Antitumor effects and mechanisms of dendritic cells stimulated by sCD40L on ovarian cancer cells in vitro

Abstract: Zheng-mao Zhang,1 Xue-mei Yang,1 Chao Zhang,2 Min-jie Zhang,1 Xia Li,1 Feng-hua Zhang,3 Shan Kang,1 Shi-jie Wang,4 Bao-en Shan21Department of Gynecology and Obstetrics, Fourth Hospital of Hebei Medical University, 2Research Center, Fourth Hospital of Hebei Medical University, 3Department of General Surgery, Hebei General Hospital, 4Department of Gastroscopy, Fourth Hospital of Hebei Medical University, Shijiazhuang, People's Republic of ChinaObjective: This study aimed to examine the expression of immune suppression factors and the mechanisms of antitumor effects of cord blood dendritic cells (DCs) stimulated by soluble cluster of differentiation 40 ligand (sCD40L) and cytokines in vitro in ovarian cancer patients.Methods: The expression levels of interleukin (IL)-10 and transforming growth factor (TGF)-β messenger RNA in peripheral blood were detected by reverse transcription polymerase chain reaction; expression levels of CD80 and CD86 in DCs stimulated by sCD40L were detected using flow cytometry and confocal laser scanning microscopy.Results: Expression levels of IL-10 and TGF-β genes in the peripheral blood of ovarian cancer patients were significantly increased compared with patients with benign ovarian tumors (P < 0.05). The expression levels of CD80 and CD86 in DCs cultured in the granulocyte-macrophage colony-stimulating factor + IL-4 + stem cell factor + Flt-3 ligand + sCD40L group were significantly increased compared with those in the control group, as assessed by flow cytometry and confocal laser scanning microscopy (P < 0.05).Conclusion: A variety of cytokines in combination with sCD40L can promote the proliferation of cord blood-derived DCs and induce their maturation as well as stimulating a specific antitumor response.Keywords: antitumor response, cytokine, immunotherapy, cord blood, IL-10, TGF-β

Abstract: 泥盆纪火山岩在辛格尔断裂以北广泛分布,且以中酸性火山岩为主。笔者在库鲁克塔格地区辛格尔断裂南侧识别出泥盆纪英安岩,40Ar/39Ar坪年龄为374.7±5.9Ma,属晚泥盆世。该火山岩样品为钙碱性系列,岩石富铝,富轻稀土(∑LREE/∑HREE=9.43),K、Rb、Sr、Ba、Th等不相容大离子亲石元素相对富集。Eu具弱负异常,Nb、Ti亏损。火山岩形成于活动大陆边缘,可能经历了大陆壳物质的混染。库鲁克塔格火山岩从时代及岩性上与辛格尔断裂北侧泥盆纪火山岩对应,说明泥盆纪火山岩不仅分布于辛格尔断裂以北地区,也见于辛格尔断裂南部。

Abstract: In this work, the effect of solid soluble temperature, cold treating temperature and ag- ing temperature on the mechanical properties of 0Cr12Mn5Ni4Mo3Al had been inves- tigated systemically. The results show that under the optimal parameters (solid soluble treatment 1050℃×40min, cold treatment -78℃×4h, aging treatment 520℃×2h) the properties (strength and tenacity) could be up to its maximum.
Localized-Surface-Plasmon Enhanced the 357 nm Forward Emission from ZnMgO Films Capped by Pt Nanoparticles

Abstract: The Pt nanoparticles (NPs), which posses the wider tunable localized-surface-plasmon (LSP) energy varying from deep ultraviolet to visible region depending on their morphology, were prepared by annealing Pt thin films with different initial mass-thicknesses. A sixfold enhancement of the 357 nm forward emission of ZnMgO was observed after capping with Pt NPs, which is due to the resonance coupling between the LSP of Pt NPs and the band-gap emission of ZnMgO. The other factors affecting the ultraviolet emission of ZnMgO, such as emission from Pt itself and light multi-scattering at the interface, were also discussed. These results indicate that Pt NPs can be used to enhance the ultraviolet emission through the LSP coupling for various wide band-gap semiconductors.

Abstract: Temperature - dependent Embedded-Atom-Method (EAM) function of hcp titanium have been determined by fitting to the lattice constants and elastic constants measured at various temperatures as well as by considering the universal Rose's relation.The EAM functions thus obtained satisfy the stable structure requirement and Cauchy discrepancy. This work shows that EAM functions do have temperature-dependence,and in the meanwhile it also provides EAM functions of titanium for the study of Ti-based metallic materials at various temperatures.
Molecular characterisation of canine parvovirus strains circulating in China

Abstract: Canine parvovirus (CPV) was first isolated at 1978 in the USA. Analysis of CPV isolates by monoclonal antibodies and restriction enzymes have shown that after the first emergence of CPV (CPV-2) it evolved to give rise to new antigenic types, which were designated CPV types 2a, 2b and 2c. These new types have replaced the original CPV type 2, although the proportions of each of the new antigenic types vary in different countries. In China, CPV infections were first observed in 1982, however, there has been no information concerning the antigenic types of CPV prevailing in China now. In this study, we designed a PCR assay to type canine parvovirus strains in fecal samples collected from symptomatic dogs from 2006 to 2009. Our data showed that the CPV prevalent strain is mostly 2b, the proportion of CPV-2a is very low, no CPV-2c and CPV-2 were observed.

Abstract: The microstructure evolution of different cold rolling reductions (from 0 to 81.6% ) was studied by SEM (scanning electron microscopy) and TEM (transmission electron microscope). The study showed that the orientation multiplicity of pearlitic lamellas resulted in inhomogeneous deformation of different pearlitic lamellas, and with the increase of reduction, the microstructure underwent a course of "homogeneity→ inhomogeneity → homogeneity ". The result of XRD (X-ray diffraction)analysis indicated that cementite did not decompose and dissolve into ferrite; the results of the mechanical property test suggested that the relationship between Rp0.2 (yield strength) and ε (true strain) was in good agreement with Hollomon relationship. With the equation Rp0.2 = 1465ε0.18, the yield strength of the steel in different reductions could be well predicted.

Abstract: A reactive co-deposition processing for obtaining high-quality single-phase Bi2Sr2Cax-1CuxOy(Bi2212) thin films has been investigated using molecular beam epitaxy (MBE) with 2.7 x10-3Pa ozone gas introduction for oxidation. The thin films with a constant composition of almost 2:2:1:2 were designed to be fabricated at the substrate temperature between 675 and 780℃, the substrate temperature dependence of the surface morphology and the emergence phases were investigated in detail. A noticeable result is that the distribution of Cu element in the thin films is sensitively changed with the substrate temperature. At 750℃ it is inclined to locate in the periphery of each grain through the diffusion process. At 780℃ the Cu-compositional fluctuation brought around the dendritic crystal growth in the thin films. Below 705℃, the Bi2212 single-phase cannot be achieved in the thin films.
