Nanoparticles Deposition at Specific Sites Using Aligned Fiber Networks  [PDF]

Abstract: We report a new method to precisely locate inorganic nanoparticles at polymeric fiber crossing sites. Polymeric fibers are aligned by a previously reported non-electrospinning Spinneret based Tunable Engineered Parameters (STEP) polymeric fiber manufac- turing technique. Cross-hatch pattern (perpendicular) of polymeric fibers embedded with inorganic nanoparticles (e.g.: Fe(NO3)3 or Bi(NO3)3) were fabricated on top of each other and the fiber compositions re-acted during annealing process, thus, producing a desired phase (e.g.: BiFeO3) only at the crossing site. Our method of-fers an easy way to mass arrange and locate nanoparticles at designed positions on virtually all types of substrates.

Abstract: We have studied the deposition of BaTiO3 (BTO) thin films on various substrates. Three representative substrates were selected from different types of material systems: (i) SrTiO3 single crystals as a typical oxide, (ii) Si wafers as a semiconductor, and (iii) Ni foils as a magnetostrictive metal. We have compared the ferroelectric properties of BTO thin films obtained by pulsed laser deposition on these diverse substrates.

Separation and Purification of Recombinant Nippostrongylus brasiliensis Acetylcholinesterase from Culture Medium of Genetic Engineering Pichia pastoris

Abstract: To develop a simple, fast and highly efficient method for the separation and purification of recombinant Nippostrongylus brasiliensis acetylcholinesterase (NbAChE) from culture medium of genetic engineering Pichia pastoris, Q-Sepharose Fast Flow chromatographic medium was used. The chromatographic column was 20 cm x 3.5 cm i. d. The elution buffer A was 20 mmol/L NaH2PO4-Na2HPO4 (pH 8) and buffer B was 1 mol/L NaCl + 20 mmol/L NaH2PO4- Na2HPO4(pH 8). The elution gradient was nonlinear. It was firstly eluted with 10% B for 300 min, then with 30% B for 300 min, finally with 100% B for 300 min. The flow rate of mobile phase was 6 mL/min. The obtained recombinant NbAChE was proved to be homogeneous on sodium-dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) and its relative molecular mass was estimated to be approximately 66 000, which was consistent with that reported in literature. The total activity recovery of this purification method was 52.6% and the purification factor was 3.87. The final specific activity of recombinant NbAChE was 2 837 U/mg. This chromatographic process is simple and highly efficient. It can be used to separate and purify recombinant NbAChE from culture medium of Pichia pastoris harboring NbAChE gene.
Characterization and Evolution of Conserved MicroRNA through Duplication Events in Date Palm (Phoenix dactylifera)

Abstract: MicroRNAs (miRNAs) are important regulators of gene expression at the post-transcriptional level in a wide range of species. Highly conserved miRNAs regulate ancestral transcription factors common to all plants, and control important basic processes such as cell division and meristem function. We selected 21 conserved miRNA families to analyze the distribution and maintenance of miRNAs. Recently, the first genome sequence in Palmaceae was released: date palm (Phoenix dactylifera). We conducted a systematic miRNA analysis in date palm, computationally identifying and characterizing the distribution and duplication of conserved miRNAs in this species compared to other published plant genomes. A total of 81 miRNAs belonging to 18 miRNA families were identified in date palm. The majority of miRNAs in date palm and seven other well-studied plant species were located in intergenic regions and located 4 to 5 kb away from the nearest protein-coding genes. Sequence comparison showed that 67% of date palm miRNA members were present in duplicated segments, and that 135 pairs of miRNA-containing segments were duplicated in Arabidopsis, tomato, orange, rice, apple, poplar and soybean with a high similarity of non coding sequences between duplicated segments, indicating genomic duplication was a major force for expansion of conserved miRNAs. Duplicated miRNA pairs in date palm showed divergence in pre-miRNA sequence and in number of promoters, implying that these duplicated pairs may have undergone divergent evolution. Comparisons between date palm and the seven other plant species for the gain/loss of miR167 loci in an ancient segment shared between monocots and dicots suggested that these conserved miRNAs were highly influenced by and diverged as a result of genomic duplication events.
Interaction of Gold Nanoparticles in Barium Titanate Thin Films

Abstract: A novel approach to control the grain size of oxide thin film materials has been investigated. Perovskite BaTiO3 shows interesting grain structures when deposited on gold predeposited, (111)-oriented, singlecrystal SrTiO3 substrates. Solid oxide films grow epitaxially on patterned seed layers and show variations in grain size relative to the films deposited on SrTiO3 directly.
Arrhenius-type domain growth in Pb(In1/2Nb1/2)O3-Pb(Mg1/3Nb2/3)O3-PbTiO3 crystals

Abstract: Single crystals of Pb(In1/2Nb1/2)O3-Pb(Mg1/3Nb2/3)O3-PbTiO3 (PIN-PMN-PT) poled along [001] were investigated by dielectric, x-ray, and polarized light (PLM) and piezo-force microscopy (PFM) methods. PLM revealed {100} macro-domain plates that formed after poling, whose size increased on heating between room temperature and a rhombohedral \rightarrow tetragonal phase transition, above which point a break-up of the macro-domain plates was observed. Corresponding PFM studies demonstrated that poling reduced the size of stripe-like domains that were internal to the macro-domain plates, whose size also increased on heating to TR-T. The temperature dependence of both the size of the macro-domain plates and internal sub-domains followed the Arrhenius relation with the activation energy of 0.4-0.5eV. The coercive field displays an abnormal increase on heating below TR-T, different than that for PMN-PT. The anomalously increased coercive field can be ascribed to the Arrhenius-type domain growth, indicating a simple thermally activated process and an important role of hierarchial domains in the improved performance of PIN-PMN-PT.

Abstract: 利用10年的TRMM卫星降水雷达观测资料,首次对青藏高原及其下游平原及海洋地区降水厚度的地区差异进行了对比分析,并对青藏高原及其周边地区对流和层云降水厚度的水平分布及其日变化和季节变化进行了统计分析,结果表明:(1)青藏高原地区对流和层云降水厚度都要比下游平原地区更为浅薄,东部海洋地区对流降水厚度比平原地区小,而层云降水厚度与平原地区相当。青藏高原及其下游平原地区对流降水厚度的日变化特征非常明显,海洋地区对流降水厚度日夜差异则不大。层云降水厚度在各地区的日变化特征都不明显。青藏高原、下游平原及海洋地区对流和层云降水厚度的季节变化都非常明显,从冬至夏,对流和层云降水逐渐变得深厚,而从夏入冬,对流和层云降水则逐渐变得浅薄。(2)青藏高原及其周边地区对流和层云平均降水厚度的分布形式和降水量分布具有较好的对应关系,降水量大的地区其降水厚度一般较为深厚,降水少的地区则降水厚度比较浅薄。对流和层云降水厚度存在明显差异,对流降水一般要比层云降水深厚。青藏高原及其周边地区降水厚度水平分布的日夜差距不大,但季节变化非常明显,且与气候系统的季节变化紧密相关。
RNA-Seq Analysis of Cocos nucifera: Transcriptome Sequencing and De Novo Assembly for Subsequent Functional Genomics Approaches

Abstract: Background Cocos nucifera (coconut), a member of the Arecaceae family, is an economically important woody palm grown in tropical regions. Despite its agronomic importance, previous germplasm assessment studies have relied solely on morphological and agronomical traits. Molecular biology techniques have been scarcely used in assessment of genetic resources and for improvement of important agronomic and quality traits in Cocos nucifera, mostly due to the absence of available sequence information. Methodology/Principal Findings To provide basic information for molecular breeding and further molecular biological analysis in Cocos nucifera, we applied RNA-seq technology and de novo assembly to gain a global overview of the Cocos nucifera transcriptome from mixed tissue samples. Using Illumina sequencing, we obtained 54.9 million short reads and conducted de novo assembly to obtain 57,304 unigenes with an average length of 752 base pairs. Sequence comparison between assembled unigenes and released cDNA sequences of Cocos nucifera and Elaeis guineensis indicated that the assembled sequences were of high quality. Approximately 99.9% of unigenes were novel compared to the released coconut EST sequences. Using BLASTX, 68.2% of unigenes were successfully annotated based on the Genbank non-redundant (Nr) protein database. The annotated unigenes were then further classified using the Gene Ontology (GO), Clusters of Orthologous Groups (COG) and Kyoto Encyclopedia of Genes and Genomes (KEGG) databases. Conclusions/Significance Our study provides a large quantity of novel genetic information for Cocos nucifera. This information will act as a valuable resource for further molecular genetic studies and breeding in coconut, as well as for isolation and characterization of functional genes involved in different biochemical pathways in this important tropical crop species.
