Heat-shock protein 90 inCandida albicans

Abstract: Researches on Candidal heat-shock protein 90 (HSP90) in recent years are summarized.Candida albicans is a commensal pathogen in human and animals. In immunocompromised individuals it behaves as an opportunist pathogen, giving rise to superficial or systemic infections. Systemic candidosis is a common cause of death among immunocompromised and debilitated patients, in which the mortality is as high as 70%. HSP90 is now recognized as an immunodominant antigen inC. albicans and plays a key role in systemic candidosis as a molecular chaperone. The 47-ku peptide is the breakdown product of HSP90. Patients who has recovered from systemic candidosis produce high titre of antibodies to 47-ku antigen, whereas the fatal cases have little antibody or falling titres. The three commonest epitopes of candidal HSP90 have been mapped, epitopes C, B and H. Epitopes C and H are immunogenic. The antibody probes of both epitopes may be developed into a new serological test agents for systemic candidosis due to rather high specificity and sensitivity. The recent results establish HSP90 as an ATP-dependent chaperone that is involved in the folding of cell regulatory proteins and in the refolding of stress-denatured polypeptides. Some researches on fungal HSP90 and the treatment of patients with candidosis are reviewed as well.
Investigation and Analysis on the Achievement Motivations of 278 Senior High School Students

Abstract: This paper, applying Achievement Motivation Scale, conducts investigation to 278 grade 2 senior high school students on their achievement motivation. The research results show that there is no significant difference between the achievement motivations of students from common senior high school and those from key senior high school; the achievement motivations of senior high school students do have significant difference in genders and male students have higher achievement motivations than female students; the achievement motivations of students studying science and students studying arts have difference closely to significant difference; motivation to pursue success has negative correlation with motivation to avoid failure. Schools, gender and science type do not have cross functions on achievement motivations.

Abstract: Objectives: To describe the development of NCMS in the past decade including three sections and to bring up relevant policy implications. Methods: Based on secondary data and literature review, the evolution and achievement of NCMS in the past decade and the expected future challenges were analyzed. Results: Impressive advances have been seen in establishing the largest medical insurance system, New Cooperative Medical Scheme (NCMS), covered more than 800 million farmers in rural China during the past decade. Remarkable achievements during the development of NCMS include universal health coverage among rural residents, rapid increasing premium, balanced pooling fund, improved service, cost and coverage of farmers, and a strengthened primary health care system. In the meantime, the NCMS also confronted certain challenges: Institutionalization and legalization lagged behind the development of NCMS; payment reform failed to control the rapid growth of medical expenditure and financial protection for enrollees was insufficient; solidarity and equity between NCMS and other medical insurance systems is still an issue that needs to be solved; sustainable financing mechanism was not established successfully and moreover, it was also not compatible with the aging population and epidemiological transition of rural China; double coverage for rural residents turned up as portability was not achieved.

Abstract: Objective: To construct to silence tissue factor (TF) expression in Human Umbilical Vein Endothelial Cells (HUVECs) for providing evidence of the gene therapy and prevention of coagulation dysfunction in placental abruption (PA) neonates. Methods: Cultured HUVECs in vitro were divided into control group and PA group, each including non-prevention, scramble siRNA and TF-siRNA subgroups. pENTRTM/U6-shRNA/TF vector expressing TF were constructed and transfected into HUVECs. The mRNA expression of TF was tested with RT-PCR, and TF protein expression was detected with immunofluorescence staining. Results: Monolayer HUVECs with short-rod and short spindle shaped were adherent to the bottom, forming paving stone arrangement. The TF mRNA expression was significantly different between control group and PA group (P < 0.01) and among different subgroups (P < 0.01). In non-prevention subgroups, significant difference was observed in TF mRNA expression between control group and PA group (P < 0.05). Abstract: This paper tries to analyze Chinese three types of bank’s efficiency changes and differences, after the financial crisis, through the integrated use of DEA and super efficiency DEA method. The results show that: the efficiency of Chinese commercial Banks in general is on the rise, the comprehensive technical efficiency growth stems mainly from scale efficiency improvement, pure technical efficiency is stagnant, and scale efficiency presents convergence among all kinds of banks, while the technical level is the core strength of banks’ current and future competition. State-owned banks with the technology competitive advantage and scale efficiency improved, began to catch up with other banks, reversed the operational inefficiency; City commercial banks due to the system advantage were more easily to be in the efficient production frontier surface.

Abstract: Equations of the reaction-diffusion type are very well known and have been extensively studied in many research areas. In this paper, the prolongation structures for the system of the reaction-diffusion type are investigated from theory of coverings. The realizations and the classifications of the one-dimensional coverings of the system are researched. And the corresponding conservation law of the one-dimensional Abelian coverings is concluded, which is closely connected with the symmetry of the system by Noether theorem.
Correlation between Innervation of Skeletal Muscles and Myoblast Stem Cells Correlación entre la Inervación de los Músculos Esqueléticos y las Células Precursoras Mioblásticas

Abstract: In order to explore the change rule of myoblast stem cells (satellite cells, SCs) in the denervated and re-innervated muscle and to investigate the cellular mechanism of the morphological and functional changes of the muscle, denervated muscle atrophy and nerve regeneration models were established in one-month-old rats. Postoperative indexes such as muscle wet weight, cell section areas, content of collagen fibers and DNA, electrophysiology, numbers of SCs in the triceps muscle of calf were dynamically tested. After denervation, the muscle wet weight and cell area reduced rapidly, and the collagen fiber content increased slowly. The number of SCs increased at first, and then declined suddenly two months later. From 4 to 5 weeks after re-neuralization, muscle action potentials could be evoked, but the best innervation effect was found in the groups, which received re-neuralization at 2 months and 3 months after denervation. Denervation causes a progressive progress of muscle atrophy. SCs proliferate within 3 months after denervation, and then atrophy becomes irreversible from 4 months. At 4 or 5 weeks after re-neuralization, muscle action potentials can be evoked. Re-neuralization at 2 months and 3 months after denervation can achieve a good effect on the functional recovery of the atrophic muscle. Con el fin de explorar la regla de cambio de las células precursoras mioblásticas (células satélite, CSs) en el músculo denervado y re-inervado e investigar el mecanismo celular de los cambios morfológicos y funcionales del músculo, se establecieron, en ratas de un mes de edad, modelos de atrofia del músculo denervado y regeneración del nervio. Fueron examinados de manera dinámica índices postoperatorios tales como, el peso húmedo del músculo, áreas celulares de la sección, contenido de fibras de colágeno y ADN, electrofisiología, número de CSs en el músculo tríceps de las crías. Luego de la denervación, el peso del músculo húmedo y el área celular se redujeron rápidamente, mientras que el contenido de fibras colágenas aumentó lentamente. El número de CSs aumentó al inicio, pero más tarde, a los dos meses, disminuyó repentinamente. Entre las 4 a 5 semanas después de la reneuralización, los potenciales de acción muscular pudieron ser evocados, pero el mejor efecto de inervación se encontró en los grupos que recibieron reneuralización a los 2 y 3 meses después de la denervación. La denervación causó un avance progresivo de la atrofia muscular. Las CSs proliferaron dentro de los primeros 3 meses post-denervación, y luego de los 4 meses la atrofia fue irreversible
Actin and nuclear myosin I are associated with RNAP II and function in gene transcription

Abstract: The presence of actin in the nucleus as well as its functions in various nuclear processes has been made clear in the past few years. Actin is known to be a part of chromatin-remodeling complexes BAF, which are required for maximal ATPase activity of the Brg1 component of the BAF complex. Moreover, the essential roles of actin in transcription mediated by RNA polymerases I, II and III have been demonstrated recently. On the other hand, a myosin I isoform, which contains a unique NH2-terminal extension for nucleus localization, has been specifically localized in nucleus. As is well known, myosin I is an actin-binding protein and plays an important role in various cellular activities. Though actin and nuclear myosin I (NM I) have been implicated to play distinct roles in gene expression, there has been no evidence for the actin-myosin interaction that might be involved in gene transcription mediated by RNA polymerase II (RNAP II). Here we show evidence that both actin and NM I are associated with RNAP II in nucleus by using co-localization and co-IP assays, and they may act together on gene transcription. The antibodies against β-actin or NM I can block RNA synthesis in a eukaryotic in vitro transcription system with template DNA comprising the promoter and the coding region of human autocrine motility factor receptor (hAMFR) gene; the antibodies pre-adsorbed with purified actin and NM I have no effect in transcriptional inhibition, indicating that the inhibition of transcription by anti-actin and anti-NM I is specific. These results suggest a direct involvement of actin-myosin complexes in regulating transcription. It also implicates that actin and NM I may co-exist in a same complex with RNAP II and the interaction of RNAP II with actin and NM I functions in the RNAP II-mediated transcription.
Non-Homologous End Joining Plays a Key Role in Transgene Concatemer Formation in Transgenic Zebrafish Embryos

Abstract: This study focused on concatemer formation and integration pattern of transgenes in zebrafish embryos. A reporter plasmid based on enhanced green fluorescent protein (eGFP) driven by Cytomegalovirus (CMV) promoter, pCMV-pax6in-eGFP, was constructed to reflect transgene behavior in the host environment. After removal of the insertion fragment by double digestion with various combinations of restriction enzymes, linearized pCMV-pax6in-eGFP vectors were generated with different combinations of 5′-protruding, 3′-protruding, and blunt ends that were microinjected into zebrafish embryos. Repair of double-strand breaks (DSBs) was monitored by GFP expression following religation of the reporter gene. One-hundred-and-ninety-seven DNA fragments were amplified from GFP-positive embryos and sequenced to analyze the repair characteristics of different DSB end combinations. DSBs involving blunt and asymmetric protruding ends were repaired efficiently by direct ligation of blunt ends, ligation after blunting and fill-in, or removed by cutting. Repair of DSBs with symmetric 3′-3′ protrusions was less efficient and utilized template-directed repair. The results suggest that non-homologous end joining (NHEJ) was the principal mechanism of exogenous gene concatemer formation and integration of transgenes into the genome of transgenic zebrafish.
Local Gateway Assisted Handover Key Derivation in Enterprise Femtocell Network  [PDF]

With the dense deployment of femtocells in enterprise femtocell network and the small coverage of femtocells, handover in enterprise femtocell network will be frequent. The general handover key derivation method which is used in handover procedures in LTE is not suitable for handover in this scenario because of its long time cost and the weak security. To solve this problem, this paper has proposed a new local gateway assisted handover key derivation schema in enterprise femtocell network. It can meet the fast derivation and good forward/backward key secrecy requirement of handover key derivation in enterprise femtocell network. The simulation result has verified that the proposed handover key derivation schema works better than the existing method.
