Abstract: The Effects of sampling schemes on construct method for core collection of Canarium album L.based on molecular markers data were studied on the basis of ISSR results of 64 germplasm materials of C.album from eastern Guangdong,and using UPGMA cluster method according to SM and Nei & Li genetic distance by stepwise clustering.The impact of sampling strategies,such as P,S,L and G,on the construction of core collection with different groupings was discussed.Compared with the number of polymorphic loci,percentage of polymorphic loci,observed number of alleles,effective number of alleles,Nei’s genetic diversity index and Shannon’s information index of different sample groups,16 samples were finally chosen as the core collection according to genetic proportion strategy based on Nei & Li genetic distance.The core collection retained 25% of germplasm of the initial samples,and the retention ratio of the number of polymorphic loci and percentage of polymorphic loci was 92.93% and 98.31%,respectively,and the observed number of alleles,effective number of alleles,Ne’is genetic diversity indexand Shannon’s information indexreached 99.26%,102.56%,107.39% and 106.29%,respectively.It suggested that the core collection constructed by this method could goodly represent the genetic diversity of initial germplasm bank.

Abstract: The mixture of five yeast strains obtained from soil could remove about 85% TOC of oil-rich wastewater in batch test. While the highest MLSS was obtained at an N:C of 1:5, the oil removal decreased with the increase of N:C during yeast sludge cultivation. Ammonium chloride was the best nitrogen source for yeast cultivation from the viewpoint of yeast growth and oil utilization. An ammonia concentration of over 1300 mg/L led to mass death of yeast at a pH of 5. The ammonia concentration should be controlled at a level of 1000 mg/L or lower.

Abstract: Knee osteoarthritis (OA) is a progressive disease in which the cartilage (cushioning between joints) wears away. As the cartilage wears away, it becomes frayed and rough, and the protective space between the bones decreases. This causes rubbing of bones, resulting in pain, swelling, stiffness, decreased ability to move and formation of bone spurs. Osteoarthritis develops slowly and the pain it causes worsens over time. Although there is no cure, there are many treatment options available to help manage pain and keep people staying active. Intra-articular (IA) hyaluronic acid (HA) injection is one of the treatment options. Hyaluronic acid is a polysaccharide, an anionic, non-sulfated glycosaminoglycan and is a naturally occurring substance found in the synovial fluid surrounding joints. It acts as a lubricant to enable bones to move smoothly over each other and as a shock absorber for joint loads. People with knee osteoarthritis have lower concentration and altered volume of hyaluronic acid than normal in their joints. Due to lower concentration and volume, exogenous hyaluronic acid may be injected into the knee joint. Treatment with exogenous hyaluronic acid contributes to restore the elastic and viscous properties of the synovial fluid, pain reduction and functional improvement. It is indicated for patients who have failed to respond adequately to conservative non-pharmacological therapy and simple analgesic.

Abstract: The old northeast industrial base has been the cradle of China after Liberation industry and has made the significant contribution to speed up the process of industrialization in China. The 16th National Party Congress definitely put forward the strategy of revitalizing the old industrial bases in Northeast China. Undoubtedly it can bring new lease and vitality for the development of the old industrial bases in Northeast China in the future. To begin with the study of the contents of a new road of industrialization, the article focuses on the necessity of taking a new road of industrialization to revitalize Northeast China and put forward the countermeasures for it according to analyze the current status. Key words: revitalization of Northeast China, new industrialization, resources, countermeasures Résumé: L’ancienne base industrielle nord-est a été le berceau de l’industrie de Chine après la Libération et a donné une contribution signifiante à l’accélération du processus d’industrialisation chinoise. Le 16e Congrès national du Parti a mis en avant définitivement la stratégie de revitalisation des anciennes bases industrielles dans le Nore-Est de Chine. Sans aucun doute, cette stratégie peut amener la vitalité et un nouveau commencement au développement des ces anciennes bases industrielles dans le futur. Débutant par l’étude du contenu de la nouvelle voie d’industrialisation, le présent article se focalise sur la nécessité de prendre une nouvelle voie d’industrialisation pour redresser le Nord-Est de Chine et propose des contre-mesures correspondantes en vertu de la situation actuelle. Abstract: Electrospray ionization mass spectrometry (ESI-MS) has become one of the most popular and powerful technologies to identify and quantify individual lipid species in lipidomics. Meanwhile, quantitative analysis of lipid species by ESI-MS has also become a major obstacle to meet the challenges of lipidomics. Herein, we discuss the principles, advantages, and possible limitations of different mass spectrometry-based methodologies for lipid quantification, as well as a few practical issues important for accurate quantification of individual lipid species. Accordingly, accurate quantification of individual lipid species, one of the key challenges in lipidomics, can be practically met.

Abstract: In order to perform the regulation and functional study and the screening of potassium uptake genes, the high affinity K+ uptake genes TRK1 and TRK2 in Saccharomyces cerevisiae were replaced with URA3 and HIS3 alternately using the methods of homologous recombination. URA3 and HIS3 of S. Cerevisiae were used as marker gene, the defective K+ uptake TRK1 and TRK2 mutant of S. Cerevisiae were selected in the basic medium without Histidine and Uracil.

Abstract: The TaqMan primers and probes for real-time PCR were designed according to the sequence of 35S promoter, NOS terminator,GUS reporter and NPTII selection gene. The most suitable TaqMan primer and probe, PCR system and PCR condition were suggested from the experiments on GMT real-time PCR quantity detecting methods using DNA Engine Opticon 2 real-time PCR. The Z-score from CORESTA genetic modified tobacco testing ring trial confirmed the accuracy and stability of this methods.

Abstract: The DNA fragment sized 2139bp which sequence is the same with AtKup1 gene from Arabidopsis thalianna was used as the templates to do DNA family shuffling. The shuffled AtKup1 gene library was expressed in the mutant of S. cerevisae in which potassium transporter gene TRK1 and TRK2 were knocked out by homologous recombination. Then the screening was carried out in the low potassium media containing 5.0 mM KCl and no histidine in it. it was found that both of diverse and wild AtKup1 genes can rescue the trk1 trk2 yeast mutant strain on low K+] medium. The growth of 2 clones yeast contained diverse AtKup1 were beter than that of AtKup1 wild gene transformant. The sequencing results of the shuffled Atkup1 showed that there were 2 nucleotides had changed and resulted in 2 amino acid variations in it compared with original AtKup1. The potassium uptaking capacity of shuffled Atkup1 gene increased significantly when it was transformed into tobacco.
Radiofrequency ablation as a treatment for hilar cholangiocarcinoma

Abstract: AIM: To explore the role of radio-frequency ablation (RFA) as a treatment for hilar cholangiocarcinoma.METHODS: Eleven patients with obstructive cholestasis underwent Computed Tomography (CT) examination, occupying lesions were observed in the hepatic hilar region in each patient. All lesions were confirmed as cholangioadenocarcinoma by biopsy and were classified as type III or IV by percutaneous transhepatic cholangiography. Patients were treated with multiple electrodes RFA combined with other adjuvant therapy. The survival rate, change of CT attenuation coefficient of the tumor and tumor size were studied in these patients after RFA.RESULTS: In a follow-up CT scan one month after RFA, a size reduction of about 30% was observed in six masses, and two masses were reduced by about 20% in size, three of the eleven masses remained unchanged. In a follow-up CT scan 6 mo after RFA, all the masses were reduced in size (overall 35%), in which the most significant size reduction was 60%. The survival follow-up among these eleven cases was 18 mo in average. Ongoing follow-up showed that the longest survival case was 30 mo and the shortest case was 10 mo.CONCLUSION: RFA is a microinvasive and effective treatment for hilar cholangiocarcinoma.
