Effects of cadmium stress on seed germination and seedling growth of Elymus dahuricus infected with the Neotyphodium endophyte

Abstract: Various cadmium (Cd) concentrations (0, 50, 100, 200 and 300 μmol L 1) affected Elymus dahuricus seed germination, seedling growth, antioxidative enzymes activities (AEA), and amounts of malondialdehyde (MDA) and proline present. These influences were determined for separate E. dahuricus cohorts known to be either infected (E+) or non-infected (E ) by a Neotyphodium endophyte. Under high Cd concentrations (100, 200 and 300 μmol L 1), E+ specimens showed a significantly (P<0.05) higher germination rate and index, as well as higher values for shoot length, root length and dry biomass. However, the germination rate and index, root length and dry weight did not show a significant (P<0.05) difference under the low Cd concentrations (0 and 50 μmol L 1). AEA and proline content increased, as did MDA content, in the E+ (vs. E ) specimens under high Cd concentrations. There was no significant (P>0.05) difference under low Cd concentrations. Endophyte infection was concluded to be of benefit to E. dahuricus exposed to high Cd concentrations.
Effects of cutting frequency and height on alkaloid production in endophyte-infected drunken horse grass (Achnatherum inebrians)

Abstract: An evaluation was performed on the influence of mowing height (2.5, 5.0 or 7.5 cm) and mowing frequency (weekly or fortnightly) on levels of ergot alkaloids (ergine and ergonovine) formed in drunken horse grass, Achnatherum inebrians, grown under greenhouse conditions. Samples were taken monthly and alkaloids were extracted and analyzed by reverse-phase HPLC. Alkaloid levels increased more or less linearly with plant age over the first four months following establishment. Levels were higher in samples cut fortnightly compared to those cut weekly, and were higher when plants were cut at a mowing height of 7.5 cm vs. 2.5 cm. In most cases, the highest alkaloid levels observed were almost three times those of the lowest. If plant protection applications are developed for the endophytic fungus, Neotyphodium gansuense, it will be necessary to be aware of the potential role of plant husbandry practices (e.g., defoliation frequency and intensity) for reducing or enhancing levels of plant alkaloids. Similar husbandry factors may affect alkaloid levels in other Neotyphodium-grass associations, which would be interesting for further study.
Rural Labor Force Transfer Training Effects Evaluation by Matching Methods: Evidence from Yunnan Province of China  [PDF]

Abstract:

Rural labor force transfer training is one of important poverty alleviation measures in China. This paper describes training participation situation and evaluates training effects by matching methods in the case of coastal autonomous minority nationality areas of Yunnan province by using 2053 rural household data. The result shows that the average training participation from 2006 to 2008 is 26.39 percent. In addition, ATE is 18.33 percent, TT is 18.99 percent, TUT is 18.09 percent. And,TUT < ATE < TT. It demonstrates that the rural labor force transfer training program is effective and well-directed in coastal autonomous minority nationality areas of Yunnan province of China.

Cloning, Sequencing and Analysis of Melatonin Receptor Subtype MT1 in Hypothalamus-Pituitary-Gonadal Axis and Pineal of Female Bactrian Camel

Abstract: It is well known that melatonin is a coordinating signal for mammalian reproduction. In order to confirm the presence of melatonin receptors in hypothalamus-pituitary-gonadal axis and pineal of female Bactrian camel, the researchers used a Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR) procedure to examine receptor MT1 expression. The length of MT1 gene was 452 bp. RT-PCR assaying revealed the presence of the mt1 (Mel1a) melatonin receptor subtype in reproduction axis and pineal which were obtained from the slaughterhouse in the Ningxia Autonomous Region, China. Sequence has been confirmed a high identity (above 85%) with melatonin receptor MT1 of other mammal known in GenBank. Comparing with other tissue s sequences, one base substitution changed the 108th TTC codon (encoding Phenylalanine) to TAC (Tyrosine) on hypothalamus. Although, there are base substitutions in pineal s gene, encoded amino acid are coincident with pituitary and ovary. The current results, the expression of MT1 receptor mRNA in brain and ovary, suggest that melatonin regulate reproduction function through not only neuroendocrine but also directly acting on the ovary in Bactrian camel.
sgRNAcas9: A Software Package for Designing CRISPR sgRNA and Evaluating Potential Off-Target Cleavage Sites

Abstract: Although the CRISPR/Cas9/sgRNA system efficiently cleaves intracellular DNA at desired target sites, major concerns remain on potential “off-target” cleavage that may occur throughout the whole genome. In order to improve CRISPR-Cas9 specificity for targeted genome editing and transcriptional control, we describe a bioinformatics tool “sgRNAcas9”, which is a software package developed for fast design of CRISPR sgRNA with minimized off-target effects. This package consists of programs to perform a search for CRISPR target sites (protospacers) with user-defined parameters, predict genome-wide Cas9 potential off-target cleavage sites (POT), classify the POT into three categories, batch-design oligonucleotides for constructing 20-nt (nucleotides) or truncated sgRNA expression vectors, extract desired length nucleotide sequences flanking the on- or off-target cleavage sites for designing PCR primer pairs to validate the mutations by T7E1 cleavage assay. Importantly, by identifying potential off-target sites in silico, the sgRNAcas9 allows the selection of more specific target sites and aids the identification of bona fide off-target sites, significantly facilitating the design of sgRNA for genome editing applications. sgRNAcas9 software package is publicly available at BiooTools website (www.biootools.com) under the terms of the GNU General Public License.
Heritable Multiplex Genetic Engineering in Rats Using CRISPR/Cas9

Abstract: The CRISPR/Cas9 system has been proven to be an efficient gene-editing tool for genome modification of cells and organisms. Multiplex genetic engineering in rat holds a bright future for the study of complex disease. Here, we show that this system enables the simultaneous disruption of four genes (ApoE, B2m, Prf1, and Prkdc) in rats in one-step, by co-injection of Cas9 mRNA and sgRNAs into fertilized eggs. We further observed the gene modifications are germline transmittable, and confirmed the off-target mutagenesis and mosaicism are rarely detected by comprehensive analysis. Thus, the CRISPR/Cas9 system makes it possible to efficiently and reliably generate gene knock-out rats.
Molecular Cloning, Bioinformatics Analysis and Expression of Insulin-Like Growth Factor 2 from Tianzhu White Yak, Bos grunniens

Abstract: The IGF family is essential for normal embryonic and postnatal development and plays important roles in the immune system, myogenesis, bone metabolism and other physiological functions, which makes the study of its structure and biological characteristics important. Tianzhu white yak ( Bos grunniens) domesticated under alpine hypoxia environments, is well adapted to survive and grow against severe hypoxia and cold temperatures for extended periods. In this study, a full coding sequence of the IGF2 gene of Tianzhu white yak was amplified by reverse transcription PCR and rapid-amplification of cDNA ends (RACE) for the first time. The cDNA sequence revealed an open reading frame of 450 nucleotides, encoding a protein with 179 amino acids. Its expression in different tissues was also studied by Real time PCR. Phylogenetic tree analysis indicated that yak IGF2 was similar to Bos taurus, and 3D structure showed high similarity with the human IGF2. The putative full CDS of yak IGF2 was amplified by PCR in five tissues, and cDNA sequence analysis showed high homology to bovine IGF2. Moreover the super secondary structure prediction showed a similar 3D structure with human IGF2. Its conservation in sequence and structure has facilitated research on IGF2 and its physiological function in yak.

Abstract:

This paper designs methods of product differences testing and preference testing by using triangle taste tests data. Binomial distribution theory and hypothesis test method are employed since that the existing statistical inference methods of triangle taste tests methods for sensory and quality analysis of cigarette products have some shortcomings. Finally, two case studies are provided, which have very important managerial implication to cigarette enterprises.

Abstract: Activity of separase, a cysteine protease that cleaves sister chromatid cohesin at the onset of anaphase, is tightly regulated to ensure faithful chromosome segregation and genome stability. Two mechanisms negatively regulate separase: inhibition by securin and phosphorylation on serine 1121. To gauge the physiological significance of the inhibitory phosphorylation, we created a mouse strain in which Ser1121 was mutated to Ala (S1121A). Here we report that this S1121A point mutation causes infertility in mice. We show that germ cells in the mutants are depleted during development. We further demonstrate that S1121A causes chromosome misalignment during proliferation of the postmigratory primordial germ cells, resulting in mitotic arrest, aneuploidy, and eventual cell death. Our results indicate that inhibitory phosphorylation of separase plays a critical role in the maintenance of sister chromatid cohesion and genome stability in proliferating postmigratory primordial germ cells.
