures such as DBS, cell-based therapies and gene therapies. The pharmacological and surgical therapies are only aiming to improve the symptoms of PD, but none are proven to have a significant effect on the underlying disease process with respect to either slowing disease progression or restoring the affected dopaminergic neurons. Although there is no cure for PD, Gene based therapy has significant prospective advantages over the conventional treatment modalities for PD, as it could theoretically be used to preserve or restore dopaminergic neurons affected by PD through the action of neurotrophic factors or alternatively increase the availability of enzymes required for dopamine synthesis. All commonly employed PD therapies focus on the amelioration of symptoms and do not cure disease. In this review only we summarize the newer therapeutic strategies for the treatment of PD such as anti-inflammatories, neurotrophic factors, neurosurgical procedures (DBS), cell based therapies and gene therapies.
Spatial Distributions of Picoplankton and Viruses in the Changjiang Estuary and Its Adjacent Sea Area during Summer

Abstract: Simultaneous determination of picoplankton (i.e., Synechococcus spp., Prochlorococcus spp., picoeukaryotes, and heterotrophic bacteria) and viruses in the Changjiang (Yangtze) River estuary and its adjacent sea area was made using flow cytometry during a cruise in June 2006. The results show that Prochlorococcus in all samples was below detectable level. The abundances of Synechococcus, picoeukaryotes, heterotrophic bacteria, and viruses ranged from 0 . 0 0 to 1 . 2 2 × 1 0 8 cell L?1, 0 . 0 1 × 1 0 6 to 1 . 4 2 × 1 0 7 cells L?1, 8 . 4 0 × 1 0 7 to 4 . 2 9 × 1 0 9 cells L?1, and 1 . 2 0 × 1 0 7 to 1 . 0 6 × 1 0 1 0 particles L?1, respectively. The determined picoplankton groups and viruses distinctly increased with the distance off the estuary and where the maximum abundance that occurred in these groups was different somewhat due to the individual sensitivity to environmental changes. Viral abundance showed a positive correlation with salinity and negative correlations with turbidity and inorganic nutrient concentrations. Positive linear relations were found between Synechococcus, heterotrophic bacteria, and viruses. 1. Introduction Autotrophic Synechococcus spp., Prochlorococcus spp., and picoeukaryotes, together with heterotrophic bacteria, are the principal components of marine picoplankton communities (0.2–2?μm in diameter) [1–4]. These picoplankton groups in the Changjiang estuary have been the subject of research in the past [5–14]. However, few data have been published on picoplankton community structure after the Three Gorges Dam construction. Previous studies were mainly focused on only one picoplankton group rather than simultaneously observing all picoplankton groups, and most of the data in the literature were obtained by epifluorescence microscope, which was not as efficient, sensitive, and precise as flow cytometry (FCM) adopted in this study [12, 13]. Marie et al. [15] reported the method of FCM measurement on marine viruses, providing a glorious prospect on studying natural viral community. Compared with other oceanic regions, the field investigation on viruses in the Changjiang estuary was much less, and, up to now, only a few reports were found [12, 14, 16, 17]. With the construction of the huge Three Gorges Dam (height 180?m, width 2?km) in the middle reaches of Changjiang River, both the amount and the timing of the terrigenous input into the East China Sea will be different [18]. Mathematical simulation method can be adopted to check flight characteristic of UAV, also can be adopted to simulate hardware experiments of unmanned aerial vehicle (UAV), then related flight experiments can be performed. The simulation method can reduce the flight periods, cost and risk. UAV flight model research play an important role in simulation and modeling in initialize periods of the UAV producing. The study of the paper focuses on the aerodynamic force modeling work of UAV based on Simulink. The designed model not only can afford mathematics simulation experiment but also will do benefits to the research of flight control, navigation guidance of UAV.

A Light-Deprivation Mouse Model Potentially for Studying the Complete Congenital Stationary Night Blindness  [PDF]

Abstract: Current rodent models of the complete congenital stationary night blindness (CSNB1) were time- consuming in breeding and validation, which makes it imperative to find a more “easily handle” animal model to broaden our understanding of this disorder. In the present study, a light-deprivation (LD) mouse model was made to validate whether it was a more “suitable” animal mode for investigating the pathogenesis of the CSNB1. Compared with controls, the LD mice exhibited a remarkable reduction in the amplitude of the dark-adapted electroretinogram (ERG) b-wave, the Max-ERG b-wave and also the oscillatory potentials (Ops), indicating an abnormal activity of rod bipolar cells in the retina. However, the ERG a-wave was relatively normal in the LD mice, which was quite consistent with what was confirmed in previously reported animal models of the CSNB1 and CSNB patients. Taken together, the LD mouse model showed CSNB1-like negative ERG responses as evidenced by the abnormal b-wave. Our study will provide a potentially useful animal model to decipher the pathogenesis of the CSNB1.

Abstract: Depression leads to a large social burden because of its substantial impairment and disability in everyday activities. The prevalence and considerable impact of this disorder call for a better understanding of its pathophysiology to improve the diagnosis, treatment and prevention. Though productive animal models and pathophysiological theories have been documented, it is still very far to uncover the complex array of symptoms caused by depression. Moreover, the neural circuitry mechanism underlying behavioral changes in some depression-like behavior animals is still limited. Changes in the neural circuitry of amygdala, dorsal raphe nucleus, ventral tegmental area, hippocampus, locus coeruleus and nucleus accumbens are related to depression. However, the interactions between individual neural circuitry of different brain areas, have not yet been fully elucidated. The purpose of the present review is to examine and summarize the current evidence for the pathophysiological mechanism of depression, with a focus on the neural circuitry, and emphasize the necessity and importance of integrating individual neural circuitry in different brain regions to understand depression.

Because these algorithms are very slow,we introduce a method based on compressive candidates, which greatly speed up the study process. Abstract: Pedro Pietri's and Kamau Brathwaite's poems suggest that a transnational and translingual comparative approach to Caribbean literature will highlight similarities across nationalities born of parallel colonial histories and emigration experiences. While the texts themselves might not articulate a transnational or translingual perspective, their intertextuality suggests that such a relationship exists. This paper will focus explicitly on how history, language, and religion play out within poems about the emigrant experience in the metropolis. I will argue that colonialism and the transnational movement from the Caribbean to urban centers have fostered similar emigrant experience even among colonial subjects with different national identities.
