Bone morphogenetic protein-15 in follicle fluid combined with age may differentiate between successful and unsuccessful poor ovarian responders

Abstract: Background The counselling of poor ovarian responders about the probability of pregnancy remains a puzzle for gynaecologists. The aim of this study was to optimise the management of poor responders by investigating the role of the oocyte-derived factor bone morphogenetic protein-15 (BMP-15) combined with chronological age in the prediction of the outcome of in-vitro fertilisation-embryo transfer (IVF-ET) in poor responders. Methods A retrospective study conducted in a university hospital. A total of 207 poor ovarian responders who reached the ovum pick-up stage undergoing IVF/intracytoplasmic sperm injection (ICSI) with three or fewer follicles no less than 14 mm on the day of oocyte retrieval were recruited from July 1, 2008 to December 31, 2009. Another 215 coinstantaneous cycles with normal responses were selected as controls. The BMP-15 levels in the follicular fluid (FF) of the 207 poor responders were analysed by western blot. Based on the FF BMP-15 level and age, poor responders were sub-divided into four groups. The main outcome measures were the FF BMP-15 level, implantation rate, pregnancy rate, and live birth rate. Results The implantation rate (24.2% vs. 15.3%), chemical pregnancy rate (40% vs. 23.7%), clinical pregnancy rate (36.5% vs. 20.4%) and live birth rate (29.4% vs. 15.1%) in the high BMP-15 group were significantly higher than those in the low BMP-15 group. Furthermore, poor responders aged less than or equal to 35 years with a higher FF BMP-15 level had the best implantation, pregnancy and live birth rates, which were comparable with those of normal responders. Conclusions Our study suggests a potential role of BMP-15 in the prediction of the IVF outcome. A high FF BMP-15 combined with an age less than or equal to 35 years may be used as a potential indicator for repeating IVF cycles in poor ovarian responders.

Abstract: Loading time is one of the most important dynamic characteristics of a magneto-optical trap. In this paper, we primarily report on a detailed experimental study of the effects of some magneto-optical trap control parameters on loading time, including the background vacuum pressure, the magnetic field gradient, and the intensities of trapping and repumping lasers. We compare the results with previous theoretical and experimental results, and give qualitative analysis. These experimental investigations offer some useful guidelines to control the loading time of magneto-optical traps. The controllable loading time achieved is helpful to enhance the signal-to-noise ratio of photoassociation spectroscopy, which is remarkably improved from 7 to 28.6.
Polymorphisms of alcohol dehydrogenase-2 and aldehyde dehydrogenase-2 and esophageal cancer risk in Southeast Chinese males

Abstract: AIM: To evaluate the impact of alcohol dehydrogenase-2 (ADH2) and aldehyde dehydrogenase-2 (ALDH2) polymorphisms on esophageal cancer susceptibility in Southeast Chinese males.METHODS: Two hundred and twenty-one esophageal cancer patients and 191 healthy controls from Taixing city in Jiangsu Province were enrolled in this study. ADH2 and ALDH2 genotypes were examined by polymerase chain reaction and denaturing high-performance liquid chromatography. Unconditional logistic regression was used to calculate the odds ratios (OR) and 95% confidence interval (CI).RESULTS: The ADH G allele carriers were more susceptible to esophageal cancer, but no association was found between ADH2 genotypes and risk of esophageal cancer when disregarding alcohol drinking status. Regardless of ADH2 genotype, ALDH2G/A or A/A carriers had significantly increased risk of developing esophageal cancer, with homozygous individuals showing higher esophageal cancer risk than those who were heterozygous. A significant interaction between ALDH2 and drinking was detected regarding esophageal cancer risk; the OR was 3.05 (95% CI: 1.49-6.25). Compared with non-drinkers carrying both ALDH2 G/G and ADH2 A/A, drinkers carrying both ALDH2 A allele and ADH2 G allele showed a significantly higher risk of developing esophageal cancer (OR = 8.36, 95% CI: 2.98-23.46).CONCLUSION: Both ADH2 G allele and ALDH2 A allele significantly increase the risk of esophageal cancer development in Southeast Chinese males. ALDH2 A allele significantly increases the risk of esophageal cancer development especially in alcohol drinkers. Alcohol drinkers carrying both ADH2 G allele and ALDH2 A allele have a higher risk of developing esophageal cancer.

Abstract: AIM: To evaluate the relationship between drinking and polymorphisms of alcohol dehydrogenase 2 (ADH2) and/or aldehyde dehydrogenase 2 (ALDH2) for risk of colorectal cancer (CRC) in Chinese males.METHODS: A case-control study was conducted in 190 cases and 223 population-based controls. ADH2 Arg47His (G-A) and ALDH2 Glu487Lys (G-A) genotypes were identified by PCR and denaturing high-performance liquid chromatography (DHPLC). Information on smoking and drinking was collected and odds ratio (OR) was estimated.RESULTS: The ADH2 A/A and ALDH2 G/G genotypes showed moderately increased CRC risk. The age- and smoking-adjusted OR for ADH2 A/A relative to G/A and G/G was 1.60 (95% CI=1.08-2.36), and the adjusted OR for ALDH2 G/G relative to G/A and A/A was 1.79 (95% CI=1.19-2.69). Significant interactions between ADH2, ALDH2 and drinking were observed. As compared to the subjects with ADH2 G and ALDH2 A alleles, those with ADH2 A/A and ALDH2 G/G genotypes had a significantly increased OR (3.05, 95% CI= 1.67-5.57). The OR for CRC among drinkers with the ADH2 A/A genotype was increased to 3.44 (95% CI= 1.84-6.42) compared with non-drinkers with the ADH2 G allele. The OR for CRC among drinkers with the ALDH2 G/G genotype was also increased to 2.70 (95% CI= 1.57-4.66) compared with non-drinkers with the ALDH2 A allele.CONCLUSION: Polymorphisms of the ADH2 and ALDH2 genes are significantly associated with CRC risk. There are also significant gene-gene and gene-environment interactions between drinking and ADH2 and ALDH2 polymorphisms regarding CRC risk in Chinese males.

Insufficient maintenance DNA methylation is associated with abnormal embryonic development

Abstract: To evaluate the possible involvement of DNA methylation in human EPL, the expression of DNMT proteins and global methylation of DNA were assessed in villous or decidua from EPL patients. The association of maintenance methylation with embryo implantation and development was also examined.We found that DNMT1 and DNMT3A were both expressed in normal human villous and decidua. DNMT1 expression and DNA global methylation levels were significantly down-regulated in villous of EPL. DNMT3A expression was not significantly changed in the EPL group compared to controls in either villous or decidua. We also found that disturbance of maintenance methylation with a DNMT1 inhibitor may result in a decreased global DNA methylation level and impaired embryonic development in the mouse model, and inhibit in vitro embryo attachment to endometrial cells.Our results demonstrate that defects in DNA maintenance methylation in the embryo, not in the mother, are associated with abnormal embryonic implantation and development. The findings of the current study provide new insights into the etiology of EPL.Early pregnancy loss (EPL) is one of the most common reproductive failures of human pregnancy. Abstract: Ultracold atom photoassociation spectroscopy is significant for studying the long-range molecular potential energy structure. The signal-noise ratio of ultracold Cesium Molecule photoassociation Rovibronic Spectrum is one of the indicators as a measure of detection technology which directly affect the distinguishing ability and detection sensitivity of the spectra. We obtain an long-range ultracold cesium molecular hyperfine rovibration spectra by modulating the fluorescence spectroscopy. We find that demodulation parameters have a significant impact to the signal-noise ratio of spectra by studying the demodulation parameters such as integration time and sensitivity, which show nonlinear relation with the ratio. We achieve the optimal control of the signal-noise ratio for spectrum according to our experiment, which laid an important foundation for further studying the long-range state rovibration level of ultracold cesium molecule.
Current-induced spin wave excitation in Pt|YIG bilayer

Abstract: We develop a self-consistent theory for current-induced spin wave excitations in normal metal-magnetic insulator bilayer systems, thereby establishing the relation between spin wave excitation and the experimentally controlled parameters. We fully take into account the complex spin wave spectrum including dipolar interactions and surface anisotropy as well as the spin-pumping at the interface. Our results focus on the mode-dependent power close to the critical currents for spin wave excitation. The major findings are (a) the spin transfer torque can excite different spin-wave modes simultaneously; (b) spin pumping counterbalances spin-transfer torque and affects the surface modes more than the bulk modes; (c) spin pumping inhibits high frequency spin-wave modes, thereby redshifting the excitation spectrum. We can get agreement with experiments on yttrium iron garnet|platinum bilayers by postulating the existence of surface anisotropy modes.

Abstract: The paper propose test statistics based on Cauchy estimation for unit root with periodical heteroscedastic shocks in seasonal time series.Under null hypothesis,the limit distributions of statistics are standard normal regardless of the period of seasonality and periodical heteroscedasticity of shocks.By Monte Carlo simulation,it is obtained that the estimators have smaller errors by this method than LSE when the autoregressive coefficient is close to 1.Simulations and a real example show the convenience and validity of this test method.
