Effect of erythropoietin on dynamic changes of TLR4,NF-κB,and IL-6 in rat model with necrotizing enterocolitis

Abstract: Objective To explore the protective effect and the mechanism of erythropoietin(EPO) on dynamic expressions of TLR-4,NF-κB,and IL-6 in the intestinal tissue of neonatal rat model with necrotizing enterocolitis(NEC).Methods A total of 90 SD rats(3 days old) were divided randomly into 3 groups(30 each) as follows: normal control group,NEC group,and EPO group.The NEC rat model was established by feeding the subjects with rat-milk substitutes after separation from the mother rats.The subjects were then exposed to hypoxia and cold stress.The EPO-treated rats were fed with the substitute of rat-milk containing EPO(0.1U/ml),and were exposed to hypoxia and cold stress as well.The rats in the control group were raised by their mothers and received no intervention.Ten rats from each group were sacrificed at 24,48,and 72h,respectively.The last 3cm of the terminal ileum was excised and the histological changes of the intestinal tissues were scored after H-E staining.Scores ≥2 were considered NEC.Fluorescence quantitative PCR was used to evaluate the dynamic levels of TLR4 mRNA and NF-κB mRNA.ELISA was used to determine the expression of IL-6.Results The TLR4 mRNA expression in the NEC group increased at 24h,peaked at 48h,and slightly decreased at 72h.The expressions in all time points increased significantly compared with those in the control group(P < 0.01).The rats in the EPO group expressed high levels of TLR4 mRNA at 24h.However,the expression rapidly declined later,and was significantly lower at 48 and 72h compared with those in the NEC group(P < 0.01).The expressions of NF-κB mRNA and IL-6 were significantly higher in the NEC group than in the control group at 48 and 72 h(P < 0.01).The expressions considerably increased in the EPO group at 48h,then declined at 72h,and were significantly lower than those in the NEC group(P < 0.01).No apparent pathological change was found in all groups at the time points of 24 and 48h.However,tissue lesion was observed in the NEC and EPO groups at 72 h,and the histological score was significantly higher in the NEC group(2.83±0.56) than in the EPO group(1.53±0.35,P < 0.01).Conclusions In the intestinal tract of the neonatal rat with NEC,innate immune response initiates earlier than the acquired immune response.Inflammatory response appears ahead of the tissue lesion,implying that the inflammatory response induces lesion.The enteral administration of human milk with a physiologic concentration of EPO may downregulate the TLR4 mRNA expression and inhibit the release of inflammatory mediators,thereby reducing intestinal tissue inju
Predictive value of neutrophil infiltration as a marker of Helicobacter pylori infection

Abstract: AIM: To evaluate the predictive value of neutrophil infiltration as a marker of Helicobacter pylori (H. pylori) infection. METHODS: A total of 315 patients with dyspepsia symptoms who underwent upper gastrointestinal endoscopy were enrolled in this study. Biopsies were evaluated using the updated Sydney system. The medication history of all patients in the preceding 4 wk was recorded. The diagnosis of H. pylori infection was based on 13C-urea breath test at least 4 wk after withdrawal of antisecretory drugs, antibiotics and related drugs. For the patients with subtotal gastrectomy, the diagnosis of H. pylori infection was based on anti-H. pylori immunoglobulin G (IgG) antibody. Serum anti-H. pylori IgG antibody was measured by enzyme-linked immunosorbent assays (Biohit, Finland). RESULTS: The sensitivity, specificity, positive predictive value and negative predictive value of neutrophil infiltration in the diagnosis of H. pylori infection were 92.3%, 83.5%, 77.4% and 94.7%, respectively. Neutrophil infiltration of gastric mucosa in the histological analysis was strongly associated with H. pylori infection (77.4% vs 5.3% in the neutrophil infiltration negative group, P = 0.000). Moderate neutrophil infiltration was more frequent in H. pylori infection when compared to mild infiltration (81.8% and 75%, respectively), but did not reach statistical significance. For those patients with negative rapid urease test, H. pylori was detected in 73.2% of patients with positive neutrophil infiltration on histology. In patients with subtotal gastrectomy, the diagnostic accuracy of neutrophil infiltration in H. pylori infection was 50%. CONCLUSION: Neutrophil infiltration is closely associated with H. pylori and may be recognized as a sign of this infection.
Optimal robust controller based on adaptive parallel genetic algorithms in uncertain domain

Abstract: In most of the existing robust controller designs based on the genetic algorithm, the search domains of variable are factitiously assigned. For undefined search domains, we propose a novel method for designing the optimal robust controller based on the adaptive parallel genetic algorithm. According to the distribution of the best individuals of each population in the current search, the algorithm narrows the uncertain search domain of variables by using the probability and statistics theory, so as to gradually achieve the optimum. Moreover, the impact of the individual fitness on the crossover probability and mutation probability is also investigated. By using this method, we design a simple, proper and low-order optimal robust controller, which effectively avoids the local optimum and improves the convergence and accuracy in simulation experiments.
An end-to-end anastomosis model of guinea pig bile duct: A 6-mo observation

Abstract: AIM: To establish the end-to-end anastomosis (EEA) model of guinea pig bile duct and evaluate the healing process of bile duct.METHODS: Thirty-two male guinea pigs were randomly divided into control group, 2-, 3-, and 6-mo groups after establishment of EEA model. Histological, immunohistochemical and serologic tests as well as measurement of bile contents were performed. The bile duct diameter and the diameter ratio (DR) were measured to assess the formation of relative stricture.RESULTS: Acute and chronic inflammatory reactions occurred throughout the healing process of bile duct. Serology test and bile content measurement showed no formation of persistent stricture in 6-mo group. The DR revealed a transient formation of relative stricture in 2-mo group in comparation to control group (2.94 ± 0.17 vs 1.89 ± 0.27, P = 0.004). However, this relative stricture was released in 6-mo group (2.14 ± 0.18, P = 0.440).CONCLUSION: A simple and reliable EEA model of guinea pig bile duct can be established with a good reproducibility and a satisfactory survival rate.

Dark matter cavity and amplification of gravitational waves

Abstract: The linearized Einstein theory in the approximately flat time-space is modified accounting for a massive graviton as a dark matter in the universe. On the basis of magnetic-type Maxwell- Proca -Vlasov equations, the interactions of the gravitoelectromagnetic fields (GEM), involving the vector potential, with gravitons (dark matter) dealing with resonant wave-particle and wave-wave one are studied. It is found that the behavior of the perturbed GEM fields and the perturbed matter density can be described by the nonlinear coupling Eqs.(4.6) and(4.7). It is shown that the perturbed field is instable with respect to the collapse, leading to enhancement of the GE field. In other words, the gravitational waves (GWs) are amplified and concurrently the cluster and cavity of the dark matter formed. And it is referred that the lower limit for graviton mass is $M_{gr}^\geqslant 10^{- 50}(g)$ by the condition to exist stable longitudinal mode.
Serum HER2 Level Measured by Dot Blot: A Valid and Inexpensive Assay for Monitoring Breast Cancer Progression

Abstract: Human epidermal growth factor receptor 2 (HER2) is one of the most important prognostic and predictive factors for breast cancer patients. Recently, serum HER2 ECD level of patients detected by enzyme-linked immunoabsorbent assay (ELISA) has been shown to predict tumor HER2 status and reveal its association with tumor progression, recurrence and poor prognosis. In this study, we established a new method, dot blot assay, to measure the serum HER2 level in breast cancer patients and further to evaluate the clinical value for monitoring breast cancer progression. We found that the serum HER2 level measured by dot blot assay was significantly correlated with tissue HER2 status in breast cancer patients (P = 0.001), and also significantly correlated with HER2 level measured by ELISA (P = 1.06×10?11). Compared with ELISA method, the specificity and sensitivity of dot blot assay were 95.3% and 65.0%, respectively. The serum HER2 levels of patients with grade III or ER-negative were higher than those with grade I–II (P = 0.004) and ER-positive (P = 0.033), respectively. Therefore, the novel dot blot method to detect serum HER2 level is a valid and inexpensive assay with potential application in monitoring breast cancer progression in clinical situations.

Abstract: A novel algorithm to detect objects across multiple views at the same instance is proposed based on object proba- bilistic random field(OOPF).OOPF is the core of our algorithm and it integrates all the information of individual views into the fusion space.Then object detection is implemented in the fu- sion space.An object window structure is designed in order to compute OOPF fast,which also provides a scheme to detect objects and the correspondence simultaneously.The experiment results demonstrate that the algorithm has a better performance against occlusion while the computational complexity is low.

Abstract: 以不对称还原4-氯乙酰乙酸乙酯(COBE)制备(S)-4-氯-3-羟基丁酸乙酯(S-CHBE)为模式反应,从实验室保藏菌种中筛选获得高效生物催化剂近平滑假丝酵母(Candida parapsilosis) CCTCC M 203011,底物浓度为20 g/L时产物产率达96%,光学纯度(e.e值)大于99%. 底(产)物分别具有对细胞(酶)的毒害作用和对酶促反应的抑制作用,是影响不对称还原反应催化效率的主要因素. 采用水-有机双相原位萃取和分批补料联用策略,原位萃取产物并控制水相底物浓度不高于4 g/L,可将底物浓度提高到60 g/L,产率达95%;同时采用高生物量策略,将产物总体积浓度提高到230 g/L,e.e值保持99%以上.
Dynamic analysis and assessment for sustainable development

Abstract: The assessment of sustainable development is crucial for constituting sustainable development strategies. Assessment methods that exist so far usually only use an indicator system for making sustainable judgement. These indicators rarely reflect dynamic characteristics. However, sustainable development is influenced by changes in the social\|economic system and in the eco\|environmental system at different times. Besides the spatial character, sustainable development has a temporal character that can not be neglected; therefore the research system should also be dynamic. This paper focuses on this dynamic trait, so that the assessment results obtained provide more information for judgements in decision\|making processes. Firstly the dynamic characteristics of sustainable development are analyzed, which point to a track of sustainable development that is an upward undulating curve. According to the dynamic character and the development rules of a social, economic and ecological system, a flexible assessment approach that is based on tendency analysis, restrictive conditions and a feedback system is then proposed for sustainable development.
