Abstract: 根据已报道的大麦黄矮病毒GAV株系(BYDV-GAV)相关基因序列,利用RT-PCR方法获得ORF4基因。在杆状病毒-昆虫细胞系统中,成功表达了ORF4和GFP(绿色荧光蛋白)的融合蛋白(GFP:ORF4),Western blot检测到目的蛋白的表达。利用激光共聚焦显微镜观察其在细胞中的积累和亚细胞分布,发现ORF4基因编码的17kD蛋白(P4)能进入细胞核,并在细胞核膜上聚集。通过对ORF4基因编码的P4蛋白的N端和C端缺失突变结合蛋白质的结构预测分析,鉴定出N端α螺旋结构对于P4蛋白的核膜定位是必需的。这些结果为进一步研究ORF4基因在黄矮病毒GAV系统侵染中的生物学功能奠定了基础。

Abstract: To investigate the relationship between X chromosome and obligatory gene of a pedigree with congenital nystagmus,we used the following markers: DXS1214, DXS1068, DXS993, DXS8035, DXS1047, DXS8033, DXS1192 and DXS1232. Genome screening and genotyping were conducted in this pedigree of congenital nystagmus, and linkage analysis by LINKAGE package was used to determine the potential location. The linkage was not found on the Xp (All LOD score <-1) but on Xq (the maximum LOD score=2). The related gene of this pedigree was located on the long arm of X chromosome.We demonstrate that Xq26-Xq28 is a common locus for CMN. It bring us closer to the identification of a gene responsible for X-linked CMN.

Abstract: We report the clinical and pathological features of two pedigrees of palmoplantar keratoderma(PPK) ,the ex-pression of keratin 9(K9) in palm tissue and the mutation of the keratin 9 gene(KRT9) .Histopathology and immunohisto-chemical assessment was performed to analyze the epidermis in the palm of the probands.Genomic DNA of 46 family indi-viduals was used for amplification of exon 1 of KRT9.The mutations were determined by direct sequencing.Epidermal abnormalities in the palm of the two probands were characterized by vacuolar changes of suprabasal keratinocytes accom-panied by thickening of the living epidermis and stratum corneum.K9 was also expressed in particular epithelial tissues.Direct sequencing of polymerase chain reaction products revealed heterozygous missense mutations in exon 1 of KRT9(N160S and L167S) in the two families,respectively.N160S and L167S of KRT9 are disease-causing mutations in these two Chinese pedigrees with epidermolytic palmoplanter keratoderma(EPPK) .
Weak Power Frequency Magnetic Field Acting Similarly to EGF Stimulation, Induces Acute Activations of the EGFR Sensitive Actin Cytoskeleton Motility in Human Amniotic Cells

Abstract: In this article, we have examined the motility-related effects of weak power frequency magnetic fields (MFs) on the epidermal growth factor receptor (EGFR)-sensitive motility mechanism, including the F-actin cytoskeleton, growth of invasive protrusions and the levels of signal molecules in human amniotic epithelial (FL) cells. Without extracellular EGF stimulation, the field stimulated a large growth of new protrusions, especially filopodia and lamellipodia, an increased population of vinculin-associated focal adhesions. And, an obvious reduction of stress fiber content in cell centers was found, corresponding to larger cell surface areas and decreased efficiency of actin assembly of FL cells in vitro, which was associated with a decrease in overall F-actin content and special distributions. These effects were also associated with changes in protein content or distribution patterns of the EGFR downstream motility-related signaling molecules. All of these effects are similar to those following epidermal growth factor (EGF) stimulation of the cells and are time dependent. These results suggest that power frequency MF exposure acutely affects the migration/motility-related actin cytoskeleton reorganization that is regulated by the EGFR-cytoskeleton signaling pathway. Therefore, upon the MF exposure, cells are likely altered to be ready to transfer into a state of migration in response to the stimuli.

Abstract: 以人工繁育的俄罗斯鲟仔鱼为对象,采用实验生态学方法,研究了初次摄食时间对生长及存活的影响。结果表明:在水温17.4 ℃下,仔鱼在9~10日龄初次摄食,初次摄食前投喂对俄罗斯鲟仔鱼的生长和存活无显著性影响,延迟2 d以上初次摄食对生长影响明显,存活率在18日龄初次摄食时显著下降,24日龄时饥饿仔鱼全部死亡;23~24日龄时仔鱼如不能建立外源营养即达到饥饿不可逆点(PNR)。人工养殖条件下,俄罗斯鲟仔鱼必须在初次摄食后14 d内建立起主动摄食能力才能保证其仔鱼的正常发育、生长和存活。
