Reciprocal Roles of Angiotensin II and Angiotensin II Receptors Blockade (ARB) in Regulating Cbfa1/RANKL via cAMP Signaling Pathway: Possible Mechanism for Hypertension-Related Osteoporosis and Antagonistic Effect of ARB on Hypertension-Related Osteoporosis

Abstract: Hypertension is a risk factor for osteoporosis. Animal and epidemiological studies demonstrate that high blood pressure is associated with increased calcium loss, elevated parathyroid hormone, and increased calcium movement from bone. However, the mechanism responsible for hypertension-related osteoporosis remains elusive. Recent epidemiological studies indicate the benefits of Angiotensin II Receptors Blockade (ARB) on decreasing fracture risks. Since receptors for angiotensin II, the targets of ARB, are expressed in both osteoblasts and osteoclasts, we postulated that angiotensin II plays an important role in hypertension-related osteoporosis. Cbfa1 and RANKL, the important factors for maintaining bone homeostasis and key mediators in controlling osteoblast and osteoclast differentiation, are both regulated by cAMP-dependent signaling. Angiotensin II along with factors such as LDL, HDL, NO and homocysteine that are commonly altered both in hypertension and osteoporosis, can down-regulate the expression of Cbfa1 but up-regulate RANKL expression via the cAMP signaling pathway. We thus hypothesized that, by altering the ratio of Cbfa1/RANKL expression via the cAMP-dependent pathway, angiotensin II differently regulates osteoblast and osteoclast differentiation leading to enhanced bone resorption and reduced bone formation. Since ARB can antagonize the adverse effect of angiotensin II on bone by lowering cAMP levels and modifying other downstream targets, including LDL, HDL, NO and Cbfa1/RANKL, we propose the hypothesis that the antagonistic effects of ARB may also be exerted via cAMP signaling?pathway.

Abstract: 肌腱膜纤维肉瘤癌基因同源物A(MafA)是成熟胰岛β细胞的重要转录因子,对胰岛素基因的转录、胰岛素分泌和β细胞团的增殖、分化至关重要。MafA与PDX-1和NeuroD1/BETA2协同作用,在胰岛β细胞和非胰岛β细胞中可诱导胰岛素基因的表达,有望成为糖尿病潜在的治疗靶点。在糖尿病小鼠模型和糖尿病患者的研究中发现,MafA基因缺陷可导致糖耐量异常和糖尿病的发生。人类MafA基因突变/变异可能与特殊类型糖尿病如新生儿糖尿病(NDM)或青少年成人起病型糖尿病(MODY)的致病及1型或2型糖尿病易感性的增加相关。
: V-maf musculoaponeurotic fibrosarcoma oncogene homolog A (MafA) is an important transcription factor of mature pancreatic islet β-cells, which is essential for insulin gene transcription, insulin secretion, and proliferation and differentiation of β-cell mass. MafA, together with PDX-1 and NeuroD1/BETA2, can synergistically induce insulin gene expression in islet β-cells and non-islet β-cells, which is expected to be a potential therapeutic target for the treatment of diabetes. Studies have shown that genetic deficiency of MafA gene in diabetic mouse models and diabetic patients can cause impaired glucose tolerance and the occurrence of diabetes. Mutations/variations of human MafA gene may be associated with the pathogenesis of special types of diabetes such as neonatal diabetes mellitus (NDM) and maturity-onset diabetes of the young (MODY), and the increased susceptibility to type 1 or type 2 diabetes mellitus
Neuroprotective, Anti-Amyloidogenic and Neurotrophic Effects of Apigenin in an Alzheimer’s Disease Mouse Model

Abstract: Alzheimer’s disease (AD) is a neurodegenerative disorder characterized by extracellular senile plaques and intracellular neurofibrillary tangles in the brain. Amyloid-β peptides (Aβ) are considered to play a critical role in the onset and progression of AD. Apigenin (4',5,7-trihydroxyflavone) is a pharmacologically active agent. Even though some evidence suggests that it has potential neuroprotective effects, no preexisting study has reported any therapeutic effects of apigenin in AD models. In the present study, we examined the effects of apigenin on cognitive function in APP/PS1 double transgenic AD mice and explored its mechanism(s) of action. Three-month oral treatment with apigenin rescued learning deficits and relieved memory retention in APP/PS1 mice. Apigenin also showed effects affecting APP processing and preventing Aβ burden due to the down-regulation of BACE1 and β-CTF levels, the relief of Aβ deposition, and the decrease of insoluble Aβ levels. Moreover, apigenin exhibited superoxide anion scavenging effects and improved antioxidative enzyme activity of superoxide dismutase and glutathione peroxidase. In addition, apigenin restored neurotrophic ERK/CREB/BDNF pathway in the cerebral cortex. In conclusion, apigenin may ameliorate AD-associated learning and memory impairment through relieving Aβ burden, suppressing amyloidogenic process, inhibiting oxidative stress, and restoring ERK/CREB/BDNF pathway. Therefore, apigenin appears to represent an alternative medication for the prevention and/or therapy of AD.

Abstract: The important parameters of camera system of TV theodolite have gotten with itself azimuth and pitching angle measure function.Camera focus can be measured through changing a little angle with constant target.Image center was gotten through comparing distances of target points in image because of symmetry of lens distortion.Through moving azimuth and pitching angle a distortion net of whole image was built.Then distortion model was set up with different models.
Design of unscented Kalman filter with correlative noises

Abstract: An Unscented-Kalman Filter(UKF) for a class of nonlinear discrete-time systems with correlated noises is designed to deal with the problem of nonlinear filtering failure in conventional UKF when system noise is correlated with measurement noise. Recursive filtering equations of UKF with correlated noises are given based on the minimum meansquare-error estimation; and unscented transformation(UT) is applied to the calculation the posterior means and covariances of the nonlinear system states. The proposed UKF breaks through the limitations on the conventional UKF that the system noise and measurement noise must be uncorrelated Gauss white noises, thus extending the applications of the conventional UKF. A simulation example shows its feasibility and effectiveness.

Unscented Kalman Filter for Nonlinear Systems with Colored Measurement Noise

Abstract: Traditional unscented Kalman filter (UKF) calls for that noise should be Gaussian white one, and can not solve nonlinear filtering problem with colored noise. For this reason, a new UKF filtering algorithm with colored measurement noise is proposed. Firstly, optimal filtering framework for a class of nonlinear discrete-time systems with colored measurement noise is derived on the basis of augmented measurement information and minimum mean square error estimation. Secondly, filtering recursive formula of UKF with colored noise is proposed through applying unscented transformation (UT) to calculation the posterior mean and covariance of the nonlinear state in this optimal framework. The proposed UKF can effectively deal with the issue that traditional UKF is failure under the condition that measurement noise is colored. A numerical simulation example also shows its feasibility and effectiveness.
Scutellaria barbate extract induces apoptosis of hepatoma H22 cells via the mitochondrial pathway involving caspase-3

Abstract: AIM: To study the growth inhibitory and apoptotic effects of Scutellaria barbata D.Don (S. barbata) and to determine the underlying mechanism of its antitumor activity in mouse liver cancer cell line H22.METHODS: Proliferation of H22 cells was examined by MTT assay. Cellular morphology of PC-2 cells was observed under fluorescence microscope and transmission electron microscope (EM). Mitochondrial transmembrane potential was determined under laser scanning confocal microscope (LSCM) with rhodamine 123 staining. Flow cytometry was performed to analyze the cell cycle of H22 cells with propidium iodide staining. Protein level of cytochrome C and caspase-3 was measured by semi-quantitive RT-PCR and Western blot analysis. Activity of caspase-3 enzyme was measured by spectrofluorometry.RESULTS: MTT assay showed that extracts from S. barbata (ESB) could inhibit the proliferation of H22 cells in a time-dependent manner. Among the various phases of cell cycle, the percentage of cells in S phase was significantly decreased, while the percentage of cells in G1 phase was increased. Flow cytometry assay also showed that ESB had a positive effect on apoptosis. Typical apoptotic morphologies such as condensation and fragmentation of nuclei and blebbing membrane of apoptotic cells could be observed under transmission electron microscope and fluorescence microscope. To further investige the molecular mechanism behind ESB-induced apoptosis, ESB-treated cells rapidly lost their mitochondrial transmembrane potential, released mitochondrial cytochrome C into cytosol, and induced caspase-3 activity in a dose-dependent manner.CONCLUSION: ESB can effectively inhibit the proliferation and induce apoptosis of H22 cells involving loss of mitochondrial transmembrane potential, release of cytochrome C, and activation of caspase-3.
Study on the effects of Lycium barbarum polysaccharides (LBP) and LBP combined with tea polyphenols on relieving the physical fatigue in mice

Abstract: 目的 评价枸杞多糖(LBP)及LBP配伍茶多酚(TP)缓解小鼠体力疲劳作用的效果,并探讨其作用机制。方法 SPF级雄性昆明小鼠336只,共分为4批,每批随机分成7组:LBP低、中、高剂量(LBP-L、LBP-M、LBP-H)组,LBP配伍TP(以下简称配伍组)低、中、高剂量(C-L、C-M、C-H)组,设生理盐水(NS)为对照,每组12只。小鼠在每日饲喂基础饲料的基础上,以每10 g体质量0.2 mL溶液灌胃,15 d后进行负重游泳实验,并检测肝糖原、血清尿素氮和血乳酸等生化指标。结果 同NS组相比,LBP-M、LBP-H组小鼠负重游泳时间显著延长(P<0.05),LBP-L、LBP-M、LBP-H组肝糖原的含量显著增加(P<0.05),血清尿素氮和血乳酸水平显著降低(P<0.05);C-L、C-M、C-H组小鼠负重游泳时间显著延长(P<0.05),肝糖原的含量显著提高(P<0.05),血清尿素氮和血乳酸水平显著降低(P<0.05)。同LBP组相比,配伍组负重游泳时间延长,肝糖原含量增加,血尿素氮和血乳酸水平降低;其中C-L组肝糖原水平显著高于LBP-L组(P=0.027),C-M组血乳酸水平显著低于LBP-M组(P=0.006)。结论 LBP及LBP配伍TP对小鼠体质量无影响,但具有缓解小鼠体力疲劳的作用;其作用机制可能与其增加了肝糖原储备量、减少蛋白质的分解代谢,进而减少血清尿素氮水平及增加了磷酸果糖激酶的活性,使血乳酸水平下降有关。
: Objective To evaluate the effects of Lycium barbarum polysaccharides (LBP) and LBP combined with tea polyphenols (TP) on relieving the physical fatigue in mice and discuss relevant mechanisms. Methods Three hundreds and thirty-six specific pathogen free (SPF) male KM mice were randomly assigned to four groups and each group was further divided into seven subgroups, i.e. three LBP groups receiving low, medium, and high doses of LBH (LBP-L, LBP-M, and LBP-H groups) and three combination groups receiving low, medium, and high doses of LBP combined with TP, and a normal saline (NS) group as the control group with 12 mice in each group. Mice were gavaged with LBP solution or LBP combined with TP solution at 0.2 mL/10 g body weight daily for 15 d. Then load swimming tests were conducted and biochemical indexes such as hepatic glycogen, serum urea nitrogen, and blood lactic acid were measured. Results LBP-M, LBP-H, C-L, C-M, and C-H groups had prolonged load swimming time (P<0.05) and LBP-L, LBP-M, LBP-H, C-L, C-M, and C-H groups had elevated hepatic glycogen level (P<0.05) and reduced serum urea nitrogen and blood lactic acid levels (P<0.05) as compared with the NS group. The combination groups had prolonged load swimming time, elevated hepatic glycogen level, and reduced serum urea nitrogen and blood lactic acid levels as compared with the LBP group. The hepatic glycogen level in C-L group was significantly higher than that in LBP-L group (P=0.027) and the blood lactic acid level in C-M group was significantly lower than that in LBP-M group (P=0.006). Conclusion LBP and LBP combined with TP can relieve the physical fatigue in mice without affecting the mouse body weight. The mechanism may be associated with increased hepatic glycogen storage, reduced catabolism for proteins, reduced serum urea nitrogen level, increased phosphofructokinase activity, and decreased blood lactic acid level
