Role of Wnt canonical pathway in hematological malignancies

Abstract: Wnt canonical signaling pathway acts a significant part in embryonic development and in maintenance of organs and tissues in adults. In the past two decades, medical scientists have devoted themselves to understanding the cellular and molecular mechanisms of Wnt signaling. A lot of studies indicate that Wnt canonical pathway involves in the pathogenesis of a range of disease including many kinds of carcinomas. Hematological malignancies are the types of carcinoma that affect blood, bone marrow and lymph nodes. They may derive from either of the two major blood cell lineages: myeloid and lymphoid cell lines. The incidence of hematological malignancies has been increasing steadily in the world for the past years, but their etiology and pathogenesis has not been well understood involving areas of chromosome aberrations, apoptosis inhibition, abnormal activation of signaling pathways, angiogenesis, et al. In this review, we focus on the role of Wnt canonical signaling in carcinomas, especially in hematological malignancies, and then disclose potential therapeutic opportunities of this pathway in hematological malignancies.Wnt signaling pathways are categorized as "canonical "and "non-canonical" Wnt pathways, which are β-catenin-dependent and β-catenin-independent signaling pathways, respectively. Here we will emphatically point out the role of Wnt canonical pathway in hematological malignancies. A simplified model of Wnt canonical pathway is delineated in Fig. 1. Wnts is a group of secreted cysteine-rich glycoproteins, which includes at least 19 identified members in diverse species ranging from round worm and insects to human[1]. In the absence of a Wnt ligand binding to its receptor complex, the cytoplasmic β-catenin is degraded by the "destruction complex". Metadherin Contributes to the Pathogenesis of Diffuse Large B-cell Lymphoma

Abstract: Background Metadherin (MTDH) has been demonstrated as a potentially crucial mediator of various types of human malignancies. However, the expression and role of MTDH in diffuse large-B-cell lymphoma (DLBCL) have not been reported yet. This study aimed to illuminate the role of MTDH in the pathogenesis of DLBCL. Methodology/Principal Findings A remarkable elevation of MTDH on mRNA level was detected in DLBCL tissues by quantitative polymerase chain reaction (PCR). Using Western-blot analysis we found that the expression of MTDH protein was significantly upregulated in DLBCL cell lines and DLBCL tissues compared with peripheral blood mononuclear cells (PBMCs) from healthy samples and tissues from patients of reactive hyperplasia of lymph node. The results showed high expression of MTDH in 23 of 30 (76.67%) DLBCL tissues by using immunohistochemical analysis and the over expression of MTDH was strongly correlated to the clinical staging of patients with DLBCL (P<0.05). Furthermore, the finding suggested that the increase of MTDH in DLBCL cells could distinctly enhance cell proliferation and inhibit cell apoptosis; meanwhile, inhibition of MTDH expression by specific siRNA clearly enhanced LY8 cell apoptosis. Upregulation of MTDH elevated the protein level of total β-catenin and translocation of β-catenin to the nucleus directly or indirectly. Knockdown of MTDH decreased the level of total, cytoplasmic β-catenin and reduced nuclear accumulation of β-catenin protein. This indicated that the function of MTDH on the development of DLBCL was mediated through regulation of Wnt/β-catenin signaling pathway. Conclusions/Significance Our results suggest that MTDH contributes to the pathogenesis of DLBCL mediated by activation of Wnt/β-catenin pathway. This novel study may contribute to further investigation on the useful biomarkers and potential therapeutic target in the DLBCL patients.

In this paper, the time-dependent invariant of the Dirac equation with time-dependent linear potential has been constructed in non-commutative phase space. The corresponding analytical solution of the Dirac equation is presented by Lewis-Riesenfield invariant method.

Numerical Studies on Flow Fields Around Buildings in an Urban Street Canyon and Cross-Road

Abstract: The questions on how vortices are constructed and on the relationship between the flow patterns and concentration distributions in real street canyons are the most pressing questions in pollution control studies. In this paper, the very large eddy simulation (VLES) and large eddy simulation (LES) are applied to calculate the flow and pollutant concentration fields in an urban street canyon and a cross-road respectively. It is found that the flow separations are not only related to the canyon aspect ratios, but also with the flow velocities and wall temperatures. And the turbulent dispersions are so strongly affected by the flow fields that the pollutant concentration distributions can be distinguished from the different aspect ratios, flow velocities and wall temperatures.
Distribution and Molecular Characterization of β-Glucans from Hull-Less Barley Bran, Shorts and Flour

Abstract: Six hull-less barley cultivars widely grown in China were roller-milled to produce bran, shorts and flour fractions. The distribution and molecular characteristics of β-glucans from the three roller-milled fractions were investigated. The β-glucan contents in the six hull-less barley cultivars varied from 4.96% to 7.62%. For all the six cultivars, the shorts fraction contained the highest concentration of β-glucan (8.12–13.01%), followed by bran (6.15–7.58%) and flour (2.48–2.95%). Crude β-glucans were prepared from the three roller-milled fractions using aqueous sodium carbonate (pH 10). These preparations contained 45.38–71.41% β-glucan, 10.81–17.26% arabinoxylan, 2.6–9.6% protein, 2.7–9.0% starch, and 5.23–9.68% ash. Purification using a-amylase and β-xylanase in combination with pH adjustment and dialysis produced high purity β-glucan preparations (91–95%). The molecular weight (Mw) of β-glucan preparations from roller-milled fractions ranged from 117,600 to 852,400 g/mol. β-Glucan from flour had higher Mw than those from shorts and bran within the same cultivar, and β-glucan preparations from bran had the lowest Mw.
Molecular Characterization of Arabinoxylans from Hull-Less Barley Milling Fractions

Abstract: Arabinoxylans were prepared from different hull-less barley milling fractions (bran, shorts and flour). The yields of hull-less bran arabinoxylan (HBB-AX), shorts arabinoxylan (HBS-AX) and flour arabinoxylan (HBF-AX) were 8.42%, 4.08% and 2.13% respectively. Sugar composition analysis showed that arabinose and xylose were the main sugars. HBF-AX had the highest Ara/Xyl ratio, followed by HBS-AX and HBB-AX. Size exclusion chromatography analysis (HPSEC) showed that HBF-AX had the highest molecular weight, followed by HBS-AX and HBB-AX, which had the lowest molecular weight. Aqueous solutions of HBB-AX, HBS-AX and HBF-AX had higher molecular weight(Mw) than in 0.5 mol/L NaOH and 1.0 mol/L NaOH. Dynamic light scattering analysis (DLS) showed that HBB-AX, HBS-AX and HBF-AX had existed in two states in distilled water, 0.5 mol/LNaOH, 1.0 mol/L NaOH, and DMSO/H2O (90/10), an unaggregated state and an aggregated state, with the latter predominating.
Simulation of wind gust structure in the atmospheric boundary layer with Lattice Boltzmann Method

Abstract: Cold fronts occur in northern East Asia during winter and spring. After cold frontal passage, airflow is downward and accompanying strong winds fluctuate significantly; this is termed wind gusts. Analysis of observation data shows that wind gust structure has coherent characteristics. This is important for entrainment of spring dust storms into the upper boundary layer, where they are transported great distances. The Lattice Boltzmann Method (LBM) is a computational fluid technique based on the Boltzmann transport equation. The LBM has been used to study complex motion such as turbulence, because it describes motion at the micro level. In this paper, Large eddy simulation is introduced in the LBM, enabling simulation of turbulent flow in the atmospheric boundary layer. The formation and development of wind gusts are simulated, and a coherent structure with a combination of wave and vortex is obtained. This explains the mechanism of soil erosion and sand entrainment by the coherent structure of wind gusts.
Stochastic modeling the effect of wind gust on dust entrainment during sand storm

Abstract: After the passage of a cold front, spring in northern China the outbreak of strong wind is often accompanied by dust emissions. Owing to the coherent structure of wind gust, dust particles can effectively overcome the systematic descending air motion and penetrate into the middle and upper levels of the atmospheric boundary layer, and then propagate further and diffuse into the troposphere where ascending air motion prevails. Here, we consider the coherent structure of wind gust in LS models, and construct a model suitable for the dust entrainment under the dust-storm canopy. With the parameter of gust, we simulate the dust entrainment during dust storms, and test that the coherent structures of wind gust make the sand particles to reach the upper of the boundary layer.

At present, a sound intellectual property management and service system have not been established yet in Chinese colleges and universities. Meanwhile, the lost of existing intellectual property in universities due to small amount, low conversion rate, low technology content and weak brand awareness, hinders the positive effect of intellectual property and affects its transformation and application in China. This article introduces the current problems with intellectual property in universities and gives some countermeasures to promote the use of intellectual property in universities for a better regional economic service.
