Research Progresses of Circulating Tumor Cells in Diagnosis and Treatment of ?Early Lung Cancer

Abstract: As one of the important liquid biopsies, circulating tumor cells (CTCs) shows more and more clinical values in the treatment of lung cancer such as diagnostic screening, treatment evaluation, postoperative monitoring, and prognosis predicting etc. A large number of small pulmonary nodules patients are detected when screening the high risk population of lung cancer. However, small lung nodules are not equal to lung cancer, and 90%-95% of them are benign lesion, therefore, to accurately and correctly differentiate whether it is benign or malignant when patients firstly detected and treat a small pulmonary nodule is become a new opportunity and challenge for clinician. With the improvement of CTCs detection technology, whether it will play an important role in early differential diagnosis of lung cancer. And whether it will have clinical significance to early lung cancer surgery operations. These require further researches and explorations so as to achieve clinical transformation in the future.
Variations in Serum CEA and CYFRA21-1 Levels Before and After Surgery Facilitate Prognosis of Non-small Cell Lung Cancer Patients

Abstract: Background and objective Serum carcinoembryonic antigen (CEA) and the soluble fragment of cytokeratin 19 (CYFRA21-1) are important tumor markers (TMs) in the preoperative examination of patients with non-small cell lung cancer (NSCLC). However, the prognostic role of these markers in NSCLC patients remains controversial. The aim of the study was to investigate the clinical significance of serum CEA variances and CYFRA21-1 levels for the prognosis of NSCLC patients following surgery. Methods This retrospective study investigated the clinical records and follow-up sessions of 175 patients with NSCLC who accepted surgery and adjuvant chemotherapy. Patients were subdivided into groups based on serum CEA and CYFRA21-1 levels. Survival analysis was conducted using Kaplan-Meier method for each group. The prognostic factor was evaluated using Cox proportional hazards model. Results The overall survival (OS) of patients with high preoperative CEA or CYFRA21-1 levels was lower than that of patients with normal preoperative CEA or CYFRA21-1 levels. The OS displayed a significant difference (P=0.001) between groups with high and normal preoperative CYFRA21-1. Compared with groups exhibiting normal preoperative and postoperative levels of CEA or CYFRA21-1, the OS was shorter for groups with high preoperative and postoperative levels of CEA or CYFRA21-1. The difference of the paired groups was significant (P<0.05). Compared with the groups with normal preoperative and postoperative levels of CEA and CYFRA21-1, the OS was lower for the groups with high preoperative and postoperative levels of CEA and CYFRA21-1, which indicated a significant difference (P<0.001). The CEACYFRA211 (HHHH), CEACYFRA211 (NNHH), CYFRA21-1 (HH), CEA (HH), and male gender were identified as independent prognostic factors (P<0.05). Conclusion This study suggested that the prognosis of NSCLC patients was not significantly satisfactory if preoperative and postoperative level of serum CEA or CYFRA21-1 was higher than standard value, especially if the preoperative and postoperative levels of CYFRA21-1 and CEA were higher than the standard values. The measurement of preoperative and postoperative levels of CYFRA21-1 and CEA proved helpful for the prognosis of patients with NSCLC.

We discuss the application of using Coh$\rho$ process to constrain the neutrino flux with the proposed Long-Baseline Neutrino Experiment Near Detector, the high resolution Straw Tube Tracker.
Characterization of Microsatellite Markers and Their Application to Genetic Diversity Analysis of Brachypodium sylvaticum var. breviglume from Yunnan, China  [PDF]

Abstract:

Southwestern China is abundant in a large species diversity of grasses and Brachypodium sylvaticum var. breviglume is unquestionably appropriate to serve as an ideal model to investigate the adaptation, evolution and diversification of grasses in this region. In this study, a total of 37 microsatellite markers were screened and genetic variation was estimated for 100 individuals from the five natural populations of the species. Our results showed that seven of them were polymorphic for the five studied populations, while the other thirty were monomorphic. These seven SSR loci exhibited a high level of genetic diversity among populations, i.e. allele number varied from 5 to 24, with an average of 13.29; expected heterozygosity (He) ranged from 0.439 to 0.561, with an average of 0.488; percentage of polymorphic loci (PPL) ranged from 85.71% to 100%, with an average of 97. 14%. The FCA and UPGMA analyses revealed that the five populations were divided into three clusters. Our results indicate that these newly characterized SSR markers are useful for the exploration of genetic diversity and evolutionary history of the B. sylvaticum var. breviglume populations in Yunnan, China.

Cloning and Expression of a b-Glucosidase Gene umcel3G from Metagenome of Buffalo Rumen and Characterization of the Translated Product

Abstract: Metagenomic cosmid libraries containing 1.26 x 10(5) clones, covering about 4.8 x 10(6) kb metagenomic DNA of uncultured microorganisms from the contents of buffalo rumens were constructed, and 118 independent clones expressing beta-glucosidase activity were isolated from the libraries. Screening of these clones showed that eight clones expressed relatively higher beta-glucosidase activity at pH 5.0 and 37 degrees C. One out of the eight clones was subcloned. Sequencing analysis showed that an open reading frame (ORF) of 2223 bp, termed umcel3G, potentially encodes a beta-glucosidase. The encoded product shared highest homology with a beta-glucosidase from Bacillus sp. at 60% identity and 73% similarity. The umcel3G was over-expressed in Escherichia coli and the size of the translated product Umcel3G on SDS-PAGE was in agreement with the predicted molecular mass. Zymogram analysis showed that Umcel3G exhibited beta-glucosidase activity, confirming that this ORF encodes a beta-glucosidase. The Umcel3G, purified with Ni-NTA column, exhibited optimal activity at pH 6.0-6.5 and 45 degrees C. Certain ions such as Ca2+, Zn2+ had significant positive effect on the activity of Umcel3G. However, some ions such as Fe3+, Cu2+ gave significant inhibitory effect on the enzyme. The Ni-NTA purified recombinant beta-glucosidase Umcel3G had a specific activity of 22.8 IU/mg at pH4.5, 35 degrees C and at the presence of 5 mmol/L Ca2+, indicating that this enzyme has potential applications in the fermentative production of ethanol by simultaneous saccharification and cofermentation (SSCF) of lignocelluloses.

Abstract: 以木质纤维素为原料、应用同步糖化共发酵工艺发酵生产酒精时需要酸性中低温高活力纤维素酶包括b-葡萄糖苷酶。本工作分6次构建了水牛瘤胃未培养微生物宏基因组文库, 获得1.26×105个克隆, 文库含外源DNA的总长度 约为4.8×106 kb。从文库中筛选到118个表达b-葡萄糖苷酶活性的独立克隆。发现其中8个克隆表达的b-葡萄糖苷酶在pH5.0、37oC条件下活性较强。对其中一个克隆进行了亚克隆, 序列分析发现一个2223 bp的潜在的编码b-葡萄糖苷酶基因(umcel3G)的开放阅读框(ORF), 其编码产物的氨基酸序列与来自于 Bacillus sp.的一个b-葡萄糖苷酶同源性最高, 具有60%的一致性和73%的相似性。该ORF在E.coli中的表达产物Umcel3G的分子量与预测大小相似, 酶谱分析表明该表达产物具有b-葡萄糖苷酶活性, 证实该基因为一个b-葡萄糖苷酶基因。测定了用Ni-NTA纯化的Umcel3G的酶学特性, 其最适pH和最适温度分别为6.0~6.5和45oC。一些金属离子如Ca2+、Zn2+能显著提高该酶的酶活, 而另外一些金属离子如Fe3+、Cu2+能抑制Umcel3G的活性。在pH4.5、35oC和5 mmol/L的 Ca2+存在的条件下, 用Ni-NTA纯化的重组酶的比活为22.8 IU/mg, 说明该酶在用SSCF工艺发酵生产酒精中有潜在的应用价值。
Effect of La2O3 on microstructure, mechanical and tribological properties of Ni-W coatings

Abstract: In this paper, a Ni-W-La2O3 composite coating was prepared by the electrodeposition method. Microhardness tester and environmental scanning electron microscope equipped energy dispersive spectroscopy were employed to investigate the microhardness and the surface morphology of the composite coatings respectively, and the high temperature friction behavior and corrosion resistance of the coatings against molten glass were investigated by using a high temperature tribometer. The results show that La2O3 can refine the microstructure effectively, and make the element distribution uniform, which leads to the increase of average microhardness. La2O3 particulates can reduce the friction coefficient between the composite coating and glass during the sliding process at about 973 K largely, and the corrosion resistance of the La2O3 added Ni-W coatings is effectively improved compared with that of the non-added one, furthermore the mechanism of friction-reducing and anti-corrosion is also discussed.

Abstract: A self-healing mechanism in key management is an important means for large-scale clustering wireless sensor networks that enable non-revoked nodes use their private information and the received broadcast messages to recover the lost session keys on their own. In this study, we propose a dynamic self-healing key management scheme for large-scale clustering wireless sensor networks that is based on Exclusion Basis System (EBS). We use forward and backward key chains to form cluster session key chain for self-healing, take t-degree polynomial keys to replace the original keys used in EBS. The analysis shows that the proposed scheme has the properties of forward and backward secrecy and resisting to a collusion attack, which is suitable for resource-constrained wireless sensor networks.

Abstract: NO$\nu$A is an accelerator-based neutrino oscillation experiment which has a great potential to measure the last unknown mixing angle $\theta_{13}$, the neutrino mass hierarchy, and the CP-violation phase in lepton sector with 1) 700 kW beam, 2) 14 mrad off the beam axis, 3) 810 km long baseline. The Near Detector on the Surface is fully functioning and taking both NuMI and Booster beam data. The far detector building achieved beneficial occupancy on April 13. This proceeding will focus on the DAQ software system.
