Application of Delphi method in traditional Chinese medicine clinical research

Abstract: ABSTRACT: In recent years, Delphi method has been widely applied in traditional Chinese medicine (TCM) clinical research. This article analyzed the present application situation of Delphi method in TCM clinical research, and discussed some problems presented in the choice of evaluation method, classification of observation indexes and selection of survey items. On the basis of present application of Delphi method, the author analyzed the method on questionnaire making, selection of experts, evaluation of observation indexes and selection of survey items. Furthermore, the author summarized the steps of application of Delphi method in TCM clinical research.
Clinical epidemiology survey of the traditional Chinese medicine etiology and syndrome differentiation of coronary artery disease: study protocol of a multicenter trial

Abstract: BACKGROUND: Coronary artery disease (CAD), a common disease with high incidence and mortality rate, has seriously threatened the health and life of the public. Traditional Chinese medicine (TCM) has an important role in the prevention and treatment of this disease. Through clinical epidemiological survey, a deeper understanding of TCM etiology and syndrome characteristics in CAD would further improve clinical efficacy in the treatment of this disease.METHODS/DESIGN: The preliminary clinical questionnaire for TCM etiology and syndrome differentiation in CAD was designed after literature reviews and analysis. Through a series of clinical pre-surveys, expert consultation and demonstration, the formal TCM clinical epidemiology questionnaire on the etiology and syndrome differentiation in CAD was finalized, after which, the study protocol, inclusive and exclusive criteria and related quality control measures were prepared. The multiregional clinical epidemiological survey with more than 5 000 participants with CAD will be carried out in 41 TCM hospitals of China for investigating the TCM etiology and syndrome differentiation of CAD.DISCUSSION: Multiregion large sample size clinical epidemiology survey on TCM etiology and syndrome differentiation in CAD will provide further evidence in preventing CAD and improving the standardization process of syndrome research.TRIAL REGISTRATION: This study protocol was registered at the Chinese Clinical Trial Registry on November 27, 2011 in both Chinese and English editions and the registration number is ChiCTR-ECS-11001728.
Expert Survey on the Prevention and Treatment Situation of Traditional Chinese Medicine for Coronary Artery Disease  [PDF]

Abstract: Objective: To grasp the current situation of Traditional Chinese Medicine (TCM) on prevention and treatment for coronary artery disease (CAD) and the possible advantages and disadvantages. Method: Using a survey in the form of questionnaire among 60 cardiovascular disease experts, to grasp current situation of TCM on prevention and treatment of CAD and the possible advantages and disadvantages. Results: In most areas of China, CAD is common, and angina is the most common clinical type. More than 91% experts choose to integrate traditional and western medicine for treatment and prevention of CAD. TCM proprietary medicine, traditional herbal decoction and intravenous TCM are widely used in the clinical work. Clinical advantages of TCM in the prevention of CAD that are listed in the questionnaire include improving symptoms, enhancing quality of life, increasing exercise tolerance, improving cardiac function, relieving angina, secondary prevention of myocardial infarction, etc. The shortcomings include troublesome brewing of herbal medicine, unpleasant taste of decoction, minimal clinical evidence, slow onset of effects and non-standardized prescription of medicine, etc. Conclusion: The survey reflects the present situation of clinical diagnosis and treatment of TCM on prevention and treatment of CAD to a certain extent; more accurate conclusions need the broader, deeper and large-scale clinical survey.
Mammalian Genes Preferentially Co-Retained in Radiation Hybrid Panels Tend to Avoid Coexpression

Abstract: Coexpression has been frequently used to explore modules of functionally related genes in eukaryotic genomes. However, we found that genetically interacting mammalian genes identified through radiation hybrid (RH) genotypes tend not to be coexpressed across tissues. This pattern remained unchanged after controlling for potential confounding factors, including chromosomal linkage, chromosomal distance, and gene duplication. Because >99.9% of the genetically interacting genes were identified according to the higher co-retention frequencies, our observation implies that coexpression is not necessarily an indication of the need for the co-presence of two genes in the genome, which is a prerequisite for cofunctionality of their coding proteins in the cell. Therefore, coexpression information must be applied cautiously to the exploration of the functional relatedness of genes in a genome.

Abstract: The title compound, [PdBr2(C24H31P)2], has a distorted trans square-planar coordination of the Pd atom, which occupies an inversion centre. The most important bond distances include Pd—P of 2.380 (2) and Pd—Br of 2.515 (2) . Weak intermolecular π–π interactions between the benzene rings of adjacent molecules [centroid–centroid distance = 3.949 (6) ] are present via crystallographic inversion centres, resulting in a one-dimensional supramolecular architecture.

Abstract: The molecule of the title compound, C16H24N2O6, which lies on a crystallographic inversion centre in the centre of the benzene ring, adopts an anti conformation in terms of the relative orientation of two amide carbonyl groups. One pair of the 2-hydroxyethyl groups is partially disordered with site occupancy factors of 0.811 (2) and 0.189 (2). The dihedral angle between the amide group and central benzene ring is 67.0 (2)°. Two O—H...O and one bifurcated O—H...(O,O) hydrogen bonds are present, resulting in a three-dimensional network.

Abstract: 目的 探讨FUNDC1蛋白在卵巢癌组织中的表达及其对卵巢癌细胞增殖和侵袭的影响。方法 通过免疫组织化学(S-P法)检测FUNDC1蛋白在卵巢癌组织中的表达;流式细胞仪检测卵巢癌SKOV3细胞沉默FUNDC1基因后细胞周期的变化;EdU检测卵巢癌SKOV3细胞沉默FUNDC1基因后细胞增殖能力变化;Transwell检测卵巢癌SKOV3细胞侵袭能力;Western blotting检测p21和p53蛋白的表达。结果 FUNDC1蛋白在卵巢癌组织细胞质中表达;在卵巢癌组织中FUNDC1蛋白呈阳性表达,在正常卵巢组织中FUNDC1蛋白呈弱阳性或不表达。与正常卵巢组织相比,FUNDC1蛋白在卵巢癌组织中的表达明显增高(P=0.021);FUNDC1蛋白在晚期卵巢癌(Ⅲ/Ⅳ期)组织中的表达明显高于早期卵巢癌(Ⅰ/Ⅱ期)组织,差异有统计学意义(P=0.012)。沉默卵巢癌SKOV3细胞FUNDC1基因后,卵巢癌细胞增殖能力(P=0.032)和侵袭能力(P=0.035)降低,S期细胞明显减少(P=0.015),p21和p53蛋白表达上调。结论 FUNDC1蛋白在卵巢癌组织中的表达上调,FUNDC1调控卵巢癌SKOV3细胞增殖可能和p53/p21途径有关。
: Objective To explore the expression of FUNDC1 protein in tissues of ovarian cancer and its effects on proliferation and invasion of ovarian cancer cells. Methods The expression of FUNDC1 protein in tissues of ovarian cancer was detected by immunohistochemical (S-P method). Changes of the cell cycle of ovarian cancer cell SKOV3 were detected by flow cytometry after the FUNDC1 gene was silenced. The abilities of proliferation and invasion of ovarian cancer cell SKOV3 were detected by EdU assay and Transwell assay, respectively. Western blotting was used to detect the expressions of p21 and p53 proteins. Results FUNDC1 expressed in the cytoplasm of ovarian cancer tissues. FUNDC1 positively expressed in tissues of ovarian cancer, while showed weakly positive or negative expression in normal ovarian tissues. Compared with normal ovarian tissues, the expression of FUNDC1 in ovarian cancer tissues was significantly higher (P=0.021). The expression of FUNDC1 in advanced ovarian cancer tissues (stage Ⅲ/Ⅳ) was significantly higher that in early stage ovarian cancer tissues (stage Ⅰ/Ⅱ) and the difference was statistically significant (P=0.012). After the FUNDC1 gene of ovarian cancer cell SKOV3 was silenced, the abilities of proliferation (P=0.032) and invasion (P=0.035)of ovarian cancer cells decreased. Cells at S phase decreased significantly (P=0.015) and expressions of p21 and p53 proteins up-regulated. Conclusion The expression of FUNDC1 protein in ovarian cancer tissues is up-regulated. The regulation of proliferation of ovarian cancer cell SKOV3 by FUNDC1 may be relevant to p21/p53 pathway

Abstract: 理论分析了超导环路与一个电阻并联结构的直流超导量子干涉器(dc-SQUID)的特性.给出了这种阻尼型dc-SQUID的电压噪声谱密度、传输函数和磁通噪声谱密度随偏置电流变化的曲线.与无阻尼型高温器件比较,调制参量β大于1的阻尼型高温dc-SQUID磁强计具有高稳定性和易调试等优点.当β大于4时,因传输函数的增大,阻尼型dc-SQUID还能降低它的磁通噪声谱密度从而提高磁强计的检测灵敏度.根据现有高温超导薄膜器件的工艺条件,设计了β等于4的阻尼型高温dc-SQUID磁强计的芯片、其磁通噪声和磁场噪声的理论值
