Purification and properties of a thermostable chitinase from thermophilic fungus Thermomyces lanuginosus

Abstract: A thermostable extracellular chitinase from culture supernatant of a thermophilic fungus Thermomyces lanuginosus was purified to SDS-PAGE homogeneity, by using ammonium sulfate fraction, DEAE-Sepharose Fast flow chromatography, Phenyl-Sepharose Fast Flow chromatography. A molecular mass of the purified enzyme was between 48-49.8 kD determined by SDS-PAGE and gel filtration chromatography. The chitinase exhibited optimum catalytic activity at pH 4.5 and 55 degrees C respectively. It was thermostable at 50 degrees C and retained 24% activity after 20 min at 70 degrees C. The half life time of the enzyme at 65 degrees C was 25 min. Different metal ions showed different effects on the chitinase activity. Ca2+, Ba2+, Na+, K+ enhanced the enzyme activity, whereas Fe2+, Ag+, Hg2+, Cu2+ caused obvious inhibition. The Km and Vmax values of chitinase on colloidal chitin were 9.56 mg/mL and 22.12 micromol/min respectively. The chitinase showed antifungal activity aginst tested fungi to different degree.

Abstract: This paper takes the color photography picture and large-scale terrain map from Northeast University as data source,and gains the DOM data and DEM by using VirtuoZoNT3.5.After pretreatment,these data can have the same coordinate system.By using 3D GIS software Skyline Terrasuite,we overlay DOM to DEM structuring the vein coming to show 3D circumstances and the building roof 3D surface.Also by making use of a digital camera to collect the side vein of buildings,we can achieve 3D landscape of city Visualization.The paper discus above-mentioned method process,and give some experience summary.

Abstract: The basic mathematic model and algorithm of independent component analysis and the apply of independent component analysis in noisy mixed images was discussed, a method was proposed of combining Wiener filtering de-noising and independent component analysis. The results show that this method can reduce the affect of noise, accordingly renew the original image.

A Classified Pre-Computed QoS Routing Algorithm

Abstract: In order to satisfy the quality of service (QoS) of the application on Internet, Internet engineering task force (IETF) has presented a set of network service mode such as IntServ/RSVP (Integrated Service/Resource Reservation Protocol), DiffServ (differentiated service) and MPLS (multi protocol label switching). All of these service modes need a correlative QoS routing mechanism and algorithm to assign an adapted route. Based on the research of existing routing algorithms, a new pre-computed QoS routing algorithm CCPF(classified minimum cost path first)is put forward,which uses bandwidth as a basic metric and gives a weighted cost function as a path optimal criterion.By simulation,this algorithm is compared with the others.The results show that it can get differet routes among different bandwidth-ranges to support multipl service modes.

Abstract: 研究了液体发酵嗜热毛壳菌Chaetomium thermophile产生的β-葡萄糖苷酶的分离纯化及特性。粗酶液经硫酸铵沉淀、DEAE-Sepharose Fast Flow阴离子层析、Phenyl-Sepharose 疏水层析、Sephacryl S-100分子筛层析等步骤后获得凝胶电泳均一的β-葡萄糖苷酶。经12.5%SDS-PAGE和凝胶过滤层析方法分别测得该酶的分子量大小约为78.4kDa和81kDa。该酶反应的最适温度和pH值分别为60℃和4.5-5.0。有较好的酸稳定性和热稳定性。金属离子对β-葡萄糖苷酶的活性影响较大, 其中Ca2+对酶有激活作用, 而Ag+、Cu2+ 、Hg2+对酶有显著的抑制作用。该酶对水杨苷具有很强的底物特异性。

Abstract: 疏绵状嗜热丝孢菌(Thermomyces lanuginosus)SY2 在以胶状几丁质为唯一碳源的诱导培养基中产生了胞外几丁质酶.该酶在50℃保温1 h,酶活稳定;65℃时半衰期为25 min;酶液在室温下保存到12周,残余酶活性为45%左右.该酶有较宽的pH范围,3.0~9.0之间保持稳定,pH值为2.5时,仍具有70%的剩余酶活性.Ca2 对几丁质酶的活性有显著的激活作用;高浓度变性剂对酶有抑制作用.结果表明该酶是一种热稳定性高且耐酸碱的新型几丁质酶,能在酸性和高温环境中发挥作用,这些特性赋予了T.lanuginosus几丁质酶在几丁质的生物转化及其它生物技术中极大的应用优势.

Effect of Trichoderma aureoviride 1010 on chilly stress-resistance of tomato in solar-greenhouse conditions

Abstract: In this study, we determined the enhancement effects of Trichoderma aureoviride 1010 (T1010) on chilly stress-resistance of tomato in solar-greenhouse conditions via soil application and 3 days of inferior chilly-stress (16 ± 2 °C in the day and 8 ± 2 °C at night ). Results show significant differences in the indicators for inferior chilly stress-resistance of functional tomato leaves and roots. SOD, CAT, POD and PPO activities of functional tomato leaves treated with T1010 increase by 266.33%, 16.85%, 20.89% and 632.89% respectively over the control. Carotenoid, ABA, proline, glucose and starch contents also increase by 50.53%, 119.40%, 55.38%, 128.57% and 23.40% respectively. For tomato roots treated with T1010, SOD, CAT, POD and PPO activities increase by 274.10%, 69.06%, 8.26% and 164.14% respectively. The contents of proline, glucose, and starch also increase by 49.81%, 66.00% and 86.76% respectively. In addition to decreasing soluble sugar content, the indicators for functional tomato leaves treated with 1/2T1010 increase at varying degrees. However, the indicators for tomato roots treated with 1/2T1010 decrease at varying degrees. ANOVA analysis shows significant difference in indicators of inferior chilly stress-resistance among CK, T1010 and 1/2T1010 treatments. T1010 positively influences chilly stress-resistance of functional tomato leaves and roots in solar-greenhouse conditions with reasonable soil organic quality.

Abstract: Reverse time migration (RTM) is an accurate migration method, which is based on two-way wave equation, eliminates angle limitation, and can be capable for imaging of reverse branch and multiple. In the development of an oilfield in Pearl River Mouth (PRM) Basin, RTM has been applied to solve the problems of fault shadow and structure in distinction and the drilling has proven that the RTM result is reliable. This paper introduces the theory of RTM and emphatically discusses parameter selection. The results of RTM showed that it had advantages in resolving fault shadow and the imaging of steep dip structure, and could be utilized in the oilfield.
