Abstract:

Neural regeneration was once considered to be impossible, especially in the central nervous system where neural regeneration comprise the generation of new neurons, glia, axons, myelin, and synapses. Until recently, neural stem cells/neural progenitor cells (NSCs/NPCs) were identified from various areas of brain and brought hopes to the neural repair and regeneration. Tissue engineering has revolutionized the current neural regeneration technology and it has become a pioneering interdisciplinary field in the areas of biomedical research. Polyhydoxyalkanoate (PHA) as one of biodegradable material has been successfully used as tissue engineering materials. It has also been applied in nerve tissue engineering due to the high biocompatibility and low cytotoxicity. Over the past 10 years, different kinds of modification strategies have been undertaken to improve the properties of PHA to fit the requirements from various fields. Several members of PHA family have been attempted for neural regeneration. This article reviewed the recent modification strategies for improving the properties of PHA and highlighted the pioneer applications in neural regeneration.

Computational Identification of Novel MicroRNAs and Their Targets in Vigna unguiculata

Abstract: MicroRNAs (miRNAs) are a class of endogenous, noncoding, short RNAs directly involved in regulating gene expression at the posttranscriptional level. High conservation of miRNAs in plant provides the foundation for identification of new miRNAs in other plant species through homology alignment. Here, previous known plant miRNAs were BLASTed against the Expressed Sequence Tag (EST) and Genomic Survey Sequence (GSS) databases of Vigna unguiculata, and according to a series of filtering criteria, a total of 47 miRNAs belonging to 13 miRNA families were identified, and 30 potential target genes of them were subsequently predicted, most of which seemed to encode transcription factors or enzymes participating in regulation of development, growth, metabolism, and other physiological processes. Overall, our findings lay the foundation for further researches of miRNAs function in Vigna unguiculata.

Introduction MicroRNAs (miRNAs) are a class of endogenous, small, noncoding, single-stranded RNAs that act as posttranscriptional regulators in eukaryotes [1]. They have been reported to be located mostly within noncoding regions of genomes, and usually transcribed from RNA polymerase II promoters [2, 3]. The generation of mature miRNA is a complicated enzyme-catalyzed process, from the initial transcript pri-miRNA to the precursor (pre-miRNA) with a characteristic hairpin structure, then a miRNA duplex (miRNA?:?miRNA*) [4]. In the end, it is assembled to the RNA-induced silencing complex (RISC) to direct its activity on a target mRNA, depending on the degree of base-pairing between the miRNA and the responsive element and results in either cleavage or translational repression of the target mRNA. Perfect complementarity generally results in cleavage, such as in plants, whereas imperfect base-pairing leads to translational repression [4, 5]. MiRNA genes represent about 1%-2% of the known eukaryotic genomes and constitute an important class of fine-tuning regulators that are involved in several physiological or disease-associated cellular processes [6]. For example, studies in plants have revealed the key roles of miRNAs in diverse regulatory pathways, including growth, development, and defense response against every sort of stress [7–17]. Considering the importance of miRNAs in gene regulation, two major categories of approaches have been applied for miRNA investigation [1]. Abstract: Enoyl acyl carrier protein (ACP) reductase (FabI) is a potential target for the development of antibacterial agents. Three-dimensional quantitative structure-activity relationships (3D-QSAR) for substituted formamides series of FabI inhibitors were investigated using comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA) techniques. Pharmacophore and molecular docking methods were used for construction of the molecular alignments. A training set of 36 compounds was performed to create the 3D-QSAR models and their external predictivity was proven using a test set of 11 compounds. Graphical interpretation of the results revealed important structural features of the formamides related to the active site of FabI. The results may be exploited for further optimization of the design of new potent FabI inhibitors.
Ocular fundus manifestation of two patients following long-term chloroquine therapy: a case report

Abstract: Chloroquine has long been used in the treatment and prevention of malaria[1-4], and for a long time, the agent is still being used to treat other diseases. For example, due to its immunosuppressive effects, chloroquine has been used in the treatment of autoimmune disorders, such as rheumatoid arthritis and lupus erythematosus. Chloroquine has also been tested in clinical trials as an antiretroviral agent against HIV [5] and as a radiosensitizer and chemosensitizer for human cancer therapy [6]. However, the side effects of Chloroquine, including ocular toxicities, limit its long-term use [2-4,6].Herein we describe two patients who developed severe ocular toxicity following long-term chloroquine treatment. The clinical characteristics of their retinopathy were extensively investigated using various techniques, and are reported in this manuscript. We are not trying to provide information which would help in the early screening for chloroquine retinopathy, but to describe the typical manifestations of chloroquine retinopathy with some advanced new technology which have not been applied on this disease before, believing that it would provide a better understanding of chloroquine retinopathy.A 43-year-old woman had taken chloroquine for her rheumatoid arthritis for 16 years, and had received a total dose of approximately 800 g of the drug (250 mg daily for two years, then 250 mg alternative daily for 14 years). Her visual acuity had decreased gradually during the previous 5 years. At her latest examination, her visual acuity was 20/100 OD and hand motions OS, Goldmann applanation intraocular pressure was normal (OD16 mmHg, OS 16 mmHg), but the center visual field (10-2 Humphrey Visual Field Analyzer, Humphrey Instruments Inc, San Leandro, Calif) was lost, and the Goldmann visual field showed a temporal visual island. Embedding and Extracting Digital Watermark Based on DCT Algorithm  [PDF]

Abstract: The principle of digital watermark is the method of adding digital watermark in the frequency domain. The digital watermark hides the watermark in digital media, such as image, voice, video, etc., so as to realize the functions of copyright protection, and identity recognition. DCT for Discrete Cosine Transform is used to transform the image pixel value and the frequency domain coefficient matrix to realize the embedding and extracting of the blind watermark in the paper. After success, the image is attacked by white noise and Gaussian low-pass filtering. The result shows that the watermark signal embedded based on the DCT algorithm is relatively robust, and can effectively resist some attack methods that use signal distortion to destroy the watermark, and has good robustness and imperceptibility.
Suppression of Jasmonic Acid-Dependent Defense in Cotton Plant by the Mealybug Phenacoccus solenopsis

Abstract: The solenopsis mealybug, Phenacoccus solenopsis, has been recently recognized as an aggressively invasive pest in China, and is now becoming a serious threat to the cotton industry in the country. Thus, it is necessary to investigate the molecular mechanisms employed by cotton for defending against P. solenopsis before the pest populations reach epidemic levels. Here, we examined the effects of exogenous jasmonic acid (JA), salicylic acid (SA), and herbivory treatments on feeding behavior and on development of female P. solenopsis. Further, we compared the volatile emissions of cotton plants upon JA, SA, and herbivory treatments, as well as the time-related changes in gossypol production and defense-related genes. Female adult P. solenopsis were repelled by leaves from JA-treated plant, but were not repelled by leaves from SA-treated plants. In contrast, females were attracted by leaves from plants pre-infested by P. solenopsis. The diverse feeding responses by P. solenopsis were due to the difference in volatile emission of plants from different treatments. Furthermore, we show that JA-treated plants slowed P. solenopsis development, but plants pre-infested by P. solenopsis accelerated its development. We also show that P. solenopsis feeding inhibited the JA-regulated gossypol production, and prevented the induction of JA-related genes. We conclude that P. solenopsis is able to prevent the activation of JA-dependent defenses associated with basal resistance to mealybugs.
