miR-346 Regulates Osteogenic Differentiation of Human Bone Marrow-Derived Mesenchymal Stem Cells by Targeting the Wnt/β-Catenin Pathway

Abstract: Osteogenic differentiation of human mesenchymal stem cells (hMSCs) is regulated by multiple transcription factors and signaling molecules. However, the molecular mechanisms underlying this process remain to be fully elucidated. MicroRNAs (miRNAs) act as key regulators in various biological processes by mediating mRNA degradation or translational inhibition of target genes. In this study, we report that miR-346 plays critical roles in regulating osteogenic differentiation of hBMSCs. The expression of endogenous miR-346 was increased during osteogenic differentiation of hBMSCs. Overexpression of miR-346 significantly promoted osteogenic differentiation, whereas miR-346 depletion suppressed this process. Further studies confirmed that miR-346 directly targeted the 3′-UTR of the glycogen synthase kinase-3β (GSK-3β) gene so as to suppress the expression of GSK-3β protein. Similar to miR-346 overexpression, GSK-3β depletion promoted osteogenic differentiation, whereas GSK-3β overexpression reversed the promotional effect of miR-346. We further found that miR-346 overexpression activated the Wnt/β-catenin pathway and increased the expression of several downstream genes including CyclinD1, c-Myc, TCF-1 and LEF-1. Depletion of β-catenin almost completely blocked the positive role of miR-346 on osteogenic differentiation. Taken together, our data indicate that miR-346 positively regulates hBMSC osteogenic differentiation by targeting GSK-3β and activating the Wnt/β-catenin pathway.

Abstract: Using the "spring" model for a weak interface between two solids, we derive the charac-teristic equation for the Lamb waves in a two-layered solid composite and make numerical computations for dispersion curves in an aluminum/copper structure with a rigid or slip interface. The influence of the shear stiffness of the interface on the low order Lamb wave modes is considered.
PROPERTIES OF GAS AND CHAR FROM MICROWAVE PYROLYSIS OF PINE SAWDUST

Abstract: Pine sawdust pyrolysis was carried out respectively using microwave and conventional electrical heating at different temperatures in order to understand the properties of pyrolytic products from microwave pyrolysis of biomass. Less char material was obtained by microwave pyrolysis compared to conventional heating at the same temperature. While comparing the components of the pyrolytic gases, it was revealed that the microwave pyrolysis gas usually had higher H2 and CO contents and lower CH4 and CO2 contents than those obtained by conventional pyrolysis at the same temperature. The texture analysis results of the microwave pyrolysis chars showed that the chars would melt and the pores would shrink at high temperatures, and hence, the specific surface areas of the chars decreased with increasing temperature. Similarly, the reactivity of the char was remarkably reduced when the microwave pyrolysis temperature exceeded 600°C.

Abstract: The influence of the structure and position of groove in a two-dimensional metallic subwavelength slit on transmission properties has been studied with the finite-difference time-domain method. The whole structure of slit with perpendicular groove could be regard as a LC oscillation circuit. The capacitance and inductance of this oscillation circuit will be altered when the structure of groove varies,so that the resonant transmission peaks shift. And the drive source of circuit changes when the position of groove moves along the slit, as a result the distribution of electromagnetic filed in the structure is changed also. So the resonant transmission peaks are split. In this paper, the change of resonant transmission peaks as each parameter of the groove changes is analyzed with the principle of LC resonance.

Abstract: We report the generation of multi-wavelength visible light through amplified spontaneous emission (ASE) in Er3+-doped and Er3+/Yb3+-doped germanosilicate single-mode optical fiber pumped by a Nd:YLF laser at 1313nm. In the Er3+-doped fiber, the intense multi-wavelength blue emission hnes around 463-510nm corre-spond to transitions born 2G7/2 etc. excited states to the metastable 4I13/2 state, and their pumping mechanists is aecomphshed by a stepwise four-photon absorption. Some emission hnes in this wavelength region are attributed to the three-wave sum-frequency process of 1313 and 1530nm (corresponds to 4I13/2 -4I15/2). The intense green emission hnes at 525 and 540 nm are also observed in the Er3+-doped fiber. In the Er3+/Yb3+-doped fiber the blue and green lines are very weak compared with those in the Er3+-doped fiber.

Synthesis of Novel Visible Light Driven BiVO4 Photocatalysts via Microemulsion Process and its Photocatalytic Performance

Abstract: The novel visiblea2lighta2driven BiVO4 photocatalyst with tetragonal and monoclinic structures was synthesized via a reverse microemulsion method using Bi(NO3)3 and NH4VO3 as starting materials. The asa2prepared BiVO4 samples were characterized by X-ray diffraction (XRD), scanning electron microscope (SEM) and Xa2ray photoelectron spectroscope (XPS). The photocatalytic activity of the BiVO4 powders was determined by degradation of methyl orange molecules in water under visible light irradiation (| gt;400nm). The results revealed that the particle and crystallite sizes of the BiVO4 powders significantly depended on synthesis temperature of the reverse microemulsion solutions. The photocatalytic tests indicated that the synthesis temperature of the microemulsion solutions can significantly affect the visible light driven photocatalytic activities. The highest methyl orange degradation of 99.9% within 3h was achieved on the BiVO4 samples with mixed tetragonal and monoclinic phases.

Abstract: High frequency somatic embryogenesis was obtained from embryonic leaflets of peanut ( Arachis hypogaea L. ) which were induced on MS medium containing 20 mg/L 2,4-D for 20 d and later subcultured on MS medium without phytohormones. Histological observations reavealed that somatic embryos originated from the cells of epidermis and several layers of subepideirois which underwent dediffemntiation and developed into embryonic cell mass with dense cytoplasm and large nudens. The embryonic cell mass continued to devide and developed further into embryos through globular, heart-shaped, torpedo-shaped and cotyledon stages.

Abstract: The calcium ion (Ca2+) is the simplest and the most versatile intracellular second messenger known to date. Over last decade, the discovery of Ca2+ sparks and a family of local elementary Ca2+ signaling events has unveiled a “digital-analog” feature of intracellular Ca2+ signaling. Brief, local “digital” Ca2+ microdomains and nanodomains are commingled with the continuous global “analog” Ca2+ signals. Patterned activation of digital Ca2+ signals confers on intracellular Ca2+ signaling a multi-scaled architecture in space, time and magnitude, and is of particular relevance in spatiotemporal regulation of high-threshold Ca2+-dependent processes (e.g., striated muscle excitation-contraction coupling, exocytosis). Investigation on spark activation, coordination, and termination has revealed new insights as well as perplexing questions about gating of Ca2+ release channels (ryanodine receptor, inositol 1,4,5-trisphosphate receptor) in intact cells, and brought about mechanistic understanding of the diverse and even opposing roles of Ca2+ signaling. Future investigation on “digital-analog” dichotomy of Ca2+ signaling thus holds the promise to unify the simplicity and complexity of Ca2+ signaling in biology.
Effects of pCEP4-PTH gene plasmid on eggshell quality and Serum hormone levels in layer

Abstract: 试验选用56周龄新扬州鸡产蛋期母鸡150只,随机分为3组,每组5个重复,每个重复10只;腿肌注射0、200、400 μg鸡甲状旁腺素基因质粒(pCEP4-PTH),研究其对蛋鸡生产性能、蛋壳质量和血液部分激素的影响.试验结果表明,pCEP4-PTH基因质粒可改善蛋鸡生产性能;200 μg注射量可显著改善蛋壳强度(P<0.05),400 μg注射量对蛋壳质量影响不显著;pCEP4-PTH基因质粒可显著提高蛋鸡体内甲状旁腺素(PTH)水平(P<0.05),且注射400 Pg时达到极显著水平(P<0.01);注射400μgpCEP4-PTH基因质粒具有促进内源降钙素(CT)和雌激素(E2)分泌的作用(P<0.05),因而不利于PTH生物学效应的发挥;两试验组骨钙素(BGP)与对照组差异不显著,提示生产性能和蛋壳质量的改善未对骨骼发育造成不良影响.说明pCEP4-PTH基因质粒可改善蛋鸡生产性能和蛋壳质量,且不影响骨骼质量.
