Toll-like receptor 4 mediates microglial activation and production of inflammatory mediators in neonatal rat brain following hypoxia: role of TLR4 in hypoxic microglia

Abstract: Background Hypoxia induces microglial activation which causes damage to the developing brain. Microglia derived inflammatory mediators may contribute to this process. Toll-like receptor 4 (TLR4) has been reported to induce microglial activation and cytokines production in brain injuries; however, its role in hypoxic injury remains uncertain. We investigate here TLR4 expression and its roles in neuroinflammation in neonatal rats following hypoxic injury. Methods One day old Wistar rats were subjected to hypoxia for 2 h. Primary cultured microglia and BV-2 cells were subjected to hypoxia for different durations. TLR4 expression in microglia was determined by RT-PCR, western blot and immunofluorescence staining. Small interfering RNA (siRNA) transfection and antibody neutralization were employed to downregulate TLR4 in BV-2 and primary culture. mRNA and protein expression of tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1β) and inducible nitric oxide synthase (iNOS) was assessed. Reactive oxygen species (ROS), nitric oxide (NO) and NF-κB levels were determined by flow cytometry, colorimetric and ELISA assays respectively. Hypoxia-inducible factor-1 alpha (HIF-1α) mRNA and protein expression was quantified and where necessary, the protein expression was depleted by antibody neutralization. In vivo inhibition of TLR4 with CLI-095 injection was carried out followed by investigation of inflammatory mediators expression via double immunofluorescence staining. Results TLR4 immunofluorescence and protein expression in the corpus callosum and cerebellum in neonatal microglia were markedly enhanced post-hypoxia. In vitro, TLR4 protein expression was significantly increased in both primary microglia and BV-2 cells post-hypoxia. TLR4 neutralization in primary cultured microglia attenuated the hypoxia-induced expression of TNF-α, IL-1β and iNOS. siRNA knockdown of TLR4 reduced hypoxia-induced upregulation of TNF-α, IL-1β, iNOS, ROS and NO in BV-2 cells. TLR4 downregulation-mediated inhibition of inflammatory cytokines in primary microglia and BV-2 cells was accompanied by the suppression of NF-κB activation. Furthermore, HIF-1α antibody neutralization attenuated the increase of TLR4 expression in hypoxic BV-2 cells. TLR4 inhibition in vivo attenuated the immunoexpression of TNF-α, IL-1β and iNOS on microglia post-hypoxia. Conclusion Activated microglia TLR4 expression mediated neuroinflammation via a NF-κB signaling pathway in response to hypoxia. Study on Advanced Water Injection Time in Low Permeability Reservoir  [PDF]

Abstract: A certain formation pressure level must be kept due to the threshold pressure of the low-permeability reser-voir during the seepage. Advanced water injection can keep the formation pressure at a higher level and keep a higher pressure gradient, which is an effective way to develop low-permeability reservoir. Based on the mechanism of advanced water injection and characteristic of porous flow in low permeability reservoir, a seepage model considering threshold pressure gradient is established to determine the formation pressure distribution at anytime as the water is injected at a constant speed. The optimum water injection time for the advanced water injection technology can be determined by using this model. The calculated result coincides basically with the numerical simulation result, which indicates that the model put forward in this paper is feasible.

Abstract: This paper discusses the trend of the emotional labor research object from the external customers to the internal customers of the organization, and separately reviews the study of subordinates’ emotional labor on supervisors and the study of supervisors’ emotional labor on subordinates. Finally, the paper identifies several future research directions, including the measurement of emotional labor between supervisors and subordinates, the research of the relationship between positive leadership behavior and subordinates’ emotional labor, and the integrated research between supervisors’ and subordinates’ emotional labor.
On Translatability of English Idioms

On Integrated Translation Approach of English Idioms

Abstract: Idioms are the gems of a language. Translating idioms is not an easy task because not all English idioms have equivalents in Chinese idioms due to linguistic, especially cultural differences. Research indicates that there are six translation tactics in translating English idioms into Chinese: literal translation, free translation, compensatory translation, explanational translation, borrowing, integrated approach. This paper aims to propose the integrated translation approach, one indispensable and important tactic to Chinese translation of English idioms based on Nida’s dynamic equivalence. Obviously, a reasonable choice and the quality of translation are decided by the translator’s cultural awareness and creativity.
On Literal Translation of English Idioms

Abstract: There are six translation tactics in translating English idioms into Chinese: literal translation, compensatory translation, free translation, explanational translation, borrowing, integrated approach. Each tactics should be reasonably employed in the process of translating, so as to keep the flavor of the original English idioms as well as to cater for the Chinese readers. In this paper, one of the tactics: literal translation will be discussed, which is the most commonly used tactics in translating English idioms.
Notch-1 Signaling Regulates Microglia Activation via NF-κB Pathway after Hypoxic Exposure In Vivo and In Vitro

Abstract: Neuroinflammation mediated by the activated microglia is suggested to play a pivotal role in the pathogenesis of hypoxic brain injury; however, the underlying mechanism of microglia activation remains unclear. Here, we show that the canonical Notch signaling orchestrates microglia activation after hypoxic exposure which is closely associated with multiple pathological situations of the brain. Notch-1 and Delta-1 expression in primary microglia and BV-2 microglial cells was significantly elevated after hypoxia. Hypoxia-induced activation of Notch signaling was further confirmed by the concomitant increase in the expression and translocation of intracellular Notch receptor domain (NICD), together with RBP-Jκ and target gene Hes-1 expression. Chemical inhibition of Notch signaling with N-[N-(3,5-difluorophenacetyl)-1-alany1- S-phenyglycine t-butyl ester (DAPT), a γ-secretase inhibitor, effectively reduced hypoxia-induced upregulated expression of most inflammatory mediators. Notch inhibition also reduced NF-κB/p65 expression and translocation. Remarkably, Notch inhibition suppressed expression of TLR4/MyD88/TRAF6 pathways. In vivo, Notch signaling expression and activation in microglia were observed in the cerebrum of postnatal rats after hypoxic injury. Most interestingly, hypoxia-induced upregulation of NF-κB immunoexpression in microglia was prevented when the rats were given DAPT pretreatment underscoring the interrelationship between Notch signaling and NF-κB pathways. Taken together, we conclude that Notch signaling is involved in regulating microglia activation after hypoxia partly through the cross talk between TLR4/MyD88/TRAF6/NF-κB pathways. Therefore, Notch signaling may serve as a prospective target for inhibition of microglia activation known to be implicated in brain damage in the developing brain.
Down-Regulation of miR-129-5p Inhibits Growth and Induces Apoptosis in Laryngeal Squamous Cell Carcinoma by Targeting APC

Abstract: miRNAs regulate gene expression and are key mediators of tumourigenesis. miR-129 has diverse effects in tumours, but its role in laryngeal squamous cell carcinoma (LSCC) remains unknown. This article focuses on the role of miR-129-5p in LSCC. We show miR-129-5p is upregulated in primary LSCC tumours and correlated with advanced disease. Down-regulating miR-129-5p suppressed cell proliferation and migration, and caused cell cycle arrest in Hep-2 cell lines. Downregulation of miR-129-5p alone is sufficient to induce apoptosis both in vivo and in vitro. Moreover, the growth of LSCC xenograft exposed to miR-129-5p antisense oligonucleotides (ASO) in BALB/c mice was markedly inhibited. In addition, we found that miR-129-5p targeted adenomatous polyposis coli (APC) to release inhibition of Wnt signalling causing cell growth and tumourigenesis. Our results suggest miR-129-5p functions as an oncogene in LSCC by repressing APC and is a potential therapeutic target for LSCC.

Abstract: Prepaid energy meters have been widely adopted by utilities in different countries across the world as an innovative solution to the problem of affordability and consumption management. However, the present smart card based systems have some inherent problems like added cost, low availability and lack of security. In the future Smart Grid paradigm, use of smart meters can completely overhaul these prepaid systems by introducing centralized accounting, monitoring and credit-control functions using state-of-the-art telecommunication technologies like WiMAX. In this paper we propose a prepaid smart metering scheme for Smart Grid application based on centralized authentication and charging using the WiMAX prepaid accounting model. We then discuss its specific application to Demand Response and Roaming of Electrical Vehicles.
