Upregulation of microRNA-451 increases cisplatin sensitivity of non-small cell lung cancer cell line (A549)

Abstract: Quantitative RT-PCR assay was performed to detect the expression of miR-451 in 10 pairs of NSCLC and noncancerous tissue samples. pcDNA-GW/EmGFP-miR-451 was stably transfected into NSCLC cell line (A549). Then, the effects of miR-451 upregulation on growth, colony formation and apoptosis of A549 cells were investigated. Finally, the effects of miR-451 upregulation on in vitro and in vivo sensitivity of A549 cells of DDP were also determined.The level of miR-451 expression in NSCLC tissues was significantly higher than that in corresponding noncancerous tissues. Ectopic overexpression of miR-451 could significantly inhibit growth and induce apoptosis of A549 cells. Moreover, ectopic overexpression of miR-451 could sensitize A549 cells to DDP possibly by increasing DDP-induced apoptosis which might be associated with the inactivation of Akt signaling pathway.This study demonstrated for the first time that combination of DDP application with miR-451 upregulation might be a potential strategy for the treatment of human NSCLC.NSCLC accounts for the majority of lung cancer cases and chemotherapy has been the mainstay of treatments of lung cancers [1]. Up to date, DDP still remains the most widely used first-line chemotherapeutic agent for NSCLC treatment. However, continuous infusion or multiple administration of DDP often cause severe side effects, including myelosuppression, asthenia, and gastrointestinal disorders, as well as long-term cardiac, renal, and neurological consequences [2]. Therefore, improving the sensitivity to drug doses strategies is still a challenge for chemotherapy efficacy. Novel therapeutic modalities combining genetic and chemotherapeutic approaches will play important roles in the fight against cancer in future.MicroRNAs (miRNAs) are small, endogenous non-coding RNAs that have been identified as post-transcriptional regulators of gene expression. Abstract: In the crystal structure of the title compound, C16H15NO3, intermolecular N—H...O hydrogen bonds link the molecules into chains parallel to the b axis and pairs of intermolecular O—H...O hydrogen bonds between inversion-related carboxylic acid groups link the molecules into dimers. The dihedral angle between the two benzene rings is 82.4 (2)°.

Abstract: The title compound, C8H8ClNO2, is almost planar, with an r.m.s. deviation of 0.0410 from the plane through the non-hydrogen atoms. In the crystal structure, intermolecular N—H...O hydrogen bonds link the molecules into chains along the b axis. An intramolecular N—H...O hydrogen bond results in the formation of a six-membered ring.

Abstract: The DNA damage response is critical for cells to maintain genome stability and survival. In this review, we discuss approaches to targeting critical elements of the DNA damage response for radiosensitization and chemosensitization. In addition, we also discuss strategies for targeting DNA damage response and DNA repair defects in cancer cells for synthetic lethality.
Effects of resveratrol on matrix metalloproteinase-9 expression in hepatoma cells

Abstract: Objective: To observe the effects of resveratrol on proliferation of human hepatocellular carcinoma cell line SMMC-7721 cells and expression of matrix metalloproteinase-9 (MMP-9) in vitro.Methods: SMMC-7721 cells were treated with different concentrations of resveratrol for 24, 48 and 72 h, respectively. The effect of resveratrol on proliferation of SMMC-7721 cells was assessed with methyl thiazolyl tetrazolium (MTT). The expression of MMP-9 mRNA was determined by reverse transcription polymerase chain reaction (RT-PCR). MMP-9 protein was identified by Western blot analysis.Results: Resveratrol could inhibit the proliferation of SMMC-7721 cells with dose- and time-dependent effects. Moreover, both MMP-9 mRNA expression and MMP-9 protein production were markedly reduced after resveratrol treatment. Conclusion: Resveratrol can inhibit the proliferation of SMMC-7721 cells and down-regulate MMP-9 expression. It is presumed that resveratrol may suppress the invasion and metastasis of hepatocellular carcinoma.
$η$ and $η^\prime$ Physics at BES-III

Abstract: Decays of both $\eta$ and $\eta^\prime$ provide very useful information in our understanding of low-energy QCD, and experimental signatures for these decays would be extremely helpful at BES-III. The rare decays of the $\eta$ and $\eta^\prime$ mesons could serve as a low-energy test of the Standard Model and its beyond. The sensitivities of the measurements of $\eta$ and $\eta^\prime$ decays are discussed at BES-III, in which the $\eta$ and $\eta^\prime$ mesons are produced in the $\psi$ decays.
Rare and forbidden decay of charm mesons at CLEO-c and BES-III

Abstract: This paper presents the results of searches for the flavor-changing neutral current, lepton-number-violating and lepton-flavor-violating processes in the charm mesons decays. Recent results from charged $D$ mesons decays to final states with dielectrons from CLEO-c are reported. The prospects of the searches for the rare charm decays at BES-III are also discussed.

Abstract: Hadron physics represents the study of strongly interacting matter in all its manifestations and the understanding of its properties and interactions. The interest on this field has been revitalized by the discovery of new light hadrons, charmonium- and bottomonium-like states. Recent results on charm physics at BESIII

Abstract: About 2.9 fb$^{-1}$ data set had been collected with the BESIII detector at the BEPCII collider. Taking advantages of data near open-charm threshold, we present preliminary results on leptonic and semileptonic charm-meson decays, as well as result on the FCNC process $D^0 \rightarrow \gamma \gamma$ from BESIII experiment. High precision charm data will enable us to validate Lattice QCD calculations at the few percent level. These can then be used to make precise measurements of CKM elements, $V_{cd}$ and $V_{cs}$, which are useful to improve the accuracy of test of the CKM unitary.
