Endometrial Carcinogenesis and Molecular Signaling Pathways  [PDF]

The Endometrial Cancer (EC) is the most common gynecologic malignancy that starts in the endometrium of women. Carcinogenesis of EC is associated with several critical regulatory molecules, which involve in different signaling pathways. A number of signaling pathways have been identified to be involved in the multiple-step development of EC, including PI3K/AKT/mTOR signaling pathway, WNT/β-catenin signal transduction cascades (including APC/β-catenin pathway), MAPK/ERK pathway, VEGF/VEGFR ligand receptor signaling pathway, ErbB signaling pathway, P53/P21 and P16INK4a/pRB signaling pathways. This review mainly focuses on the molecular signaling pathways relevant to human endometrial cancer and discusses those critical capabilities of transforming endometrial cells, including evading apoptosis; enhancing cell proliferation; blocking differentiation; and inducing angiogenesis.

SPOC domain of mint protein induces hematopoietic differentiation via Bmp4/Smad5 pathway  [PDF]

Abstract: Mint is a newly identified molecule that mediates signal transduction and modulates chromatin repression. Mint family members contain a highly conserved C-terminus SPOC domain (SpenParalog and OrthologsC-terminal domain) commonly associated with proliferation and related diseases (for example: cancer) due to its role in cell differentiation and apoptosis. In this study, we addressed the SPOC function using a tetracycline-inducible system to express the target domain in Ain V15 embryonic ES cells and bone marrow stem cells from SPOC transenic mice. In vitro differentiation of Ain V15 ES cells as a model of early hematopoietic development, we found expression of SPOC domain induces hematopoietic differentiation via up-regulation of transcription factors Bmp4 and Smad5, which induce the expression of hematopoietic factors Eklf1 and hematopoietic proliferation associated factor Gata2, the SPOC domain also plays the regulation function in the differentiation of hematopoitic progenitor by colony forming Unit (CFU) assays. Further, we determined SPOC expression enhances erythrocyte and granulocyte maturationusing bone marrow cells derived from tiSPOC chimeric mice. Finally, we identified that overexpression of full length Mint in ES cells drive Smad5 and Bmp4 up-regulation under culture conditions, and up-regulation of endogenous Mint when induceshematopoitic differentiation of EML, M1 and WT18 cells. In summary, our study reveals the conserved SPOC domain of Mint protein induces differentiation both in the stages of embryonic stem cells and hematopoietic progenitor cells.
Isoflavone Ameliorates H2O2 Induced Injury by Activating the Antioxidant System of Sow Mammary Gland Cell  [PDF]

Abstract: The antioxidant and protective properties of a synthetic soybean isoflavone (ISO) were investigated using sow mammary gland cell. Cells were cultured with 10, 20 and 30 uM ISO, respectively, under 80 uM FeSO4·7H2O/H2O2 conditions. After 48 h of incubation, the cells in the presence of ISO were lost less compared with that of control under oxidative damage by H2O2/FeSO4; ISO decreased the cell number at G1 and G2 stages, increased the cell number at S stage (all P < 0.05), it also reduced apoptosis of the cells (P < 0.01, P < 0.01, P < 0.05). The addition of ISO significantly promoted cell proliferation (P < 0.05) from 3rd to 6th days. Upon these, the activities of total superoxide dismutase (SOD), glutathione peroxidase (GPX), total antioxidant ability (T-AOC) also were increased and the activities of NADPH oxidase (NOX) decreased by ISO treatment (all P < 0.05). ISO decreased the concentration of ROS (P < 0.05) and MDA (P < 0.05). 10 uM, 20 uM, 30 uM ISO increased the relative mRNA abundance of SOD1 (all P < 0.05) and SOD2 (P < 0.05, P < 0.01, P < 0.05) and SOD3 (all P < 0.05); ISO significantly increased the relative mRNA abundance of GPX4 (all P < 0.01) and NOX4 (all P < 0.05), 20 uM ISO also increased the relative mRNA abundance of NOX2 (P < 0.05). It was concluded that supplementation of ISO enhanced the anti-oxidative function and prevented lipid peroxidation, possibly through the activation of the antioxidant enzymes and inhibition of cell apoptosis.
Cloning and expression of swine myostatin gene and its application in animal immunization trial

Abstract: We have amplified swine myostatin (MSTN) gene by reverse transcription polymerase chain reaction (RT-PCR) and cloned it into pGEM-T Easy vector. The cloned swine MSTN gene consists of 1128 nucleotides, which has been submitted to GenBank (acquired registered code-AY448008). The cloned swine MSTN gene was successfully expressed in E. coli without the first 25 amino acids. Crude extraction of the expressed recombinant MSTN protein was used to immunize mice to investigate the effects on their bodyweights. We show here that the body weights of the immunized mice were higher than that of the controls, even though the difference was not significant. Surprisingly, the progenies of the immunized mice also were heavier than the controls. Especially at day 3, the average body weight of the immunized mice was 10.5% higher than that of the controls, which is significant (p < 0.05).
Association of IGF-I gene polymorphisms with milk yield and body size in Chinese dairy goats

Abstract: the association of igf-i gene polymorphisms with certain traits in 708 individuals of two chinese dairy-goat breeds (guanzhong and xinong saanen) was investigated. polymerase chain reaction-single strand conformation polymorphism (pcr-sscp) and dna sequencing methods were employed in screening for genetic variation. two novel mutations were detected in the 5'-flanking region and in intron 4 of igf-i gene, viz., g.1617 g > a and g.5752 g > c (accession d26119.2), respectively. the associations of the g.1617 g > a mutation with milk yield and the body size were not significant (p > 0.05). however, in the case of g.5752 g > c, xinong saanen dairy goats with the cg genotype presented longer bodies (p < 0.05). chest circumference (p < 0.05) was larger in guanzhong goats with the gg genotype. in xinong saanen dairy goats with the cc genotype, milk yields were significantly higher during the first and second lactations (p < 0.05). hence, the g.5752 g > c mutation could facilitate association analysis and serve as a genetic marker for chinese dairy-goat breeding and genetics.
Emergence pattern of weed seedling community in direct-seeding cotton fields in the southwest of Anhui Province

Abstract: 为明确皖西南直播棉田杂草群落出苗模式及其与气象因子的相关性,采用定点定时调查法,于2010—2012年对棉田杂草种类、数量及群落结构进行调查,分析物种多样性,并以灰色关联度法分析直播棉田杂草群落出苗数量与气象因子的相关性.结果表明,皖西南棉区直播棉田杂草种类有13科24种.棉田杂草出苗有2次高峰,分别在5—6月和8—9月.棉田杂草群落多样性指数年度间变化较一致,但物种丰富度变化较大.周积温、周最高积温和周最低积温是影响皖西南直播棉田杂草群落出苗数量季节变化的主要因素,另外还受降水量影响.5—6月的杂草出苗高峰期是棉田杂草重点防治时期;相对占优势的杂草有牛筋草、千金子、旱稗、通泉草和马齿苋,是棉田杂草重点防治对象.
To clarify the emergence pattern and its influential factors of weed communities in direct-seedling cotton fields, weed density, species number and community structure were investigated by fixed time and location survey in southwestern Anhui Province. The species diversity and the relationship between meteorological elements and the seedling pattern were analyzed by using grey correlation analysis. The results showed that 24 weed species, which belong to 13 families, existed in direct-seedling cotton fields. Weed emergence showed two peaks, the first between May and June, and the second between August and September. The diversity characteristic values in weed community had the same trend among different years, while species richness was different. The seasonal fluctuation of weed seedlings was affected dominantly by weekly accumulative temperature, maximum weekly accumulative temperature and minimum weekly accumulative temperature; rainfall was the secondly important factor. The weed emergence peak between May and June was key weed control period in cotton fields. The key control objects were the relatively dominant species, such as Eleusine indica, Leptochloa chinensis, Echinochloa hispidula, Mazus japonicus and Portulaca oleracea.
Efficient Multi-resolution Detection of Binary Object

Abstract: This study presents an efficient multi-resolution method to detect binary object directly. Both intensity and geometry differences are used to measure the similarity between the source object template and target objects inside the test image. Especially for the geometrical difference, the histogram of oriented gradients is proposed to measure the similarity of the edge images between the source object template and the target object. Based on these two measurements, a multi-resolution strategy is used to accurately locate the object position in a coarse-to-fine way, where intensity difference and geometry difference applied consequently in each layer. Experimental results demonstrate the efficiency of the proposed method.
Neurotensin Receptor 1 (NTSR1) Overexpression in Breast Carcinomas Is Common and Independent of ER/PR/Her2 Expression  [PDF]

Neurotensin (NT) is a 13-amino acid peptide with trophic effects on some neoplasms. Its bioactivities are mainly mediated by neurotensin receptor 1 (NTSR1). Both NT and NTSR1 were found to be upregulated in breast cancer. NT/NTSR1 thus becomes a potential therapeutic target. We studied whether any correlation exists between the expression of NTSR1 in breast carcinomas and the expression of ER, PR, and Her2. A total 85 cases of invasive ductal (62) and lobular (23) breast carcinomas were studied. Based on their ER/PR profiles, the ductal carcinomas (DCs) were subcategorized into ER+/PR+ (21), ER+/PR (20), and ER/PR (21). All of the lobular carcinomas (LCs) were ER+/PR+. 21.57% of all DCs and 5.56% of LCs were Her2 positive. 77.78% of ER/PR DCs were also Her2 negative (triple negative). The expression of NTSR1 was detected by immunohistochemistry and was semiquantitated (as negative, 1+, 2+, 3+). Both 2+ and 3+ were collectively defined as overexpression. The expression of NTSR1 was weak and focal in non-neoplastic mammary epithelial cells. It is increased in 74.19% of DCs (80.95% in ER+/PR+, 75% in ER+/PR, and 66.67% in ER/PR group), and in 95.65% of LCs. The overexpression of NTSR1 is similar between ER+ DCs and ER DCs (75% vs 66.67%, p > 0.05) as well as between PR+ DCs and PR DCs (80.95% in ER+/PR+ DCs vs 75% in ER+/PR DCs, p > 0.05). And it was seen in 77.78% of Her2+ DCs, 78.38% of Her2 DCs, 94.12% of Her2 LCs, and 78.57% of triple negative DCs. Overall, NTSR1 is commonly overexpressed in both ductal and lobular breast carcinomas and is independent of the ER/PR/Her2 profiles of the tumors. Mechanochemical synthesis and high-capacity performances of transition-metal borides as aqueous anode materials

Abstract: Transition-metal borides MB0.5 (M = Co, Mo, V) were synthesized by high-speed mechanical ball-milling of the corresponding elemental metals and boron, and investigated as aqueous anode materials. The as-synthesized borides can achieve an excellent discharge capacity, about twice that of their parent transition metals. The metal boride electrodes also exhibit polarizations about 100–300 mV lower than those of their parent metals. The galvanostatic discharge curve of CoB0.5 shows a single discharge voltage plateau as a result of simultaneous electro-oxidation of elemental cobalt and/or amorphous cobalt boride. Both MoB0.5 and VB0.5 show two well-defined voltage plateaus, corresponding to the electro-oxidation of the corresponding metal and boride. These results show that the coexisting transition metal and boride in the metal borides co-activate each other in the ball-milling process, thereby significantly enhancing their electrochemical performances.
