Plasma proteomic study in patients with high altitude pulmonary edema (HAPE)

Abstract: Objective To investigate the differential expressions of protein in the plasma proteome in patients suffering from high altitude pulmonary edema (HAPE) and their implications. Methods The plasmas of six HAPE patients and six healthy controls were studied. The high-abundant proteins in the plasma were removed. The low-abundant proteins in the plasma/serum were segregated by 2-DE. MALDI-TOF/MS was adopted to measure the peptide fingerprints after the differential protein spots were digested by enzymes. Comparison and analysis were made in the GenBank. Results The immunoglobulin K1 light chain, serum transferrin protein precursor, and α-trypsin inhibitor heavy chain-related protein expressions were upregulated in HAPE patients compared with the control group. However the human fibrin glue coagulation protein 3 was down-regulated. Conclusion The differential expression of the above four proteins in the plasma of HAPE patients may be related to the occurrence of HAPE and can be used as the target point for the prediction of HAPE.
Studies of Cost-Income Modelof the optimum spatial scale of cities

Abstract: Optimum scale of cities is inexistent in historical view. But the optimum scale of cities in a concrete region exists in aperiod of time. Firstly, this paper proposes the hypothesis that the oper ating objective of cities is maximization of their economic profits.Then it an alyzes the transformation trend of incomes and costs of the cities when the scales of cities heighten. Next, the necessary and sufficient condition when optimu mscale of cities exists is resulted. The optimum scale of cities is in a stabl estate of the cities. Cities regulate their scales to reach the optimum scale basing on raising economic level or enlarging scale, when their scales are not optimum.The scales of cities are the urban ranges of cities. This paper chooses GDP sas incomes and budget expenditures as cost of cities. The dynamical models of incomes and costs of cities are established using territorial area and populati on, GDP per capita as independent variables based on statistical data of Jiangsu province. The model to calculate the optimum scale of cities in Jiangsu Province is drawn. The evolutionary trend of the optimum scale of cities with different population sizes is simulated using GDP percapita as independent variable.Some important results are concluded. The more the population is, the more the optimum scale, when the GDP per capita of cities keeps invariant. The last opti mum scale of cities covers an area of 2030 km~2 in Jiangsu Province when th e GDP percapita is high enough, whatever the population of cities. The speed to reach 2030 km~2 is faster when the population of cities reaches 2.8 million.Finally, this paper evaluates the adjustment of the administrative divisions of cities in Jiangsu Province in the last two years.The adjustments of Suzhou , Wuxi and Nanjing are appropriate and the adjustment extents of Huaiyin and Yan gzhou are too great. It is necessary for Changzhou, Zhenjiang and Nantong to enlarge their urban ranges. The existing scales are greater than the optimum scale of Xuzhou, Lianyungang, Yancheng, Taizhou and Suqian because of their low GDP percapita.
The Strategy of Security of Source Specific Multicast Data Forwarding Based on Single Encrypted Key

Abstract: In forwarding multicast the way of switching data is not point to point, the traditional security model on end to end is not conformable. The approaches solve the problem, which forwarding multicast data, all of multicast receivers must know a common key code. Before forwarding secure multicast data, the receiver may not know the password, which the source encrypt data with, and it only announces its public key code and the session password, which they negotiate about when the receiver join a multicast group. While the multicast source send multicast data, it stochastically choice the session key, which is encrypted multicast data and is informed the receivers directly. Before the receivers decrypt the cryptograph, they must obtain the session key that is encrypted multicast data.
Establishment of a rat model of cervical syndrome with qi deficiency, blood stasis and kidney deficiency

Abstract: Objective: To establish a rat model of cervical syndrome with qi deficiency, blood stasis and kidney deficiency.Methods: A total of 30 three-month-old female Sprague-Dawley rats were randomly divided into normal group, cervical syndrome group and cervical syndrome with qi deficiency, blood stasis and kidney deficiency group (combined group), with 10 rats in each group. Rats in the normal group received no treatment, rats in cervical syndrome group underwent resection of cervical muscles and ligaments as unbalanced dynamic and static animal model, and rats in combined group underwent resection of both cervical muscles and ovaries, swimming and irregular diet, and injection of adrenal cortex hormone and adrenaline two and a half months after resection as combined model. The qi deficiency, blood stasis and kidney deficiency were determined by observing behaviors and physical signs of the rats, detecting the contents of plasma cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), the hemorrheology, the expression of alpha-granular membrane protein (CD62p) and the serum estradiol (E2) content. The aggrecan-1, type Ⅱ procollagen gene (Col2a1), matrix metalloproteinases-13 (MMP-13) and tissue inhibitor of metalloproteinases-1 (TIMP-1) mRNAs in cervical intervertebral discs were detected by histopathology, immunohistochemistry and real-time polymerase chain reaction. The cataplasia of the intervertebral discs was determined by detecting the histopathology, the expressions of type Ⅱ collagen and type Ⅹ collegen proteins, and the expressions of aggrecan-1 (Agc1), type Ⅱ procollagen gene (Col2a1), matrix metalloproteinase-13 (MMP-13) and tissue inhibitor of metalloproteinase-1 (TIMP-1) mRNAs. Results: Compared with those in the normal group and cervical syndrome group, the rats in the combined group were noted with obvious signs of deficiency of vital energy, such as depression, tiredness, ptosis, obvious weight loss and blue tail. And the ratio of cAMP/cGMP was decreased; the reducing viscosity was significantly up-regulated; the expression of CD62p was increased; the content of serum E2 was decreased; the intervertebral disc structure was destructed; the cervical intervertebral disc was more seriously deteriorated. There exhibited a decrease in type Ⅱ collagen protein expression, an increase in type Ⅹ collagen protein expression, as well as decreases of Agc1, Col2a1 and TIMP-1 mRNA expressions in intervertebral disc, and the expression of MMP-13 mRNA was noted an increase.Conclusion: The rat model of cervical syndrome with qi-deficienc
Regulation effects of Gegen recipe and Guizhi recipe on expression of IL-1β, iNOS, TNFα, TGFβ mRNA in degenerated cervical intervertebral disk in rabbits

Abstract: Objective: To study the effects of Gegen Recipe and Guizhi Recipe regulating the expression of IL-1β, iNOS, TNFα and TGFβ mRNA in degenerated intervertebral disk caused by stimulation of wind, cold and damp. Methods: Thirty-six rabbits were randomly separated into six groups, viz. control group, stimulation by wind, cold and damp group, head in low-location group, compound model group(head in low-location plus stimulation of wind, cold and damp), Gegen Recipe group and Guizhi Recipe group. The gene expression of IL-1β, iNOS, TNFα and TGFβ mRNA in degenerated cervical intervertebral disks were assayed by RT-PCR. Results: Compared with the control group, IL-1β mRNA and iNOS mRNA expression were up-regulated in each model group (P<0.01). Both Gegen Recipe and Guizhi Recipe down-regulated IL-1β mRNA, iNOS mRNA expression, which had significant differences compared with the stimulation model group (P<0.01 or<0.05). TNFα mRNA expression was significantly enhanced in the compound model group compared with the control group (P<0.01). There was no significant difference between the head in low-location group and the stimulation model group. Gegen Recipe down-regulated TNFα mRNA expression compared with the stimulation group (P<0.01), while the effect of Guizhi Recipe was not significant. Compared with the control group, TGFβ mRNA expression went down in each model group (P<0.01). Gegen Recipe up-regulated TGFβ mRNA expression (P<0.01), while the effect of Guizhi Recipe on adjusting TGFβ mRNA was not significant. Conclusion: Stimulation by wind, cold and damp, head in low-location, and compound of both can down-regulate the expressions of IL-1β, iNOS, TNFα mRNA, and up-regulate the expression of TGFβ mRNA. These researches certify that these factors take part in the progression of intervertebral disk degeneration. IL-1β, iNOS, TNFα accelerate the degeneration and TGFβ defers the degeneration. Gegen Recipe down-regulates the expression of TNFα mRNA and up-regulates the expression of TGFβ mRNA. The effect on regulating the cell factors expressions by Guizhi Recipe is lower than that of Gegen Recipe.
A rabbit model of cervical spondylosis established by stimulation of wind, cold and dampness

Abstract: Objective: To make an animal model of cervical spondylosis (arthralgia syndrome type) with stimulation of wind, cold, and dampness. Methods: Twenty-four 8 months old male New Zealand white rabbits were randomly allocated into four groups: normal control group, light stimulation group, moderate stimulation group and severe stimulation group. The wind speed was 10.8-13.8 m/s, the temperature was (5±0.5) ℃, and the humidity was 100%. The rabbits of light, moderate, and severe stimulation groups were kept in the above-mentioned environments for 4 hours everyday, and for a total of 32, 64, and 128 hours, respectively. The intervertebral discs were stained with HE method, and observed with a light microscope. Prostaglandin E2 (PGE2), 6-ketone-prostaglandin F1α (6-K-PGF1α) and thromboxane B2 (TXB2) contents were measured by ELISA. Fas and Bcl-2 expressions were examined by immunohistochemical avidin-biotin peroxidose complex technique. Interleukin-1β (IL-1β), tumor necrosis factor α (TNF-α), and transforming growth factor β (TGF-β) mRNA expressions were examined by reverse transcription-polymerase chain reaction. Results: The nucleus pulposus of rabbits in the light and moderate stimulation groups shrunken, and in the severe stimulation group, the anulus fibrosus loosed or ruptured, and the cartilage end-plate became proliferated. Compared with rabbits in the normal control group, the PGE2 content rose in the light stimulation group, the contents of PGE2, 6-K-PGF1α, and TXB2 increased, the expressions of IL-1β and TNF-α mRNAs and Fas were up-regulated, and the expressions of TGF-β mRNA and Bcl-2 were down-regulated in the moderate and severe stimulation groups. The expression of Fas was up-regulated mostly and Bcl-2 was down-regulated mostly in the severe group. Conclusion: Moderate and severe stimulations of wind, cold and dampness can lead to degeneration of cervical intervertebral discs of rabbits. The model corresponds to the theory of traditional Chinese medicine about arthralgia syndrome caused by wind, cold and dampness.
Study of Yiqi Huayu Bushen Recipe and its decomposed formulas in regulating gene expressions in degenerated cervical intervertebral discs of rats

Abstract: Objective: To investigate the gene expression changes in the degenerated cervical intervertebral discs of rats, and to study the function of Yiqi Huayu Bushen Recipe, a compound Chinese herbal medicine, and its discomposed formulas in regulating gene expressions in the degenerated cervical intervertebral discs. Methods: The rat model with degenerated cervical intervertebral discs caused by imbalance between the dynamic and static forces was established. The mRNA was extracted from the cervical intervertebral discs of rats in the normal control and experiment groups, and the cDNA probes were obtained by inverse transcript. The cDNA probes were hybridized with the gene chips. The gene expression pattern was gained with a laser scanner. Image analysis, standardized ratio value and cluster analysis were used to investigate the differential of gene expressions between the control and experiment groups. Results: Cluster analysis showed that the gene chips of No.1 (Yiqi Huayu group), No.2 (Yiqi Bushen group) and No.3 (Huayu Bushen group) were in one class, while the gene chips of No.4 (untreated group) and No.5 (Yiqi Huayu Bushen group) were in the other. The gene expression of No.4 was different from the others mostly, and the gene expressions of No.2 and No.3 were similar. There were 96 genes expressed differently in three cases and among them 77 genes were already known and the expression of 48 genes were up-regulated (ratio>1.0), and 29 down-regulated (ratio<0.5). There were 25 genes expressed differently between the untreated group and the herb-treated groups. Conclusions: The gene expressions of the degenerated rat intervertebral discs are changed. Yiqi Huayu Bushen Recipe and its discomposed formulas have the effect of regulating the expressions of related genes, such as PI3K, PTK, ERK3, and PH1B1.
Effects of Yiqi Huayu Recipe on gene expression pattern of normal and apoptotic chondrocytes in cervical intervertebral disc in rats

Abstract: Objective: To study the effects of Yiqi Huayu Recipe on the gene expression pattern in normal and apoptotic chondrocytes in the cervical intervertebral disc of rats. Methods: The intervertebral disc endplates of rats were digested enzymatically. The apoptosis of the chondrocytes was induced by anti-Fas antibody. BiostarR-40s microarray chips were used to investigate the gene expression pattern in chondrocytes in the normal control, apoptosis and Yiqi Huayu Recipe groups. The results were scanned with Scan Array 4000 and analyzed with GenePix Pro 3.0, and then subjected to standardization and ratio analysis. Results: In the apoptosis group, 30 kinds of genes expressed in cervical intervertebral disc chondrocytes were screened out, in which 10 were up-regulated (ratio>2) and 20 down-regulated (ratio<0.5) as compared with the normal control group. On the other hand, in the Yiqi Huayu Recipe group, 97 kinds of genes expressed in cervical intervertebral disc chondrocytes were screened out, in which 44 were up-regulated (ratio>2) and 53 down-regulated (ratio<0.5) as compared with the apoptosis group. Conclusion: There are some signal transduction pathways that control the apoptosis of the intervertebral disc chondrocytes. Yiqi Huayu Recipe regulates the gene express of the apoptotic chondrocytes in intervertebral disc. The study gives a further understanding to the mechanism of the cervical spondylosis and enriches the theories of qi and blood of traditional Chinese medicine.
Mechanism of Yiqi Huayu Bushen Recipe in treating cervical syndrome in rats with qi deficiency, blood stasis and kidney deficiency

Abstract: Objective: To explore the mechanism of Yiqi Huayu Bushen Recipe (YHBR), a compound traditional Chinese herbal medicine, in treating cervical syndrome (CS) with qi deficiency, blood stasis and kidney deficiency in rats.Methods: A total of 30 three-month-old female Sprague-Dawley rats were randomly divided into normal control group, untreated group and YHBR group; there were ten rats in each group. The rat model of cervical syndrome with qi deficiency, blood stasis and kidney deficiency was established by combining disease and syndrome models. After one-month YHBR treatment in YHBR group, all rats were sacrificed. Serum, plasma and cervical intervertebral discs were detected and observed by radio-immunology, histopathology, immunohistochemistry, etc.Results: Compared with those in the normal control group, rats in the untreated group showed obvious signs of deficiency in vital energy, such as tiredness, ptosis, few movement, bluish-purple tongue and tail and weight loss; the weight of uterus and appendages, and the ratio of cyclic adenosine monophosphate/cyclic guanosine monophosphate were decreased; hemorheological parameters and the expression of alpha-granular membrane protein (CD62p) were increased; the content of serum estradiol was decreased. YHBR could improve the above indexes except for the weight of uterus and appendages. HE staining showed annulus fibrosus with cracks, diminish of pulposus nucleus, and decrease in the height of intervertebral disc and the thickness of end-plate in the untreated group; and YHBR could improve the changes of cervical intervertebral discs, but no obvious changes in end-plate. Type Ⅱ collagen protein (Col2a1)was distributed in every part of the annulus fibrosus in normal control group, but was decreased in untreated group; YHBR could increase Col2a1 expression in annulus fibrosus as compared with the untreated group. The expressions of Col2a1 and tissue inhibitor of metalloproteinase-1 (TIMP-1) mRNAs were decreased in untreated group as compared with the normal control group (P<0.01); the expressions of Col2a1 and TIMP-1 mRNAs were higher and matrix metalloproteinase-13 mRNA was lower in the YHBR group than those in the untreated group (P<0.05, P<0.01).Conclusion: YHBR may improve the condition of cervical syndrome with qi deficiency, blood stasis and kidney deficiency by regulating immune system, coagulation system and endocrine system, and delay the degeneration of cervical intervertebral disc by regulating extracellular matrix and metalloproteases in intervertebral disc.
Entropy Production and Viscosity of A Dilute Gas

Abstract: It is known that the viscosity of a dilute gas can be derived by using kinetic theory. We present here a new derivation by using two entropy production principles: the steepest entropy ascent (SEA) principle and the maximum entropy production (MEP) principle. The known result is reproduced in a similar form.
