Effects of Panax notoginseng saponins on mRNA expressions of interleukin-1β, its correlative factors and cysteinyl-aspartate specific protease after cerebral ischemia-reperfusion in rats

Abstract: Objective: To investigate the effects of Panax notoginseng saponins (PNS) on mRNA expressions of interleukin-1β (IL-1β), interleukin-1 receptor type Ⅰ (IL-1RⅠ), interleukin-1 receptor antagonist (IL-1ra), intercellular adhesion molecule-1 (ICAM-1), cysteinyl-aspartate specific protease-1 (caspase-1), caspase-3 and caspase-8 after cerebral ischemia-reperfusion in rats.Methods: Focal cerebral ischemia reperfusion in rats was induced by the method of nylon monofilament via the internal carotid artery. PNS was administered intraperitoneally respectively five minutes before cerebral ischemia and twelve hours after cerebral ischemia. After cerebral ischemia for two hours followed by reperfusion for twenty two hours, the mRNA expressions of IL-1β, IL-1RⅠ, IL-1ra, ICAM-1, caspase-1, caspase-3 and caspase-8 in brain tissue were determined by reverse transcription polymerase chain reaction assay.Results: After cerebral ischemia for two hours followed by reperfusion for twenty two hours, the mRNA expression levels of IL-1β, IL-1RⅠ, IL- 1ra, ICAM-1, caspase-1, caspase-3 and caspase-8 in brain tissue in the untreated group were obviously elevated as compared to those in the sham-operation group (P<0.05 or P<0.01). The mRNA expression levels of IL-1β, IL-1RⅠ, IL-1ra in brain tissue in the PNS group were lower than those in the untreated group, but higher than those in the sham-operation group, and without statistical differences as compared to those in the sham-operation group and in the untreated group (P>0.05). The mRNA expression level of caspase-3 in brain tissue in the PNS group was significantly lower than that in the untreated group (P<0.05), but PNS had no effect on the mRNA expression levels of ICAM-1, caspase-1 and caspase-8 in brain tissue.Conclusion: PNS can inhibit the mRNA expression of caspase-3, slightly inhibit the mRNA expressions of IL-1β and its correlative inflammatory factors in brain tissue. The protective effects of PNS on cerebral injury induced by ischemia-reperfusion may be related to inhibiting the mRNA expressions of caspase-3, IL-1β and its correlative inflammatory factors in brain tissue.

Abstract: Combining improved injector, gas line and valve-driving modules, a GC equipped with FID and ECD, could simultaneously measure CH4, CO2 and N2O in an air sample within 4 min. Test results showed that the system has high sensitivity, resolution and precision; the linear response range of the system meets the requirement of in situ flux measurements. Thus, the system is suitable for monitoring fluxes of main greenhouse gases in terrestrial ecosystem since it is easy to use, efficacious, stable and reliable to collect data.
Phase Transitions and Vortex Line Entanglement in a Model High Temperature Superconductor

Abstract: Monte Carlo simulations of the uniformly frustrated 3d XY model are used to model vortex line fluctuations in high temperature superconductors in an applied magnetic field. We find two distinct phase transitions. At a lower T_{c\perp}, the vortex lattice melts and coherence is lost in planes perpendicular to the magnetic field. At a higher T_{cz}, a vortex tangle percolates throughout the system, and coherence is lost parallel to the magnetic field. Cooling below T_{cz}, high energy barriers for vortex line cutting lead to an entangled glassy state. Figures available upon request to stte@buckwheat.pas.rochester.edu.
Vortex Line Fluctuations in Model High Temperature Superconductors

Abstract: We carry out Monte Carlo simulations of the uniformly frustrated 3d XY model as a model for vortex line fluctuations in a high Tc superconductor. A density of vortex lines of f=1/25 is considered. We find two sharp phase transitions. The low T phase is an ordered vortex line lattice. The high T normal phase is a vortex line liquid with much entangling, cutting, and loop excitations. An intermediate phase is found which is characterized as a vortex line liquid of disentangled lines. In this phase, the system displays superconducting properties in the direction parallel to the magnetic field, but normal behavior in planes perpendicular to the magnetic field.

Abstract: In this paper, the generic intersection theory for difference varieties is presented. Precisely, the intersection of an irreducible difference variety of dimension $d > 0$ and order $h$ with a generic difference hypersurface of order $s$ is shown to be an irreducible difference variety of dimension $d-1$ and order $h+s$. Based on the intersection theory, the difference Chow form for an irreducible difference variety is defined. Furthermore, it is shown that the difference Chow form of an irreducible difference variety $V$ is transformally homogenous and has the same order as $V$.
Profiling of the Tetraspanin CD151 Web and Conspiracy of CD151/Integrin β1 Complex in the Progression of Hepatocellular Carcinoma

Abstract: Tetraspanin CD151 has been implicated in metastasis through forming complexes with different molecular partners. In this study, we mapped tetraspanin web proteins centered on CD151, in order to explore the role of CD151 complexes in the progression of hepatocellular carcinoma (HCC). Immunoprecipitation was used to isolate tetraspanin complexes from HCCLM3 cells using a CD151 antibody, and associated proteins were identified by mass spectrometry. The interaction of CD151 and its molecular partners, and their roles in invasiveness and metastasis of HCC cells were assayed through disruption of the CD151 network. Finally, the clinical implication of CD151 complexes in HCC patients was also examined. In this study, we identified 58 proteins, characterized the tetraspanin CD151 web, and chose integrin β1 as a main partner to further investigate. When the CD151/integrin β1 complex in HCC cells was disrupted, migration, invasiveness, secretion of matrix metalloproteinase 9, and metastasis were markedly influenced. However, both CD151 and integrin β1 expression were untouched. HCC patients with high expression of CD151/integrin β1 complex had the poorest prognosis of the whole cohort of patients. Together, our data show that CD151 acts as an important player in the progression of HCC in an integrin β1-dependent manner.
Tree growth and soil nutrients in response to nitrogen deposition in a subtropical Chinese fir plantation

Abstract: To investigate the response of subtropical forests to increased nitrogen deposition a field experiment was conducted in Chinese fir plantation involving four N treatments designated as N0(control),N1,N2 and N3 at the doses of 0,60,120 and 240 kg N hm-2 a-1,respectively.Based on three years of manipulation,medium to high N loads(N2,N3) increased average annual increment of diameter at breast height(DBH),but low N(N1) produced no significant effects.Nitrogen deposition also had a positive effect on tree height,but the increment magnitude decreased with the increasing N additions.The stand volume,calculated from binary tree volume equation,responded positively to N treatments,with a minor gain in N1,an annual gain of 3.81 m3 hm-2 stemwood in N2 and 4.86 m3 hm-2 stemwood in excess of the control(N0) in N3,although no significant difference statistically existing between treatments.The soil NH 4-N and NO-3-N increased,but pH values and the contents of organic matter,available P and K,exchangeable Ca and Mg decreased with the increasing N additions.Nutrient status of the needles exhibited similar sensitivity to N treatments,N1,N2 and N3 increased N concentrations in the leaves by 18.25%,11.68% and 13.14%,respectively,but reduced to some extents the contents of P,K,Ca and Mg.
Measurement and Study on the Atmospheric Trace BTEX in Beijing

Abstract: Atmospheric trace BTEX in Beijing was observed by a gas chromatography/mass spectrometry in the latest 4 years. The diurnal, lunar and annual concentration variations of these pollutants were reported. In winter and spring, the diurnal profiles exhibited two peak values corresponding to the daily rush hours 08:00-10:00 am and 17:00 pm; in autumn, there was the third peak value in the night at 22:00; the highest concentration of BTEX in summer appeared in the midday and totally three daily maxima were in the sampled day. In the lunar variation of the BTEX from 2000 to 2002, the average concentration was about (44.1 +/- 24.5) nmol/mol (C unit), and the seasonal maximum value appeared in summer. The trend of BTEX was decreased greatly from 1999 to 2002.
Measurement Method Research on Trace Volatile Organic Compounds in Atmosphere

Abstract: An automated accumulation sampling system and a method of two-step Cryo-Concentrated System combined with Gas Chromatography/Mass Spectrometry (CCS-GC/MS) are introduced. The method is evaluated by a set of special experiments and the results are demonstrated. The lowest limit volume concentrations of the measurement is expanded from 10 (-6) to 10(-12) using CCS-GC/ MS instead of the normal method of GC/MS and the average response factor of 39 object compounds is 2.9 x 10(-12) x A(-1). When injection volume of air sample reaches 1000 cm3, the lowest limit is 7 x 10(-12) - 40 x 10(-12); CCS-GC/MS may identify all the objective chemical species in the atmosphere samples, with average 2.5s bias of retention time for 39 GC peaks. Within 0 - 400 x 10(-9), the concentration of all the objective compounds can be calculated by standard curve individually (average r2 above 0.99) and accurately. The recollect rate is 88% - 111%, at average of 100.8% +/- 5.6%; and the bias of precision is 2% - 14%, at average of 6.6%.
